Abstract
Kerang Simping Placuna placenta merupakan komoditas bernilai ekonomis bagi masyarakat pesisir di area tangkapan kerang di Socah Bangkalan dan Ujungpangkah Gresik. Tekanan ekologis berupa zat pencemaran semakin meningkat akibat aktivitas manusia. Tekanan ekologis ini dapat menyebabkan tekanan oksidatif pada biota perairan yang direspon dalam bentuk pertahanan primer dengan enzyme antioksidan. Tujuan penelitian yaitu 1. Mendapatkan respon antioksidan dan Nilai Indeks Multimarker Polusi (IMP) pada kerang simping Placuna placenta dari area tangkapan nelayan Ujungpangkah dan Socah, 2. Mendapatkan keterkaitan respon biomarker antioksidan terhadap parameter habitat kerang simping, 3. Mendapatkan keterkaitan respon biomarker antioksidan terhadap nilai indeks kondisi pada kerang simping. Metode penelitian yang dilakukan ada 3 Tahapan yaitu: 1). Pengukuran respon biomarker pada organ insang, saluran pencernaan dan otot pada kerang simping meliputi antioksidan SOD, CAT, GPx dan MDA, serta dilakukan penghitungan nilai Indeks Multimarker Polusi berdasarkan hasil respon antioksidan dengan analisis diskriminan dan Analisa PCA. 2). Analisa respon biomarker antioksidan terhadap parameter kualitas air dan sedimen dan kerang dengan pengukuran kandungan logam berat dan PAH untuk menghitung nilai biokonsentrasi logam berat dan PAH, dan komposisi mineral pada cangkang kerang. 3). Analisa respon biomarker antioksidan terhadap nilai indeks Kondisi kerang Placuna placenta. Analisis respon biomarker antioksidan terhadap kualitas air dan sedimen serta nilai Indeks Kondisi dilakukan dengan menggunakan PCA. Respon antioksidan yang mempunyai korelasi positif tertinggi didapatkan pada kandungan MDA di organ otot aduktor. Nilai IMP menunjukkan pada kedua lokasi masuk dalam kategori Kontaminan berat. Respon antioksidan pada kerang simping di perairan Socah lebih banyak dipengaruhi oleh logam berat, sedangkan pada perairan Ujungpangkah lebih dipengaruhi oleh PAH di air dan sedimen. Respon antioksidan terhadap nilai indeks kondisi mempunyai korelasi positif terhadap proses adaptif kerang simping pada lingkungan yang terpapar logam berat dan PAH, sedangkan MDA dari proses lipid peroksidase menyebabkan kerusakan membrane sel yang berkorelasi negatif pada nilai indeks kondisi. Perlu kajian batas aman konsumsi kerang simping dan potensi pengembangan antioksidan endogenous dari kerang simping Placuna placenta.
English Abstract
Placuna placenta scallops are a valuable commodity for coastal communities in the clam catchment area of Socah Bangkalan and Ujungpangkah Gresik. Ecological pressure in the form of pollutant substances is increasing due to human activities. This ecological pressure can cause oxidative stress on aquatic biota which is responded in the form of primary defense with antioxidant enzymes. The aims of the study were 1. To obtain antioxidant responses and Multimarker Pollution Index (IMP) values in scallop Placuna placenta from the fishing areas of Ujungpangkah and Socah fishermen, 2. To obtain the relationship between antioxidant biomarker responses to scallop habitat parameters, 3. To obtain the relationship between antioxidant biomarker responses to condition index value on scallop shells. There are 3 stages of the research method, namely: 1). Measurement of the response of biomarkers in gill organs, digestive tract and muscles in scallop shells including the antioxidants SOD, CAT, GPx and MDA, and the calculation of the Pollution Multimarker Index value was carried out based on the results of the antioxidant response with discriminant analysis and PCA analysis. 2). Analysis of the response of antioxidant biomarkers to water and sediment and shellfish quality parameters by measuring the content of heavy metals and PAHs to calculate the bioconcentration values of heavy metals and PAHs, and the mineral composition of the shells. 3). Analysis of the response of antioxidant biomarkers to the condition index value of Placuna placenta. Analysis of the response of antioxidant biomarkers to water and sediment quality and Condition Index values was carried out using PCA. The antioxidant response that had the highest positive correlation was found in the MDA content in the adductor muscle organ. The IMP value shows that both locations are in the category of heavy contaminants. The antioxidant response of scallops in Socah waters was more influenced by heavy metals, whereas in Ujungpangkah waters it was more influenced by PAHs in water and sediment. The antioxidant response to the condition index value has a positive correlation with the adaptive process of scallop shells in an environment exposed to heavy metals and PAHs, while MDA from the lipid peroxidase process causes cell membrane damage which is negatively correlated with the condition index value. It is necessary to study the safe limits of consumption of scallop shells and the potential for developing endogenous antioxidants from scallops Placuna placenta.
