Abstract
Coronavirus -disease 2019 (COVID-19) adalah | penyakit- yang sangat menular yang. disebabkan oleh SARS-CoV-2.. Coronavirus menginfeksi melalui situs mukosa. Salah satu pencegahan yang efektif adalah melalui vaksinasi yang dapat- ' menginduksi ' imunitas ” mukosa.- “Vaksin — mukosa - memungkinkan penghantaran: melalui -rute -~oral menggunakan live | bacterial- vector yaitu Lactococcus . actis. -Penelitian... ini ~bertujuan- .untuk, ;. mengetahui pengaruh pemberian protein spike High Conserved Region (HCR) SARS-CoV-2 dengan live bacterial vector L: lactis' dan asam retinoat sebagai adjuvan terhadap ekspresi sel T CD8+ pada limpa mencit BALB/c. Penelitian ' ini ‘menggunakan desain -penelitian - post . test. only control - group | design.--untuk ~mengetahui perbandingan_ekspresi sel T CD8+ setelah pemberian. vaksin oral-berbasis L. lactis “rekombinan “dengan ‘penambahan asam retinoat. Hewan coba dibagi menjadi 3 kelompok, yaitu kontrol negatif (KN), vaksin' lactis pNZHCR (KP),-dan vaksin L. factis| pPNZHCR dengan asamretinoat 300 ug- (P): -Vaksin- L. lactis PNZHCR 5x10° CFU diberikan pada hari ke-1, 21, dan 42. Adjuvan asam retinoat diberikan satu jam sebelum’pemberian vaksinasi. Mencit dieutanasia pada hari ke-60-untuk’ diambil ‘organ limpa. Pengamatan-ekspresi' sel-T-CD8+ dilakukan menggunakan:metode immunofiuorescence-dengan_antibodi: anti-CD8(SC-177 PES phycoerythrin). dan. pengukuranekspresi.menggunakan . perangkat . lunak ImageJ. Analisis menunjukkan bahwa. pemberian vaksin L. lactis pPNZHCR dan asam retinoat sebagai adjuvan menyebabkan peningkatan ekspresi-sel' T CD8+ dibandingkan :dengan’ kelompok: kontrol: negatif dan_pemberian-vaksin L. lactis pNZHCR saja,.namun peningkatan tidak signifikan secara statistik (p:>-0,05; p.= 0,204). Pemberian_vaksin dan asam retinoat disimpulkan belum adekuat dalam meningkatkan jumlah sel T'CD8+ secara signifikan'pada limpa mencit BALB/c
English Abstract
Coronavirus disease 2019 (COVID-19) is a highly contagious disease caused' 'by SARS-CoV-2.' The coronavirus infects through'the mucosal'sites. One of the most effective prevention, is by vaccination 'which-can ;induce-mucosal immunity. Mucosal.vaccines .can allow_delivery. via the oral.route using a. live bacterial vector Lactococcus lactis. This study aimed to determine the effect of administering the SARS-CoV-2/High Conserved Region (HCR) protein spike ‘with live bacterial vector. L. lactis'NZ3900 and retinoic acid as an adjuvant on:CD8+ T+ cell expression: in, the: spleen of BALB/c mice. This study used.a-post test only control _group .design to compare the expression of CD8+ T cells after administration of ‘recombinant L- /actis-based oral vaccines with the addition 'of retinoic/acid: Experimental animals were divided into 3 groups, namely negative control (KN), L. lactisspNZHCR vaccine (KP); and L. lactis-pNZHCR vaccine with 300 g retinoic acid (P). L. lactis-pNZHCR 5x10° CFU vaccine was given on days 1, 21, and 42. Retinoic ‘acid was given one hour before vaccination: Mice were euthanized“on 'the: 60th''day to collect 'spleens. /Observation of CD8+ T cell expressionwas carried . out . using- the, immunofluorescence: method -and measurement of expression using the-imageJ software. The analysis showed that administration of L. /actis-pNZHCR vaccine and- retinoic acid as an adjuvant caused an increase in 'CD8+ T-cell expression compared to the negative control group. and administration of L./ factis-pNZHCR vaccine alone, but the increase was not statistically. significant (p-> 0,05; p =.0,204). The conclusion;of this study is the administration of vaccines and retinoic acid as an adjuvant is not adequate to significantly increase the number of CD8+ T cells in spleen BALB/c mice
