Abstract

Udang vannamei (Litopenaeus vannamei) adalah salah satu jenis komoditas budidaya perikanan dengan nilai produksi tinggi. Sistem budidaya udang vannamei di Indonesia salah satunya menerapkan model budidaya intensif. Permasalahan utama yang sering ditemukan dalam kegagalan produksi udang vaname adalah buruknya kualitas air selama masa pemeliharaan, terutama pada tambak intensif. Salah satu faktor penyebab penyakit infeksi yaitu adanya patogen pada perairan budidaya dan Vibrio spp. adalah patogen yang sering ditemui di perairan budidaya. Kualitas air memiliki peranan penting bagi pertumbuhan udang vaname dan pencegahan adanya patogen. Total Organic Matter (TOM) merupakan salah satu parameter kualitas air dengan metode penentuan kandungan zat organik total dalam perairan. Penelitian hubungan Total Organic Matter dengan kelimpahan Total Vibrio Count diharapkan mampu menjadi sebuah indikator terhadap hubungan antara angka kandungan bahan organik dalam perairan dengan pertumbuhan bakteri merugikan yaitu bakteri Vibrio spp. Penelitian dilakukan pada bulan Oktober – Desember 2022 di Laboratorium Mandiri CP. Prima, Sumenep, Jawa Timur. Penelitian dilakukan pada 2 tambak yaitu tambak CV. Indah Grup dan tambak Kasmani A. Prosedur penelitian yang dilakukan yaitu dengan sterilisasi alat dan bahan, sterilisasi ruangan, pembuatan media (Tryptic Soy Agar dan Thiosulphate Citrate Bile Sucrose), perlakuan TOM, pengambilan sampel air, pengenceran sampel, penanaman sampel bakteri dan perhitungan hasil baik TOM maupun bakteri pada media TSA dan TCBS. Kelimpahan bakteri Vibrio spp. didapatkan dengan membandingkan hasil pada media TSA dengan TCBS. Parameter suhu, salinitas, ammonium dan nitrit digunakan sebagai parameter penunjang. Analisis data dilakukan dengan uji normalitas menggunakan uji normalitas Kolmogorov-smirnov dan uji korelasi menggunakan uji Rank Spearman dengan bantuan aplikasi SPSS. Hasil pengukuran TOM pada tambak CV. Indah Grup memiliki rata-rata 105,6 ppm, pada tambak Kasmani A. memiliki rata-rata 174,34 ppm. Hasil perhitungan bakteri pada media TCBS di tambak CV. Indah Grup memiliki rata-rata 9,6 x 10² CFU/ml dan pada media TSA 4,6 x 10⁴ CFU/ml. Hasil perhitungan bakteri pada media TCBS di tambak Kasmani A. memiliki rata-rata 2,4 x 10³ CFU/ml dan pada media TSA 3,8 x 10⁴ CFU/ml. Hasil uji normalitas didapatkan nilai signifikansi sebesar 0,001 pada Total Organic Matter dan Total Bakteri Vibrio 0,004. Uji korelasi diperoleh nilai koefisien korelasi sebesar 0,247 dengan nilai signifikansi 0,146. Hasil penelitian menunjukkan bahwa tidak terdapat hubungan signifikan antara perbedaan nilai TOM dengan kelimpahan bakteri Vibrio spp. pada perairan budidaya udang vanname.

English Abstract

Vannamei shrimp (Litopenaeus vannamei) is one of aquaculture commodity that has high production value. One of the vannamei shrimp cultivation systems in Indonesia is applying an intensive cultivation model. The main problem that is often found in vannamei shrimp production failure is poor water quality during the rearing period, especially in intensive ponds. One of the factors causing infectious diseases is the presence of pathogens in aquaculture waters and Vibrio spp. are pathogens that are often found in aquaculture waters. Water quality has an important role for the growth of vannamei shrimp and prevention of pathogens. Total Organic Matter (TOM) is one of the water quality parameters with a method of determining the total organic matter content in waters. Research on the relationship between Total Organic Matter and the abundance of Total Vibrio Count is expected to be an indicator of the relationship between the number of organic matter content in waters and the growth of Vibrio spp. The research was conducted in October - December 2022 at the Lab Mandiri CP. Prima, Sumenep, East Java. The research was conducted on 2 ponds, CV. Indah Group was sampled in 3 plots and Kasmani A. The research procedures carried out were sterilization of tools and materials, room sterilization, media preparation (Tryptic Soy Agar dan Thiosulphate Citrate Bile Sucrose), TOM treatment, water sampling, sample dilution, bacterial sample planting and calculation of the results of both TOM and bacteria on TSA and TCBS media. The abundance of Vibrio spp. obtained by comparing the results on TSA media with TCBS. Parameters of temperature, salinity, ammonium and nitrite are used as supporting parameters. Data analysis was performed by normality test using the Kolmogorov–Smirnov normality test and correlation test using the Rank Spearman test with the help of the SPSS application. TOM measurement results on CV. Indah Group has an average of 105.6 ppm, Kasmani A. pond has an average of 174.34 ppm. The results of bacterial counts on TCBS media in CV. Indah Group has an average of 9.6 x 10² CFU/ml and on TSA media 4.6 x 10⁴ CFU/ml. The results of the bacteria count on TCBS media in Kasmani A ponds have an average of 2.4 x 10³ CFU/ml and on TSA media 3.8 x 10⁴ CFU/ml. The normality test results obtained a significance value of 0.001 for Total Organic Matter and 0.004 for Total Vibrio Bacteria. The correlation test obtained a correlation coefficient value of 0.247 with a significance value of 0.146. The results showed that there was no significant relationship between the difference in TOM values and the abundance of Vibrio spp. in vannamei shrimp aquaculture.
