Abstract
Coronavirus Disease 2019 (COVID-19) merupakan penyakit infeksius yang disebabkan oleh severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Salah satu tindakan pencegahan dan pengendalian penyebaran COVID-19 yaitu dengan vaksinasi. Vaksin subunit protein spike SARS-CoV-2 berbasis bakteri Lactococcus lactis rekombinan yang diberikan secara oral dapat menjadi alternatif pengembangan vaksin COVID-19. Pada penelitian sebelumnya, diketahui bahwa pemberian vaksin subunit HCR dengan karier L. lactis melalui rute oral kurang dapat menginduksi antibodi IgG spesifik antigen dibandingkan dengan pemberian intranasal sehingga dibutuhkan adjuvan vaksin yang potensial untuk menginduksi respon imun yang kuat. Tujuan dilakukannya penelitian ini yaitu untuk mengetahui pengaruh pemberian adjuvan asam retinoat dalam pemberian vaksin oral COVID-19 berbasis L. lactis rekombinan terhadap kadar IgG anti-spike SARS-CoV-2 pada serum mencit BALB/c. Penelitian ini menggunakan hewan coba mencit BALB/c usia 6-8 minggu sebanyak 30 ekor yang dibagi menjadi 3 kelompok, yaitu kontrol negatif (tanpa perlakuan), kontrol positif (vaksin oral L. lactis rekombinan), dan perlakuan (vaksin oral L. lactis rekombinan ditambah adjuvan asam retinoat). Selama masa percobaan, mencit kelompok kontrol positif dan perlakuan divaksinasi sebanyak tiga kali (hari ke-1, -21, -42) melalui rute oral. Sampel darah hewan coba diambil pada hari ke-0, -42, dan -60 untuk diukur kadar IgG anti-spike SARS-CoV-2 serum menggunakan metode ELISA dan dikonfirmasi dengan uji Dot blot. Analisis statistik menunjukkan bahwa penambahan adjuvan asam retinoat pada pemberian vaksin oral subunit SARS-CoV-2 berbasis L. lactis rekombinan belum dapat menginduksi respon IgG anti-spike SARS-CoV-2 serum secara signifikan (p > 0,05) dibanding kelompok lainnya. Kesimpulan penelitian ini adalah penambahan adjuvan asam retinoat dosis 300 μg yang diberikan sebelum pemberian vaksin oral COVID-19 berbasis L. lactis rekombinan belum dapat menginduksi respon IgG anti-spike SARS-CoV-2 secara signifikan pada mencit galur BALB/c.
English Abstract
Coronavirus Disease 2019 (COVID-19) is an infectious disease caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). One of the measures to prevent and control the spread of COVID-19 through vaccination. The SARS-CoV-2 spike protein subunit vaccine is based on recombinant Lactococcus lactis bacteria that is given orally can be an alternative for developing a COVID-19 vaccine. In previous studies, it was known that administration of HCR subunit vaccines with L. lactis carriers via the oral route was less able to induce antigen-specific IgG antibodies compared to intranasal administration, so a potential vaccine adjuvant was needed to induce a strong immune response. The purpose of this study was to determine the effect of giving retinoic acid adjuvants in the administration of recombinant L. lactis-based oral COVID-19 vaccines on levels of IgG anti-spike SARS-CoV-2 in the serum of BALB/c mice. This study used 30 BALB/c mice aged 6-8 weeks which were divided into 3 groups, namely negative control (no treatment), positive control (recombinant L. lactis oral vaccine), and treatment (recombinant L. lactis oral vaccine plus retinoic acid adjuvant). During the experimental period, mice in the positive control and treatment groups were vaccinated three times (days 1, -21, -42) via the oral route. Blood samples from experimental animals were taken on days 0, -42, and -60 to measure levels of serum IgG anti-spike SARS-CoV-2 using the ELISA method and confirmed by Dot blot testing. Statistical analysis showed that the addition of retinoic acid adjuvants to the administration of recombinant L. lactis-based SARS-CoV-2 subunit oral vaccine was not able to induce a serum IgG anti-spike SARS-CoV-2 response to significantly (p > 0.05) compared to the other groups. This study concluded that the addition of a 300 μg dose of retinoic acid adjuvant given before administration of the recombinant L. lactis-based COVID-19 oral vaccine had not been able to induce a significant IgG anti-spike SARS-CoV-2 response in BALB/c strain mice.
