Abstract

Tujuan : Mengetahui perubahan histologi dan perbedaan ekspresi Insulin-like growth factor binding protein-7 (IGFBP-7) pada kornea tikus normal dan kornea tikus model Limbal stem cell deficiency (LSCD). Metode : Penelitian ini true experimental in vivo pada kornea tikus model LSCD yang dipapar Benzalkonium Klorida (BAK) 0.5% empat kali sehari selama 30 hari. Pada akhir perlakuan dilakukan pemeriksaan slit lamp biomikroskop (Inami L-0189) dan pewarnaan fluoresen untuk melihat tanda klinis tikus model LSCD, kemudian dilakukan enukleasi dan pembuatan preparat untuk pengamatan morfologi jaringan kornea dengan pemeriksaan Hematosiklin Eosin (HE), serta imunofluoresen untuk melihat ekspresi IGFBP-7. Hasil : Pemberian BAK 0.5% empat kali sehari selama 30 hari dapat menginduksi LSCD pada kornea tikus model yang ditandai dengan neovaskularisasi kornea, defek epitel kornea serta fluoeresin positf. Pada pemeriksaan HE didapatkan adanya neovaskularisasi dan hyperplasia sel keratosit pada lapisan stroma. Ekspresi IGFBP-7 pada kornea tikus normal dan kornea tikus model LSCD didapatkan perbedaan signifikan (p<0.05). Kesimpulan : Terdapat perubahan histologi dan perbedaan signifikan ekspresi IGFBP- 7 pada kornea tikus normal dan kornea tikus model LSCD.

English Abstract

Objective: To determine the histological change and Insulin-like Growth Factor Binding Protein-7 (IGFBP-7) expression differences in cornea of normal mouse and LSCD model. Methods: This was a true, in vivo experimental study of the LSCD mouse model exposed to Benzalkonium chloride (BAC) 0.5% four times a day for 30 days. At the end of the treatment, slit lamp biomicroscope (Inami L-0189) examination and fluorescent staining were performed to evaluate clinical signs of LSCD. Enucleation of euthanized mouse and hematoxylin eosin (HE) staining were performed to observe the morphology of corneal tissue. Moreover, the immunofluorescence staining was performed to examine IGFBP-7 expression. Results: BAC 0.5% four times per day for 30 days successfully induced the typical manifestations of LSCD including neovascularization, corneal epithelial defect, and positive fluoerescen. Neovascularization and hyperplasia of keratocyte cells in the stromal layer were obtained in HE staining. IGFBP-7 expression was significantly difference between the cornea of normal mouse and LSCD model (p 0.05). Conclusion: There are histological change and significant differences in IGFBP-7 expression in normal mouse corneas and LSCD-model mouse corneas.
