Abstract

Bakteri merugikan merupakan penyebab munculnya suatu penyakit, salah satunya adalah jenis bakteri Vibrio spp. yang dominan menyerang pada udang vaname. Perlakuan perbedaan salinitas dalam budidaya udang vaname memiliki dampak bagi organisme budidaya yang diharapkan mampu menekan kelimpahan bakteri merugikan. Rumusan masalah dalam penelitian ini adalah membandingkan perlakuan salinitas yang berbeda dalam menekan kelimpahan bakteri Vibrio spp. tambak udang vaname (Litopenaeus vannamei). Tujuan peneltian ini adalah mengetahui pengaruh perbandingan perlakuan dalam menekan kelimpahan bakteri Vibrio spp. Penelitian ini dilaksanakan pada bulan November - Desember 2022 di Laboratorium Kualitas Air Binaan PT. Central Proteina Prima Tuban, Jawa Timur. Metode penelitian yang digunakan adalah metode eksperimental. Perlakuan dilakukan dengan membandingkan antar kelompok “Salinitas 11” dan “Salinitas 35”. Masing-masing menggunakan 2 petakan dan perhitungan bakteri dilakukan setiap minggu selama 4 minggu. Perhitungan koloni bakteri menggunakan rumus ((jumlah koloni x (1/d)). Persentase kelimpahan vibrio dihitung dengan rumus ((total vibrio/total bakteri umum) x 100%)). Analisis data yang digunakan dalam penelitian ini menggunakan uji T-Tidak Berpasangan dibantu software IBM SPSS Statistics versi 26.0 for Windows dengan taraf kepercayaan 95% (α=0,05). Rancangan penelitian yang digunakan adalah menggunakan statistik parametrik Independent Sampel T-Test (Uji T-Tidak Berpasangan) didukung dengan uji normalitas dan uji homogenitas. Hasil penelitian identifikasi bakteri Vibrio spp. menggunakan media TCBS (Thiosulfat Citrate Bile Salts) didapatkan pertumbuhan bakteri dominan berwarna kuning 70%. Kelimpahan bakteri umum tertinggi terjadi pada minggu 3 di petakan 22 “Salinitas 35” (3,22 x 103 cfu/ml) dan paling rendah pada minggu 3 petakan 20 “Salinitas 11” (1,3 x 102 cfu/ml). Total bakteri Vibrio spp. tertinggi terjadi pada minggu 4 petakan 22 “Salinitas 35” (2,91 x 103 cfu/ml) dan paling rendah pada minggu 1 pada petakan 22 “Salinitas 35” (1,1 x 102 cfu/ml). “Salinitas 11” menunjukkan performa yang relatif stabil dalam menurunkan persentase Vibrio spp. namun hanya dalam waktu singkat, sedangkan “Salinitas 35” menunjukkan performa yang relatif cepat walaupun sempat terjadi kenaikan bakteri. Hasil uji T-Tidak Berpasangan menggunakan SPSS sebesar 0,412 (Sig. 2 tailed > 0,05). Kesimpulan penelitian ini tidak terdapat perbedaan yang signifikan antara perlakuan salinitas <15 maupun >30 (“Salinitas 11” dan “Sallinitas 35”) dalam menekan kelimpahan bakteri Vibrio spp. pada tambak budidaya udang vaname. Cuaca fluktuatif, namun rentang nilai parameter kualitas air cenderung stabil dan memiliki pola yang sama setiap minggunya.

English Abstract

Harmful bacteria are the cause of the emergence disease. Bacteria Vibrio spp. are one of the dominant bacteria which attack vaname shrimp. The treatment of differences salinity in vaname shrimp cultivation has an impact on cultured organisms which are expected to suppress the abundance of harmful bacteria. The formulation of the problem in this study was to compare different salinity treatments for suppressing the abundance of Vibrio spp. in vaname shrimp (Litopenaeus vannamei) ponds. The purpose of this study was to determine the effect of treatment comparisons in suppressing the abundance of Vibrio spp. bacteria. This research was conducted in November - December 2022 at Water Quality Laboratory of PT. Central Proteina Prima Tuban, East Java. The research method used is the experimental method. The treatment was carried out by comparing the "Salinity 11" and "Salinity 35" groups between groups. Each used 2 pond and bacterial counts were carried out every week for 4 weeks. Calculation of bacterial colonies using the formula ((number of colonies x (1/d)). The percentage of vibrio abundance was calculated by the formula ((total vibrios/total common bacteria) x 100%)). Data analysis used in this study used the unpaired T-test assisted by IBM SPSS Statistics software version 26.0 for Windows with a 95% confidence level (α=0.05). The research design used was the Independent Sample T-Test parametric statistic (Independent T-Test) supported by the normality test and homogeneity test. The results of the research on the identification of Vibrio spp. using TCBS (Thiosulfate Citrate Bile Salts) medium, the dominant bacterial growth was obtained in color yellow 70%. The highest abundance of common bacteria occurred at week 3 on the pond 22 "Salinity 35" (3.22 x 103 cfu/ml) and lowest at week 3 pond 20 “Salinity 11” (1.3 x 102 cfu/ml). Total bacteria Vibrio spp. highest occurred at week 4 pond 22 “Salinity 35” (2.91 x 103 cfu/ml) and lowest at week 1 on pond 22 “Salinity 35” (1.1 x 102 cfu/ml). "Salinity 11" shows a relatively stable performance in reducing the percentage of Vibrio spp. but only for a short time, while "Salinity 35" showed a relatively fast performance even though there had been an increase in bacteria. The results of the Unpaired T-test using SPSS were 0.412 (Sig. 2 tailed > 0.05). The conclusion of this study was that there was no significant difference between the treatments of salinity <15 and >30 ("Salinity 11" and "Sallinity 35") in suppressing the abundance of Vibrio spp. in vaname shrimp aquaculture ponds. The weather fluctuates, but the range of water quality parameter values tends to be stable and has the same pattern every week.
