Abstract
Cumi salah satu komoditas perikanan yang digemari oleh masyarakat. Proses pengawetan berupa cumi asin bertujuan untuk mencegah pembusukan. Formalin disalahgunakan sebagai bahan pengawet berbahaya pada cumi asin. Formalin dilarang digunakan sebagai pengawet pangan berdasarkan Peraturan Menteri Kesehatan Republik Indonesia Nomor 33 Tahun 2012. Tujuan penelitian ini untuk mengetahui metode ekstraksi formalin dalam sampel yang optimum menggunakan metode UAE dan RSM – BBD serta untuk mendeteksi adanya kandungan formalin pada sampel cumi asin yang dijual di Pasar Tradisional Kota Malang. Metode penelitian ini menggunakan Spektrofotometer UV-VIS dengan reagen Nash dan RSM - BBD. Hasil ekstraksi formalin optimum diperoleh pada suhu 43oC, rasio pelarut terhadap sampel 10:1 mL/g, dan waktu reaksi 12 menit. Hasil uji spesifitas cumi asin dengan spektrofotometer UV-VIS memperoleh panjang gelombang maksimum 412 nm. Hasil uji linearitas dengan konsentrasi 2 - 10 ppm, y = 0,1665x – 0,217 dengan r2 = 0,9968. Hasil uji LOD dan LOQ diperoleh sebesar 0,099 ppm dan 0,301 ppm. Hasil uji akurasi dan presisi memperoleh %recovery sebesar 98,23% dan %RSD sebesar 1,6%. Hasil penetapan kadar pada sampel cumi asin diperoleh hasil positif kandungan formalin dengan kadar sebesar 19 – 22 ppm. Hasil uji ANOVA menujukkan nilai p<0,05 artinya terdapat perbedaan signifikan pada populasi data penelitian.
English Abstract
Squid is one of the fisheries commodities favored by the community. The preservation process in the form of salted squid aims to prevent spoilage. Formalin is misused as a dangerous preservative in salted squid. Used of formalin is prohibited as a food preservative based on the Law of the Minister of Health of the Republic of Indonesia number 33 of 2012. This study aims to determine the optimal method of formalin extraction in samples using the UAE and RSM – BBD methods and to detect the presence of formalin in salted squid samples sold in the traditional market of Malang City. This research method used a UV-VIS spectrophotometer with Nash reagents and RSM - BBD. The result is optimum extraction at a temperature of 43oC, a solvent-to-sample ratio of 10:1 mL/g, and a reaction time of 12 minutes. The specificity test results for salted squid with a UV-VIS spectrophotometer obtained a maximum wavelength of 412 nm. The results of the linearity test with a concentration of 2-10 ppm, y = 0.1665x - 0.217 with r2 = 0.9968. The LOD and LOQ test results obtained were 0.099 ppm and 0.301 ppm. The results of the accuracy and precision tests obtained a %recovery of 98.23% and % an RSD of 1.6%. The results of the method in the salted squid sample were positive formalin with levels of 19 – 22 ppm). The results of the ANOVA test showed p-value = 0.00 (p<0.05), which means there is a significant difference in the study data.
