Abstract
Coronavirus disease 2019 (COVID-19) merupakan penyakit infeksi menular yang disebabkan oleh severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Vaksinasi dengan rute oral diketahui dapat menginduksi respon imun humoral. Pada penelitian ini plasmid bakteri Lactococcus lactis (L. lactis) dikonstruksi dengan gen highly conserved region (HCR) dari protein spike SARS-CoV-2. Pada vaksin berbasis bakteri asam laktat, strategi penambahan adjuvan diperlukan untuk menginduksi respon imun yang kuat dan tahan lama, salah satunya adalah adjuvan asam retinoat. Penelitian ini bertujuan untuk membandingkan kadar IgA anti-spike SARS-CoV-2 antara mencit galur BALB/c yang diberikan vaksin oral COVID-19 berbasis L. lactis rekombinan dengan penambahan atau tanpa penambahan adjuvan asam retinoat. Mencit dikelompokkan menjadi kontrol negatif (tanpa perlakuan) (n=5), kontrol positif (vaksinasi L. Lactis rekombinan 5x109 CFU) (n=12), dan perlakuan (vaksinasi L. Lactis rekombinan 5x109 CFU dan adjuvan asam retinoat 300 μg 1 jam sebelum vaksinasi secara per oral) (n=13). Vaksinasi dilakukan pada hari ke 1, 21, dan 42. Sampel serum darah diambil pada hari ke 0, 42, dan 60. Kadar IgA diukur menggunakan uji ELISA. Hasil uji one way ANOVA menunjukkan tidak terdapat perbedaan kadar IgA yang siginifikan antar kelompok. Hasil uji repeated ANOVA antar waktu pengambilan sampel pada tiap kelompok menunjukkan bahwa kelompok perlakuan mengalami peningkatan kadar IgA yang signifikan (p < 0,05) pada hari ke 0 dan 60. Kesimpulan dalam penelitian ini yaitu penambahan adjuvan asam retinoat pada pemberian vaksin dapat meningkatkan kadar IgA namun tidak secara signifikan.
English Abstract
Coronavirus disease 2019 (COVID-19) is an infectious disease caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Vaccination through the oral route is known to induce a humoral immune response. In this study, the Lactococcus lactis (L. lactis) plasmid was constructed with the highly conserved region (HCR) gene of the SARS-CoV-2 spike protein. In lactic acid bacteria-based vaccines, the strategy of adding adjuvants is necessary to induce a strong and long-lasting immune response, one of which is retinoic acid adjuvant. This study aims to compare the levels of SARS-CoV-2 anti-spike IgA between BALB/c strain mice given the recombinant L. lactis COVID-19 oral vaccine with or without the addition of retinoic acid adjuvant. Mice were grouped into negative control (no treatment) (n=5), positive control (recombinant L. lactis vaccination 5x109 CFU) (n=12), and treatment (recombinant L. lactis vaccination 5x109 CFU and Retinoic acid adjuvant was administered at a dose of 300 μg one hour prior to oral vaccination) (n=13). Vaccinations were carried out on days 1, 21, and 42. Blood serum samples were taken on days 0, 42, and 60. IgA levels were measured using the ELISA test. The one way ANOVA test results indicated no significant difference in IgA levels among the groups. The repeated ANOVA test results for the sampling time within each group showed that the treatment group had a significant increase in IgA levels (p < 0.05) on days 0 and 60. The conclusion of this study is that the addition of retinoic acid adjuvant to vaccine administration can increase IgA levels, but not significantly.
