Abstract
Di Indonesia penyakit dan parasit potensial menyebar dan menginfeksi pada sistem budidaya. Penyakit utama ikan adalah penyakit yang disebabkan oleh bakteri maupun virus. Beberapa penyakit akibat bakteri Edwardsiella tarda, Aeromonas hydrophila, Vibrio sp., dan Mycobacterium sp. Berbagai masalah dalam budidaya di Indonesia sejak tahun 1990 hingga sekarang yang disebabkan oleh berbagai jenis bakteri vibrio, salah satunya yaitu Vibrio harveyi. V. harveyi adalah spesies bakteri penyebab penyakit kunang-kunang (lumirescent vibriosis) atau penyakit fluorescent. Penularan penyakit ini sangat mudah, karena dapat diturunkan oleh organisme yang bergerak. Penelitian dilaksanakan pada bulan Desember 2023 hingga Februari 2023 di Laboratorium Budidaya Ikan Divisi Parasit dan Penyakit Ikan dan Laboratorium Eksplorasi Sumberdaya Perikanan dan Kelautan, Fakultas Perikanan dan Ilmu Kelautan, Universitas Brawijaya Malang. Metode penelitian yang digunakan metode eksperimental. Penelitian dengan menggunakan variable bebas berupa perlakuan pemberian ekstrak kasar daun pandan wangi dengan perlakuan yang diberikan adalah perbedaan konsentrasi ekstrak kasar daun pandan wangi terhadap bakteri V. harveyi. Dengan melakukan uji pendahuluan terlebih dahulu. Setelah mendapatkan dosis, dilakukan uji inti dimulai dari dosis 50 ppm sebagai dosis terkecil dan dosis terbesar sebesar 250 ppm dengan selisih masing-masing perlakuan 50 ppm. Dengan langkah-langkah persiapan sampel, sterilisasi alat, pembuatan media, identifikasi bakteri dan uji cakram. Setelah itu dilakukan analisis secara statistic menggunakan analisis keragaman atau uji F (ANOVA) sesuai dengan rancangan yang digunakan yaitu Rancangan Acak Lengkap (RAL) dengan selang kepercayaan yaitu 95% Hasil penelitian yang sudah dilakukan tentang uji daya hambat ekstrak kasar daun pandan wangi (Pandanus amaryllifolius Roxb) terhadap bakteri Vibrio harveyi secara in vitro didapatkan zona daya hambat pada perlakuan yang digunakan yaitu dengan 5 dosis yang berbeda A (50 ppm), B (100 ppm), C (150 ppm), D (200 ppm), E (250 ppm) dan kontrol positif menggunakan antibiotik jenis ciprofloxacin dengan dosis sebesar 30 ppm, kontrol negative hanya diberi perlakuan direndam akuades. Didapatkan bahwa ekstrak kasar daun pandan wangi berpengaruh terhadap bakteri V. harveyi. Zona bening dengan nilai terbaik yaitu pada perlakuan E (250 ppm) dengan nilai rerata zona hambat sebesar 8,89 mm. Nilai daya hambat yang dihasilkan ekstrak kasar daun pandan wangi terhadap V. harveyi yaitu semakin tinggi pemberian dosis maka semakin tinggi juga zona hambat yang dihasilkan. Bakteri V. harveyi sensitif terhadap pemberian perlakuan ekstrak kasar daun pandan wangi dengan cara menghambat dan membunuh dapat dikatakan sebagai bakterisidal.
English Abstract
In Indonesia, diseases and parasites can potentially spread and invade aquaculture systems. The primary fish diseases are those caused by bacteria and viruses—several diseases are caused by bacteria such as Edwardsiella tarda, Aeromonas hydrophila, Vibrio sp., and Mycobacterium sp. Various problems in aquaculture in Indonesia since 1990 until now have been caused by various types of vibrio bacteria, one of which is Vibrio harveyi. V. harveyi is a bacterial species that causes luminescent vibriosis, or fluorescent disease. Transmission of this disease is effortless, as moving organisms can pass it on. The research was conducted from December 2023 to February 2023 at the Fish Cultivation Laboratory of the Division of Fish Parasites and Diseases and the Fisheries and Marine Resources Exploration Laboratory, Faculty of Fisheries and Marine Sciences, Universitas Brawijaya Malang. The research method used was an experimental method. The research using independent variables in the form of treatment of fragrant pandanus leaf extract with the treatment given is the difference in concentration of fragrant pandanus leaf extract against V. harveyi bacteria. By doing preliminary tests first. After getting the dose, the core test starts with a dose of 50 ppm as the smallest dose and the largest dose of 250 ppm, with a difference of 50 ppm for each treatment. With the steps of sample preparation, tool sterilization, media preparation, bacterial identification and disc testing. After that, statistical analysis was carried out using the analysis of variance, or F test (ANOVA), in accordance with the design used, namely, a Completely Randomized Design (RAL) with a confidence interval of 95%. The results of research that has been carried out on the inhibition test of fragrant pandanus leaf extract (Pandanus amaryllifolius Roxb) against Vibrio harveyi bacteria in vitro obtained inhibition zones in the treatments used, namely with five different doses A (50 ppm), B (100 ppm), C (150 ppm), D (200 ppm), E (250 ppm), and positive control using ciprofloxacin type antibiotics at a dose of 30 ppm, a negative control only treated with distilled water. It was found that fragrant pandanus leaf extract affected V. harveyi bacteria. The clear zone with the best value is treatment E (250 ppm), with an average inhibition zone value of 8.89 mm. The value of the inhibition produced by fragrant pandanus leaf extract against V. harveyi is that the higher the dose, the higher the inhibition zone produced. V. harveyi bacteria are sensitive to the treatment of fragrant pandanus leaf extract by inhibiting and killing it, which can be said to be bactericidal.
