Abstract
Coronavirus disease 2019 (COVID-19) merupakan penyakit infeksius pada saluran pernapasan akibat severe acute respiratory syndrome coronavirus 2 (SARS�CoV-2). Vaksinasi rute oral banyak menarik perhatian karena bersifat non-invasif tetapi memiliki keterbatasan dalam segi stabilitas, sehingga dikembangkan sistem penghantaran menggunakan bakteri seperti Lactococcus lactis (L.lactis). Pada penelitian ini plasmid bakteri L.lactis dikonstruksi dengan gen highly conserved region (HCR) dari protein spike SARS-CoV-2. Pada penelitian yang telah dilakukan sebelumnya, desain vaksin dengan vektor L. Lactis rekombinan mampu menginduksi respon imun seluler melalui peningkatan sel T CD4+ dengan hasil yang tidak signifikan, sehingga pada penelitian ini dikembangkan dengan menambahkan adjuvan. Penelitian ini bertujuan untuk mengukur ekspresi sel T CD4+ pada limpa mencit yang diberikan vaksinasi dengan atau tanpa penambahan adjuvan asam retinoat. Mencit BALB/c jantan berusia 6-8 minggu dibagi dalam 3 kelompok yaitu, kontrol negatif (tanpa perlakuan), kontrol positif (vaksin L.lactis rekombinan), perlakuan (vaksin L.lactis rekombinan dan adjuvan asam retinoat). Vaksinasi dilakukan pada hari ke-1, 21, dan 42, asam retinoat diberikan 1 jam sebelum vaksinasi. Mencit dikorbankan pada hari ke-60 dengan mengambil organ limpa untuk dibuat menjadi preparat histologi. Intensitas sel T CD4+ diukur dengan metode imunofluoresensi. Analisa statistik menunjukkan bahwa penambahan adjuvan asam retinoat mampu menginduksi respon imunitas seluler sel T CD4+ pada mencit galur BALB/c, tetapi tidak terdapat perbedaan signifikan dibandingkan dengan kontrol. Kesimpulan dari penelitian ini yaitu pemberian vaksin oral COVID-19 menggunakan karier L. lactis rekombinan dengan penambahan adjuvan asam retinoat mampu meningkatkan ekspresi sel T CD4+ pada limpa dibandingkan kelompok yang tidak menerima adjuvan walaupun tidak signifikan secara statistik.
English Abstract
Coronavirus disease 2019 (COVID-19) is an infectious respiratory disease due to severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Oral route vaccination has attracted much attention because it is non-invasive but has limitations in terms of stability, therefore delivery system using bacteria such as Lactococcus lactis (L. lactis). In this study, bacterial plasmids were constructed with highly conserve region (HCR) gene from spike protein of SARS-CoV-2. In previous studies, a vaccine design with recombinant L. Lactis-based was able to induce a cellular immune response by increasing CD4+ T cells with insignificant results, so this study was developed by adding adjuvants. This study aims to measure the expression of CD4+ T cells in the spleens of mice vaccinated with or without retinoic acid adjuvant. Male BALB/c mice aged 6-8 weeks were divided into 3 groups: negative control (no treatment), positive control (L.lactis recombinant vaccine), and treatment (L.lactis recombinant vaccine and retinoic acid adjuvant). Vaccination was carried out on day�1, 21 and 42, and also retinoic acid was given 1 hour before vaccination. Mice were sacrificed on the 60th day by harvesting the spleen to make histological preparations. The intensity of CD4+ T cells was measured by the immunofluorescence method. Statistical analysis showed that the addition of retinoic acid adjuvant was able to induce a cellular immune response of CD4+ T cells in BALB/c mice, but there was no significant difference compared to controls. The conclusion of this study is that the administration of oral COVID-19 vaccine using recombinant L.lactis carrier with the addition of retinoic acid adjuvant can enhance CD4+ T cell expression in the spleen compared to the group that did not receive the adjuvant, although not statistically significant.
