Abstract

Androgen Deprivation Therapy (ADT) merupakan terapi hormonal pada pasien kanker prostat untuk menurunkan kadar hormone testosterone dalam sirkulasi. Efek samping ADT, khususnya orkidektomi adalah menurunkan massa otot skeletal. Selective Androgen Receptor Modulator (SARM) RAD 140 adalah kelas ligan reseptor androgen yang digunakan untuk mencegah efek samping ADT. Tujuan penelitian ini adalah untuk mengetahui efek pemberian terapi SARM-RAD 140 terhadap massa otot skeletal pasca orkidektomi bilateral. Desain penelitian true experimental-randomized post test only control group design secara in vivo pada Rattus norvegicus yang dilakukan orkidektomi bilateral. Masing-masing kelompok adalah kontrol negatif, kontrol positif, dan perlakuan SARM RAD 140 dengan dosis 0.03 mg/kg, 0.1 mg/kg, 1 mg/kg, 3 mg/kg, 10 mg/kg selama 6 minggu. Variabel terikat adalah besar cross sectional area, myonuclei, massa otot gastrocnemius pada tikus. Analisis data menggunakan SPSS versi 20. Tidak terdapat perbedaan data berat badan tikus yang signifikan antar kelompok perlakuan dosis SARM RAD 140 (p>0,05). Terdapat perbedaan yang signifikan pada CSA otot gastrocnemius tikus (p=0.000), myonuclei sel otot gastrocnemius (p=0.000), dan massa otot gastrocnemius (p=0.000) antar kelompok perlakuan dosis SARM RAD 140. Prosedur orkidektomi secara signifikan mampu menurunkan berat badan hewan coba. Selama delapan minggu, berat badan tikus yang diberi terapi SARM meningkat setiap tujuh hari sebesar 10%. Pemberian SARM pada hewan dengan orkidektomi bilateral mampu meningkatkan massa otot melalui gambaran peningkatan ukuran CSA dan meningkatkan jumlah myonuclei. SARM RAD 140 3mg/kg efektif untuk meningkatkan ukuran CSA otot, jumlah myonuclei otot, dan massa otot tikus Rattus norvegicus dengan orkidektomi bilateral.

English Abstract

Androgen Deprivation Therapy (ADT) is a hormonal therapy for prostate cancer patients to reduce testosterone levels in circulation. A side effect of ADT, especially orchidectomy, is a decrease in skeletal muscle mass. Selective Androgen Receptor Modulator (SARM) RAD 140 is a class of androgen receptor ligand used to prevent the side effects of ADT. The purpose of this study was to determine the effect of SARM-RAD 140 therapy on skeletal muscle mass after bilateral orchidectomy. The research design was true experimental-randomized post test only control group design in vivo on Rattus norvegicus which underwent bilateral orchidectomy. Each group was a negative control, positive control, and SARM RAD 140 treatment at a dose of 0.03 mg/kg, 0.1 mg/kg, 1 mg/kg, 3 mg/kg, 10 mg/kg for 6 weeks. The dependent variable is cross sectional area, myonuclei,, gastrocnemius muscle mass in rats. Data analysis using SPSS version 20. There was no significant difference in mice body weight data between groups treated with SARM RAD 140 dose (p>0.05). There were significant differences in CSA gastrocnemius muscle of mice (p=0.000), myonuclei, of gastrocnemius muscle cells (p=0.000), and gastrocnemius muscle mass (p=0.000) between groups treated with SARM RAD 140 dose. The orchidectomy procedure significantly reduced the weight of the experimental animals. For eight weeks, the body weight of the mice treated with SARM increased every seven days by 10%. Administration of SARM to animals with bilateral orchidectomy can increase muscle mass by increasing the size of the CSA and increasing the number of myonuclei. SARM RAD 140 3mg/kg was effective to increase CSA muscle size, number of myonuclei, and muscle mass of Rattus norvegicus mice with bilateral orchidectomy.
