Abstract
Modified Cassava Starch (MCS) merupakan salah satu jenis starch yang dapat membentuk gel sehingga cenderung memiliki viskositas yang tinggi sehingga dapat menyebabkan pelepasan obat lebih lambat dan dapat digunakan sebagai matriks pada sediaan sustained release. Hal ini dapat terjadi karena MCS memiliki kandungan amilopektin yang besar. Selain penggunaan starch, digunakan pula HPMC dan Na CMC sebagai matriks karena keduanya mampu membentuk lapisan gel yang menyebabkan pelepasan obat lebih lambat. Tujuan penelitian ini adalah untuk mengetahui formulasi optimum tablet sustained release menggunakan MCS, HPMC, dan Na CMC, memperoleh polimer terbaik, serta melihat perbedaan antar formula melalui profil disolusinya. Pembuatan tablet menggunakan metode kempa langsung dengan asam mefenamat sebagai model obat hingga mendapatkan 7 rancangan formula yaitu: F1 (100% MCS), F2 (100% HPMC), F3 (100% Na CMC), F4 (50% MCS dan 50% HPMC), F5 (50% MCS dan 50% Na CMC), F6 (50% HPMC dan 50% Na CMC), dan F7 (33,33% MCS, 33,33% HPMC, dan 33,33% Na CMC). Dilakukan uji evaluasi tablet meliputi uji keseragaman ukuran, uji keseragaman bobot, uji kekerasan, uji friabilitas dan uji profil disolusi yang dilakukan selama 12 jam. Hasil dari uji profil disolusi didapatkan formula optimum dengan kombinasi polimer MCS dan Na CMC karena dapat melepaskan zat aktif paling besar pada jam ke-6. Pada formula ini, Na CMC memiliki pengaruh paling besar dalam mempertahankan laju disolusi dan MCS memiliki kelarutan yang baik sehingga kombinasi dari MCS dan Na CMC dapat mempertahankan dan melarutkan zat aktif dengan baik sehingga pelepasannya dapat terkontrol. Formula ini memiliki metode pelepasan gabungan antara difusi dan erosi yang dapat dilihat dari nilai eksponen pelepasannya. Disarankan pada penelitian lebih lanjut untuk penyempurnaan formula kembali agar dapat melepaskan obat segera setelah penggunaan dan dapat bertahan hingga 12 jam serta penggunaan alat kempa dengan kekuatan tetap.
English Abstract
Modified Cassava Starch (MCS) is a type of starch that can form a gel so it tends to have a high viscosity that can cause slower drug release and can be used as a matrix in sustained release preparations. This can happen because MCS has a large amylopectin content. In addition to the use of starch, HPMC and Na CMC were also used as matrices because both of them were able to form a gel layer which resulted in slower drug release. The aim of this study was to determine the optimum formulation of sustained release tablets using MCS, HPMC, and Na CMC, to obtain the best polymer, and to see the differences between the formulas through their dissolution profiles. Preparation of tablets using direct compression method with mefenamic acid as a drug model to obtain 7 draft formulas namely: F1 (100% MCS), F2 (100% HPMC), F3 (100% Na CMC), F4 (50% MCS and 50% HPMC ), F5 (50% MCS and 50% Na CMC), F6 (50% HPMC and 50% Na CMC), and F7 (33,33% MCS, 33,33% HPMC, and 33,33% Na CMC). Tablet evaluation tests were carried out including size uniformity tests, weight uniformity tests, hardness tests, fribility tests and profil dissolution tests which were carried out for 12 hours. The results of the dissolution profile test obtained the optimum formula with the combination of MCS polymer and CMC Na because it can release the active substance the greatest in the 6th hour. In this formula, Na CMC has the greatest influence in maintaining the dissolution rate and MCS has good solubility so that the combination of MCS and Na CMC can retain and dissolve the active substance well so that its release can be controlled. This formula has a combined release method between diffusion and erosion which can be seen from the exponen release. It is recommended for further research to refine the formula again that can release the drug immediately after use and can last up to 12 hours and use a press with constant strength.
