Abstract
Sumber daya perikanan di Indonesia berasal dari perikanan tangkap dan perikanan budidaya. Perikanan budidaya berdasarkan pengelolaannya dibagi menjadi 3, yakni tradisional, semi intensif, dan intensif. Budidaya intensif menerapkan sistem padat tebar yang tinggi sehingga memberikan hasil produksi yang tinggi. Masalah utama yang menyebabkan produktivitas rendah yakni adanya serangan penyakit. Bakteri V. harveyi merupakan bakteri yang dapat menyebabkan penyakit vibriosis yang menyerang hewan laut dan payau, bersifat sangat akut dan ganas karena menyebabkan mortalitas sampai dengan 100% dalam waktu 1-3 hari. Penyakit vibriosis dapat diatasi dengan penggunaan antibiotik, namun dapat menyebabkan resistensi dan residu. Alternatif pengobatan dapat menggunakan fitoterapi dari ekstrak daun jambu air (S. aqueum). Daun jambu air dipilih karena mengandung senyawa metabolit sekunder seperti alkaloid, saponin, tanin, dan flavonoid yang bermanfaat sebagai antibakteri. Penelitian ini bertujuan untuk mengetahui pengaruh penggunaan ekstrak kasar daun jambu air (S. aqueum) dengan dosis yang berbeda sebagai salah satu bahan uji daya hambat bakteri V. harveyi secara in vitro. Penelitian ini dilakukan di Laboratorium Budidaya Ikan Divisi Penyakit dan Kesehatan Ikan, Fakultas Perikanan dan Ilmu Kelautan Universitas Brawijaya pada bulan Januari – Februari 2023. Penelitian ini menggunakan metode eksperimental yang digunakan untuk mengetahui keterkaitan variabel bebas dengan variabel terikat. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan lima perlakuan meliputi dosis A (25 ppm), B (50 ppm), C (75 ppm), D (100 ppm), dan E (125 ppm) dengan ulangan masing-masing tiga kali serta kontrol positif berupa antibiotik ciprofloxacin 30 ppm dan kontrol negatif menggunakan kertas cakram tanpa perlakuan apapun. Hasil uji fitokimia ekstrak kasar etanol 96% daun jambu air (S. aqueum) mengandung senyawa metabolit sekunder berupa flavonoid, tanin, saponin, dan alkaloid yang dapat menghambat pertumbuhan bakteri. Hasil penelitian ini menunjukkan bahwa pemberian ekstrak daun jambu air (S. aqueum) dengan dosis yang berbeda memberikan pengaruh yang berbeda nyata terhadap pertumbuhan bakteri V. harveyi. Diameter zona bening tertinggi diperoleh pada perlakuan E (125 ppm) dengan rata-rata diameter zona bening 9,26 mm dan diameter terendah diperoleh pada perlakuan A (25 ppm) dengan rata-rata diameter sebesar 7,94 mm. Hubungan antara dosis ekstrak kasar daun jambu air (S. aqueum) terhadap diameter zona bening yang terbentuk pada bakteri V. harveyi menghasilkan persamaan linier y= 0,0127x + 7,526 dengan nilai determinasi R2 = 0,789. Kekuatan hambat ekstrak daun jambu air (S. aqueum) terhadap bakteri V. harveyi termasuk sedang dan bersifat bakteriostatik.
English Abstract
Indonesian fishery resources these kinds of resources mostly come from direct fishing and aquaculture. When it comes to aquaculture, this kind of fishery industry is divided into 3 methods, called traditional, semi-intensive and intensive. Intensive cultivation applies a high stocking density system provide high production yields. The main problem that causes low productivity in intensive culture is disease. V. harveyi is a bacterium can cause vibriosis which infect marine and brackish organisme, is very acute and malignant because it causes mortality of up to 100% within 1-3 days. Vibriosis disease can be overcome by using antibiotics, but it can cause resistance and residue. Alternative treatment that can be used is phytotherapy from crude extract of watery rose apple leaves (S. aqueum). watery rose apple were chosen because it contain secondary metabolites such as alkaloids, saponins, tannins, and flavonoids which are useful as antibacterials. This research aims to determine the effect of using crude extract of guava leaves (S. aqueum) with different doses as a test material for the inhibition of V. harveyi bacteria in vitro. This research was conducted at the aquaculture Laboratory, Fish Disease and Health Division, Faculty of Fisheries and Marine Sciences, Universitas Brawijaya, from January to February 2023. This research used an experimental method which was used to determine the relationship between independent variables and dependent variables. This research used a completely randomized design (CRD) with five treatment doses A (25 ppm), B (50 ppm), C (75 ppm), D (100 ppm), and E (125 ppm) with three replications in each. Treatment well as a positive control using ciprofloxacin 30 ppm and a negative control without any treatment. Based on phytochemicals test results of 96% ethanol crude extract of watery rose apple leaves (S. aqueum), it shows the result contain secondary metabolites in the form of flavonoids, tannins, saponins, and alkaloids which can inhibit bacterial growth. The results of this study showed that crude extract of watery rose apple leaves (S. aqueum) with different doses had a significantly different effect on the growth of V. harveyi bacteria. The highest clear zone diameter was obtained in treatment E (125 ppm) with an average clear zone diameter of 9,26 mm and the lowest diameter was obtained in treatment A (25 ppm) 7,94 mm. The relationship between the dose of crude extract of watery rose apple leaves (S. aqueum) and the diameter of the clear zone formed on V. harveyi bacteria produces a linear equation y = 0,0213x + 6,719 with a determination value of R2 = 0,789. The inhibition power of crude extract of watery rose apple leaves (S. aqueum) against V. harveyi bacteria was included in the moderate category and was bacteriostatic.
