Abstract
Indonesia merupakan salah satu negara yang memiliki kawasan hutan mangrove yang menjadi habitat mikroalga Aurantiochytium sp.. Aurantiochytrium sp. mampu menghasilkan asam lemak seperti lemak tak jenuh tunggal (Monounsaturated Fatty Acid / MUFA) dan asam lemak tak jenuh ganda (Polyunsaturated Fatty Aicd / PUFA), terutama docosahexaenoic acid (DHA). Salah satu faktor pembatas dalam pertumbuhan Aurantiochytrium sp. adalah karbon yang digunakan untuk tumbuh dan melakukan pembelahan sel. Salah satu sumber karbon yang dapat digunakan utuk pertumbuhan Aurantiochytrium sp. adalah gliserol. Penentuan dosis gliserol sebagai sumber karbon alternatif dilakukan untuk efisiensi dalam budidayanya. Oleh karena itu, penelitian ini bertujuan untuk menentukan dosis optimal gliserol sebagai sumber karbon terhadap pertumbuhan, biomassa, kandungan nutrisi, dan karotenoid Aurantiochytrium sp.. Metode yang digunakan dalam penelitian ini adalah metode eksperimen menggunakan Rancangan Acak Lengkap (RAL) dengan 5 perlakuan dan 3 kali ulangan. Analisa data dilakukan secara statistik menggunakan analisa keragaman atau uji F (ANOVA) sesuai Rancangan Acak Lengkap (RAL). Parameter yang diuji adalah pertumbuhan, biomassa, kandungan nutrisi, dan karotenoid Aurantiochytrium sp.. Hasil penelitian menunjukkan bahwa dosis gliserol tidak memberi pengaruh signifikan terhadap laju pertumbuhan spesifik. Pengukuran pada parameter yang lain menunjukkan bahwa pemberian dosis gliserol 5%, memberikan hasil yang terbaik untuk biomassa, lipid, karbohidrat dan karotenoid yang dihasilkan. Hasil biomassa tertinggi yang dihasilkan sebesar 4,385g/L pada dosis gliserol 5%, sedangkan kandungan lipid, karbohidrat dan karotenoid menunjukkan hasil berturut-turut sebesar 33,62%, 55,11% dan 213,545μg/g. Hasil produksi protein menunjukkan hasil yang tidak berbeda jauh pada tiap perlakuan pemberian dosis gliserol. Berdasarkan hasil produksi biomassa, kandungan nutrisi dan karotenoid pada penelitian, menunjukkan penggunaan dosis gliserol 5% merupakan dosis yang terbaik untuk produksi biomasa, kandungan nutrisi dan karotenoid Aurantiochytrium sp..
English Abstract
Indonesia is one of the countries that has the largest mangrove forest area in the world with one of the microalgae that becomes a decomposer in the mangrove forest area, namely Aurantiochytium sp.. Aurantiochytrium sp. able to produce fatty acids that have many uses such as monounsaturated fatty acids (MUFA) and polyunsaturated fatty acids (Polyunsaturated Fatty Aicd / PUFA), especially docosahexaenoic acid (DHA). This microalgae has limiting factors in its growth, one of which is carbon which is used to grow and carry out cell division. One of the carbon sources that can be used is glycerol, because it is easy to obtain and the price is relatively cheap. Determination of the dosage of glycerol as an alternative carbon source is carried out in order to obtain efficient results in its cultivation. Therefore, this study aims to determine the optimal dose of glycerol as a carbon source for growth, biomass, nutrient content, and carotenoids of Aurantiochytrium sp. This research was conducted at the Laboratory of PT. Indoalgae Aquaculture on March – April 2023. The study was conducted to observe the effect of glycerol doses on growth, biomass, nutrient content, and carotenoid Aurantiochytrium sp.. The method used in this study was a Completely Randomized Design (CRD) method with 5 treatments and 3 repetitions. The treatment used was control (glucose 0.5%) and glycerol as a substitute for carbon sources at doses of 2%, 5%, 8% and 11%. Data analysis was performed statistically using analysis of variance or F test (ANOVA) according to Completely Randomized Design (CRD). The analysis was continued with the Duncan test to determine differences between treatments and the orthogonal polynomial test to determine the relationship between treatments with biomass, nutrient content, and carotenoid Aurantiochytrium sp.. The results showed that the dose of glycerol had no significant effect on the specifict growth rate. Measurements on other parameters showed that the dose of 5% glycerol gave the best result for the biomass, lipids, carbohydrates and carotenoids produced. The hingest yield of biomass produced was 4.385g/L at a dose of 5% glycerol, while the contents of lipids, carbohydrates dan carotenoids showed successive results of 33,62%, 55,11% and 213.545 μg/g. The results of proteins production showed results that were not much different in each treatment of glycerol dosing. The decrease in protein conten is affected by the high biomass produced. From the results of biomass production, nutrient content and carotenoids in the study, it was shown that the use of 5% glycerol dose was the best dose for biomass production, nutrient conten and carotenoid Aurantiochytrium sp..
