Abstract
Jeroan ikan tenggiri (Scomberomorus sp.) merupakan salah satu limbah yang dapat menyebabkan pencemaran lingkungan. Kapasitas yang banyak namun metode untuk menguranginya belum banyak dilakukan. Salah satu metode yang dapat dilakukan adalah dengan mengubahnya menjadi enzim protease. Tingginya kandungan protein dari usus ikan tenggiri (Scomberomorus sp.) dapat berpotensi menjadi enzim protease. Pembuatan enzim protease dari jeroan ikan khususnya usus ikan tenggiri (Scomberomorus sp.) dapat dilakukan dengan ekstraksi lalu diuji aktivitas enzim. Penelitian ini dilakukan dalam dua tahap yaitu penelitian pendahuluan dan penelitian utama. Penelitian pendahuluan dilakukan dengan membuat ekstrak kasar protease dari jeroan ikan tenggiri (Scomberomorus sp.) dan penelitian utama dilakukan dengan menguji aktivitas ekstrak kasar protease. Ekstrak dari usus ikan tenggiri (Scomberomorus sp.) disebut sebagai ekstrak kasar protease. Faktor-faktor yang mempengaruhi aktivitas enzim protease adalah pH optimum, suhu optimum dan konsentrasi substrat. Pengujian pH optimum, suhu optimum dan konsentrasi substrat dilakukan dalam enam level. pH optimum diuji terlebih dahulu sebagai awal Pengujian pH buffer yang dapat digunakan untuk pengujian suhu optimum dan konsentrasi substrat. Pengujian kedua yaitu suhu optimum dan dilanjutkan konsentrasi substrat. Konsentrasi substrat pada ekstrak kasar protease berpengaruh terhadap KM dan Vmax. Uji yang digunakan adalah uji One Way ANOVA dan dilanjutkan uji lanjutan Uji Duncan. Hasil dari pH optimum ekstrak kasar protease sebesar 12, suhu optimum 70 dan konsentrasi substrat dengan substrat casein bovine milk sebesar 2%. Hasil dari KM sebesar 0.753 mmol.L-1 dan hasil dari Vmax sebesar 4.47 mmol.L-1.menit-1. Berdasarkan penelitian perlakuan pH, suhu dan konsentrasi substrat berpengaruh terhadap aktivitas enzim. Apabila ekstrak kasar protease dari usus ikan tenggiri (Scomberomorus sp.) bekerja diatas titik optimum maka aktivitasnya akan menurun.
English Abstract
Viscera of spanish mackerel (Scomberomorus sp.) is one of the wastes that can cause fisheries waste. There is a lot of capacity, but there are not many methods to reduce it. One method that can be done is to convert it into a protease enzyme. Height content protein from the intestines of spanish mackerel (Scomberomorus sp.) can potentially become a protease enzyme. Manufacture of protease enzymes from fish innards, especially spanish mackerel intestine (Scomberomorus sp.) can be done by extraction and then tested for enzyme activity. This research was conducted in two stages, preliminary research and main research. Preliminary research was carried out by preparing protease crude extract from spanish mackerel (Scomberomorus sp.) and the main research was conducted by testing the protease activity of the crude extract. Extract from the intestines of spanish mackerel (Scomberomorus sp.) is referred to as crude protease extract. The factors that affect the activity of the protease enzyme are the optimum pH, optimum temperature and substrate concentration. Optimum pH, optimum temperature and substrate concentration tests were carried out in six levels. The optimum pH is tested first as a starting point for determining the pH buffer that can be used for testing the optimum temperature and substrate concentration. The second test is the optimum temperature and continues with the concentration of the substrate. Substrate concentration in the protease crude extract has an effect on KM and Vmax. Data from research will test with One Way ANOVA and advanced test with Duncan. The results of the optimum pH of the crude protease extract were 12, the optimum temperature was 70 and the concentration of substrate with substrate casein bovine milk by 2%. Results from Km is 0.753 mmol. L−1 and the result of Vmax is 4.47 mmol.L−11.minute−11.. Based on research on the treatment of pH, temperature and substrate concentrations affect enzyme activity. If the crude protease extract from the intestines of spanish mackerel (Scomberomorus sp.) works above the optimum point, it can make activity decrease.
