Abstract

Aurantiochytrium sp. dikategorikan sebagai salah satu jenis mikroalga yang hidupnya berada di perairan payau dan laut. Aurantiochytrium sp. bersifat heterotrof bersel tunggal yang mampu menghasilkan asam lemak tak jenuh dalam jumlah besar. Kandungan nutrisi Aurantiochytrium sp. dipengaruhi asupan nutrien dalam kultivasi. Yeast extract sebagi sumber N berguna memberikan suplai nutrisi organik yang diperlukan untuk pertumbuhan sel mikroalga. Konsentrasi nitrogen menyebabkan osmolaritas yang dapat mempengaruhi penyerapan nutrisi. Oleh karena itu, penelitian ini dilakukan untuk mengevaluasi pengaruh konsentrasi yeast extract terhadap pertumbuhan, biomassa, kandungan nutrisi, dan karotenoid pada Aurantiochytrium sp. Penelitian skripsi ini dilaksanakan di Laboratorium PT Indoalgae Akuakultur yang berlokasi di Pasuruan, Jawa Timur, pada bulan November-Desember 2022. Metode yang digunakan dalam penelitian ini yaitu metode eksperimen menggunakan Rancangan Acak Lengkap (RAL) dengan 4 perlakuan dan 3 ulangan. Perlakuan yeast extract yang digunakan adalah 5, 10, 15, dan 20 g/L. Parameter utama yang diuji adalah pertumbuhan, biomassa, lipid, karbohidrat, protein dan karotenoid. Parameter penunjang yang diukur dalam penelitian adalah pH, suhu, salinitas, dan serapan glukosa. Hasil penelitian ini menunjukkan bahwa yeast extract yang berbeda memiliki pengaruh nyata terhadap pertumbuhan, biomassa, kandungan nutrisi, dan karotenoid Aurantiochytrium sp. Yeast extract 15 g/L memiliki hasil yang optimal terhadap pertumbuhan dengan nilai kepadatan 2.254 serta laju pertumbuhan spesifik maksimum sebesar 0,134 sel/jam dengan doubling time 5,176 sel/jam, biomassa sebesar 9,412 g/L, karbohidrat sebesar 2,467 g/L, lipid sebesar 46,97%, protein sebesar 0,490 μg/gr dan karotenoid sebesar 9,48 μg/gr. Hasil pengukuran parameter penunjang menunjukkan pH berkisar antara 6 - 8, suhu berkisar antara 28-30oC, dan salinitas berkisar antara 65 - 107 ppt. Berdasarkan penelitian ini, dapat disimpulkan bahwa konsentrasi yeast extract berbeda memberikan pengaruh berbeda nyata terhadap pertumbuhan, biomassa, kandungan nutrisi, dan karotenoid Aurantiochytrium sp.. Konsentrasi yeast extract 15 g/L memiliki hasil yang optimal terhadap pertumbuhan, biomassa, karbohidrat, dan karotenoid Aurantiochytrium sp. Peneliti menyarankan untuk dilakukan penelitian lebih lanjut tentang konsentrasi yeast extract yang berbeda dan analisis kandungan nutrisi lainnya seperti PUFA, DHA, EPA, squalene.

English Abstract

Aurantiochytrium sp. categorized as a type of microalgae that live brackish and sea waters. Aurantiochytrium sp. are single-celled heterotrophs which capable of producing large amounts of unsaturated fatty acids. The nutritional content of Aurantiochytrium sp. influenced by nutrient intake during cultivation. Yeast extract as a source of N is useful in providing the supply of organic nutrients needed for the growth of microalgae cells. Nitrogen concentration causes osmolarity which can affect nutrient absorption. Therefore, this study conducted to evaluate the effect of yeast extract concentration on growth performance, biomass, nutrient content, and carotenoids content in Aurantiochytrium sp. This research was carried out at the PT Indoalgae Aquaculture Laboratory located in Pasuruan, East Java, in November-December 2022. The method used in this study was an experimental method using a Completely Randomized Design (CRD) with 4 treatments and 3 replications. The yeast extract treatments used were 5, 10, 15, and 20 g/L. The main parameters tested were growth, biomass, lipids, carbohydrates, proteins and carotenoids. Supporting parameters measured in the study were pH, temperature, salinity, and glucose uptake. The results of this study indicate that different yeast extracts have significant effects on growth, biomass, nutrient content, and carotenoids of Aurantiochytrium sp. Yeast extract 15 g/L has optimal results for growth with a density value of 2,254 and a maximum specific growth rate of 0.134 cells/hour with a doubling time of 5.176 cells/hour, biomass of 9.412 g/L, carbohydrates of 2.467 g/L, lipids of 46.97%, protein of 0.490 μg/gr and carotenoids of 9.48 μg/gr. The results of the supporting parameter measurements showed that the pH ranged from 6 - 8, the temperature ranged from 28 - 30oC, and the salinity ranged from 65 - 107 ppt. Based on this study, it can be concluded that different concentrations of yeast extract had significantly different effects on growth, biomass, nutrient content, and carotenoid Aurantiochytrium sp.. Concentration of yeast extract 15 g/L has optimal results on growth performance, biomass, carbohydrates, and carotenoid Aurantiochytrium sp. Researchers suggest further research on different concentrations of yeast extract and analysis of other nutritional content such as PUFA, DHA, EPA, squalene.
