Abstract
Likopen mengandung antioksidan yang banyak ditemukan dalam makanan seperti sayur dan buah terutama buah yang memiliki warna merah seperti tomat. Pemberian antioksidan terhadap embrio sebagai media maturasi dalam proses in vitro culture sudah terbukti dapat meningkatkan kualitas embrio. Akan tetapi, saat ini masih belum ada penelitian yang menjelaskan bahwa suplementasi likopen memiliki pengaruh terhadap perkembangan kualitas embrio ikan zebra. Tujuan dari penelitian ini yaitu, untuk menganalisis pengaruh suplementasi likopen terhadap perkembangan embrio ikan zebra dan menganalisis kondisi daya tetas, serta frekuensi detak jantung dari embrio ikan zebra yang dipapar heat stress. Prosedur penelitian meliputi kultur embrio, pemberian perlakuan, pengamatan parameter uji, dan analisis data. Embrio ikan zebra yang fertil dan sudah memasuki fase gastrula, dimasukkan ke dalam waterbath untuk dipapar heat stress dengan suhu mencapai 36oC. Dalam pemberian perlakuan likopen, telur ikan zebra dibagi menjadi dua kelompok yaitu kelompok yang dipapar heat stress dan tidak dipapar heat stress. Masing-masing kelompok diberikan tiga perlakuan yang berbeda yaitu perlakuan kontrol dengan 4 μL solution (P1), 2 μL solution + 2 μL likopen (P2), dan 4 μL likopen (P3) serta dilakukan ulangan sebanyak empat kali. Pengamatan embrio ikan zebra akan dilakukan menggunakan mikroskop inverted setiap tiga jam sekali hingga 48 hpf, kemudian dilanjutkan dengan pengamatan lima jam sekali hingga 72 hpf dan 96 hpf. Analisis data dilakukan dengan uji one way ANOVA untuk mengetahui pengaruh perlakuan menggunakan SPSS
English Abstract
Lycopene is one of the antioxidant that found in many vegetables and fruit, especially the fruit with red-colored such as tomato. Antioxidant given to embryo as a maturation media in the in vitro process has proven to improve the quality of the embryo itself. However, there is no research yet which explains how lycopene could affect the quality of zebrafish embryo. This research intends to analyze the influence of lycopene on the improvement of zebrafish embrio, to anaylze hatchability condition, and the frequency of heartrate of the zebrafish embryo under the heat stress. This research procedure includes embryo culture, treatment giving, observation to testing parameter, and data analysis. The fertile embryo of zebrafish that has entered gastrula stage, will be put into the waterbath to be exposed by heat- stress until the temperature reaches up to 36°C. In the giving of lycopene, zebrafish's eggs will divided into two groups, which are not exposed and exposed to heat-stress. Each group will be given three different treatment, which are control treatment with 4 μL solution (P1), 2 μL solution + 2uL lycopene (P2), dan 4 μL lycopene (P3) and there will be repetition for 4 times. The observation of zebrafish embryo will be performed using microscope inverted once every 3 hours until 48 hpf, then it will be continued with observation once every 5 hours until 72 hpf and 96 hpf. Data analysis was carried out by one way ANOVA test to determine the effect of treatment using SPSS.
