Abstract
Diabetes mellitus tipe 1 merupakan penyakit yang ditandai dengan meningkatnya kadar glukosa darah di atas normal. Streptozotocin (STZ) merupakan obat yang digunakan untuk pembuatan hewan coba model diabetes mellitus dengan cara mengganggu produksi insulin dan merusak sel b pankreas. Hal tersebut menyebabkan keadaan hiperglikemia yang kemudian akan meningkatkan Reactive Oxygen Species (ROS) sehingga terjadi peningkatan radikal bebas yang memicu kerusakan hepar dan ginjal. Penelitian ini bertujuan untuk menganalisis dan membandingkan perubahan histopatologi hepar dan ginjal akibat diabetes mellitus tipe 1 yang dibuat dengan cara induksi streptozotocin (STZ) melalui intraperitoneal metode single dose dan multiple dose. Hewan coba yang digunakan sebanyak 18 ekor tikus Sprague dawley jantan yang dibagi menjadi 3 kelompok; kontrol (K0), single dose 50mg/kg BB (P1) dan multiple dose 45mg/kg BB (P2). Pengukuran kadar glukosa darah dilakukan pada minggu ke-2 dan ke-4 setelah perlakuan menggunakan glukometer (Gluco. Dr). Organ hepar dan ginjal dikoleksi pada minggu ke-5 kemudian diamati perubahan histopatologi menggunakan pewarnaan Hematoxylin Eosine (HE) dengan mikroskop perbesaran 400x serta pengambilan gambaran histopatologi menggunakan aplikasi opti lab. Uji One Way Anova kadar glukosa darah pada minggu ke-2 dan minggu ke-4 berbeda signifikan (p<0,05). Uji Kruskal-Wallis histopatologi organ hepar dan ginjal berbeda signifikan (p<0,05). Hasil penelitian menunjukan induksi streptozotocin (STZ) metode multiple dose mampu menyebabkan kerusakan di hepar dan ginjal yang lebih parah daripada metode single dose. Kerusakan hepar ditandai dengan degenerasi dan nekrosis dengan rerata derajat kerusakan P2=2,30±0,24. Kerusakan pada ginjal ditandai dengan glomerulosklerosis dan nekrosis tubulus dengan rerata derajat kerusakan P2=1,93±0,48.
English Abstract
Type 1 diabetes mellitus is a disease characterized by increased blood glucose levels above normal. Streptozotocin (STZ) is a drug used to manufacture experimental animal models of diabetes mellitus by interfering with insulin production and damaging pancreatic β cells. This causes a state of hyperglycemia which will then increase Reactive Oxygen Species (ROS) resulting in an increase in free radicals which trigger liver and kidney damage. This study aims to analyze and compare the histopathological changes of the liver and kidneys due to type 1 diabetes mellitus produced by induction of streptozotocin (STZ) via intraperitoneal single dose and multiple dose methods. The experimental animals used were 18 male Sprague Dawley rats which were divided into 3 groups; control (K0), single dose 50mg/kg BW (P1) and multiple dose 45mg/kg BW (P2). Blood glucose levels were measured on the 2nd and 4th week after treatment using a glucometer (Gluco. Dr). The liver and kidney organs were collected in the 5th week and then observed for histopathological changes using Hematoxylin Eosine (HE) staining with a 400x magnification microscope and taking histopathological images using the opti lab application. One Way Anova test blood glucose levels at week 2 and week 4 were significantly different (p<0.05). The Kruskal-Wallis test for liver and kidney histopathology was significantly different (p<0.05). The results showed that the multiple dose induction of streptozotocin (STZ) could cause more severe liver and kidney damage than the single dose method. Liver damage is characterized by degeneration and necrosis with an average degree of damage P2 = 2.30±0.24. Damage to the kidney is characterized by glomerulosclerosis and necrosis tubules with an average degree of damage P2 = 1.93±0.48
