Abstract

Organofosfat dapat meninggalkan residu pada tanaman yang menyebabkan efek samping pada hewan pememakan tanaman tersebut. Organofosfat menyebabkan hilangnya fungsi enzim AchE dan menyebabkan akumulasi asetilkolin yang berefek pada reseptor saraf, sehingga memicu gangguan hipotalamus dan mengganggu sekresi hormon reproduksi. Jintan hitam memiliki kandungan thymoquinone sebagai antioksidan sehingga mengurangi radikal bebas. Tujuan dari penelitian untuk mengetahui efek terapi jintan hitam terhadap jumlah folikel deGraaf dan folikel atresia pada mencit yang terpapar oleh organofosfat. Mencit dibagi 5 kelompok perlakuan yaitu kontrol negatif tidak diberikan perlakuan, kontrol positif diinduksi diazinon 40 mg/kgBB, dan perlakuan I, II, III dengan masing-masing diinduksi diazinon 40 mg/kgBB 8 minggu ditambah pemberian ekstrak jintan hitam dengan dosis yaitu 200 mg/kgBB, 400 mg/kgBB, dan 600 mg/kgBB secara bertahap dimulai minggu ke 5 hingga minggu ke 8 satu kali sehari. Koleksi ovarium dilakukan untuk pengamatan histopatologi jumlah folikel deGraaf dan atresia. Hasil menunjukan pada folikel deGraaf kelompok K+ berjumlah 2.8, peningkatan kelompok K+ sebanyak 250%, peningkatan kelompok P1 sebanyak 25%, peningkatan kelompok P2 sebanyak 100%, dan peningkatan kelompok P3 sebanyak 50%, Sementara untuk folikel atresia Kelompok K+ sebanyak 19.6, penurunan kelompok K- sebanyak 48%, penurunan kelompok P1 sebanyak 22%, penurunan kelompok P2 sebanyak 43%, dan penurunan kelompok P3 sebanyak 14%. Data dianalisis menggunakan SPSS dengan analisis ANOVA serta Uji Turkey taraf kepercayaan 95%. Kesimpulan penelitian ini yaitu dosis ekstrak jintan hitam 400 mg/kgBB menjadi dosis paling baik dalam meningkatkan jumlah folikel deGraaf dan menurunkan jumlah folikel Atresia pada mencit yang diinduksi diazinon, meskipun peningkatannya tidak berbeda nyata.

English Abstract

Food product often come from plants and can’t be seperated with the use of pestiside. One type of pestiside often used for pest control in indonesia is oganophospate. Organophospate can leave residues in plants and transfered to animal if they eat it and cause side effect, one of the side effect is disturbing the work of acetilcolinesterase enzyme and will cause the accumulation of asetilcoline as well as increasing free radicals. This side effect will cause damage in hypotalamus and will distrupt secretion of hormone, especially reproductive hormone. The administration of black cumin that have thymoqionone as the main content can reduce the excessive of free radicals because it have antioxidant property. Purpose of this study was to find out the effect of black cumin therapy on the number of Graafian follicle and atretic follicle of female mice that had been induced by organofosfat. Female mice were divided into 5 groups wich consist f negatif control, positive control wich has been induced with diazinon 40 mg/kg BB for 8 weeks, and the treatment groups I, II, II eAch induced with the same dosage as positive control group and adding administration of black cumin extract with dose 200 mg/kg BB, 400 mg/kg Bg, and 600 mg/kg BB respectively starting in week 5 until week 8 using gastric sonde everyday. At the end of the treatment ovarium collection was peroformed for histopathological observation to count the number of Graafian follicle and atretic follicle. The result showed for graafian follicle K- group have 2.8 average follicle, K+ group have 0.8 average follicle, P1 have 1 average follicle, P2 group have 1.6 average follicle, and P3 group have 1.2 average follicle, and for atretic follicle K- group have 9.6 average follicle, K+ group have 19.6 average follicle, P1 group have 15.2 average follicle, P2 group have 11 average follicle, and P3 group have 16.8 average follicle. The data were analyzed using SPSS for windows with ANOVA variety analysis and Turkey Test with 95% confidence level. Conclusion from this research is that 400 mg/kg BB dose of cumin extract have the most effect for curing the diazinon effect, it showed from higher graafian follicle and lower atretic follicle
