Abstract
Kolitis merupakan inflamasi kolon yang muncul sebagai gejala klinis penyakit saluran penceraan ayam. Kolitis dapat mengganggu fungsi usus besar dan menimbulkan respon limpa terhadap peradangan yang terjadi. Hewan model kolitis dibuat dengan induksi Dextran Sodium Sulfate (DSS). Penelitian ini bertujuan untuk mengetahui parameter histomorfometri persentase luas pulpa putih dan pulpa merah beserta gambaran histopatologi limpa ayam model kolitis yang diinduksi DSS. Digunakan ayam broiler strain Cobb yang dibagi menjadi empat kelompok dengan lima ekor ayam pada masing-masing kelompok. Ayam dipelihara selama 14 hari, kemudian diinduksi DSS peroral melalui air minum 1 x 24 jam selama enam hari. Dosis DSS yang diberikan adalah 0,75% untuk kelompok P1, 1,5% untuk kelompok P2, 2,25% untuk kelompok P3, dan tidak diberikan DSS untuk kelompok kontrol negatif. Pada hari ke-21 dilakukan eutanasi pada ayam dan diambil organ limpa untuk pengukuran persentase luas pulpa merah dan pulpa putih serta pengamatan histopatologi limpa. Analisa data kuantitatif persentase luas pulpa dilakukan dengan uji One Way ANOVA dilanjutkan dengan uji Duncan. Gambaran histopatologi limpa disajikan secara deskriptif kualitatif. Pengamatan gejala klinis dan nekropsi menunjukkan terdapat perubahan konsistensi feses, penurunan pertambahan berat badan, serta adanya hemoragi pada sekum. Histomorfometri pulpa putih menunjukkan peningkatan signifikan pada kelompok DSS 2,25% dan pulpa merah menunjukkan penurunan signifikan pada kelompok DSS 2,25%. Hasil pengamatan histopatologi menunjukkan adanya kongesti pada pulpa merah pada kelompok DSS 2,25%. Kesimpulan dari penelitian ini adalah persentase luas pulpa putih dan pulpa merah berbeda nyata pada pada kelompok DSS 2,25% dan perubahan histopatologi diamati pada kelompok DSS 2,25% berupa kongesti.
English Abstract
Colitis is an inflammation of the colon. Colitis can interfere with the function of the large intestine and cause a splenic response to the inflammation that occurs. Colitis animal models were made by induction of Dextran Sodium Sulfate (DSS). This study aims to determine the histomorphometric parameters of the percentage area of white pulp and red pulp along with histopathological features of the spleen of chicken colitis induced by DSS. Cobb strain broiler chickens were used which were divided into four groups with five chickens in each group. Chickens were reared for 14 days, then DSS was induced orally through drinking water 1 x 24 hours for six days. The dose of DSS given was 0.75% for the P1 group, 1.5% for the P2 group, 2.25% for the P3 group, and no DSS was given for the negative control group. On the 21st day, the chickens were euthanized and the spleen was taken to measure the percentage of red and white pulp areas as well as histopathological observations of the spleen. Quantitative data analysis on the percentage of pulp area was carried out using the One Way ANOVA test followed by Duncan's test. Histopathological picture of the spleen is presented in a qualitative descriptive manner. Observation of clinical symptoms and necropsy showed changes in stool consistency, decreased weight gain, and the presence of hemorrhage in the cecum. The histomorphometry of the white pulp showed a significant increase in the 2.25% DSS group and the red pulp showed a significant decrease in the 2.25% DSS group. The results of histopathological observations showed congestion in the red pulp in the 2.25% DSS group. The conclusion of this study was that the percentage area of white pulp and red pulp was significantly different in the 2.25% DSS group and the histopathological changes observed in the 2.25% DSS group were congestion
