Abstract
Thraustochytrids adalah sekelompok protista heterotrofik laut yang termasuk dalam kelas Labyrinthulomycetes. Thraustochytrids adalah organisme bersel tunggal yang umumnya ditemukan di lingkungan laut, terutama di sedimen dan air laut. Thraustochytrids dikenal karena kemampuannya menghasilkan asam lemak dalam jumlah tinggi, terutama asam docosahexaenoic (DHA) dan asam lemak omega-3. Karena produksi DHA mereka yang signifikan, Thraustochytrids mendapat perhatian di bidang bioteknologi dan akuakultur. Nitrogen adalah nutrisi penting untuk pertumbuhan dan perkembangan Thraustochytrids, dan sumber nitrogen organik telah terbukti secara efektif meningkatkan produksi biomassa dan senyawa bioaktif. Sumber nitrogen anorganik yang berbeda telah digunakan untuk penanaman Thraustochytrids. Sumber nitrogen ini mengandung berbagai asam amino, peptida, dan vitamin, yang tidak hanya menyediakan nitrogen tetapi juga nutrisi penting lainnya yang dibutuhkan untuk pertumbuhan dan metabolisme Thraustochytrids yang optimal. Namun dibutuhkan lebih banyak variasi sumber nitrogen anorganik pada penelitian kali ini. Sumber nitrogen anorganik yang digunakan yaitu amonium sulfat, amonium nitrat, potassium nitrat dan sodium nitrat. Oleh karena itu, penelitian ini dilakukan untuk menguji pengaruh pemberian sumber nitrogen anorganik pada media terhadap pertumbuhan, biomassa, kandungan nutrisi, dan karotenoid pada Thraustochytrids. Metode yang digunakan dalam penelitian ini yaitu metode eksperimen menggunakan Rancangan Acak Lengkap (RAL) dengan 4 perlakuan dan 3 ulangan. Perlakuan pemberian sumber nitrogen anorganik yang digunakan adalah amonium sulfat, amonium nitrat, potassium nitrat dan sodium nitrat. Parameter utama yang diuji adalah pertumbuhan, biomassa, lipid, karbohidrat, protein dan karotenoid. Parameter penunjang yang diukur dalam penelitian adalah pH, suhu, salinitas. Hasil penelitian ini menunjukkan bahwa pemberian sumber nitrogen anorganik memiliki pengaruh nyata terhadap pertumbuhan, biomassa, dan kandungan nutrisi Thraustochytrids. Sumber nitrogen anorganik Potassium nitrat memiliki hasil yang optimal terhadap biomassa sebesar 2,853 g/L, karbohidrat sebesar 22,90%, karotenoid sebesar 0,1312 μg/gr, lipid sebesar 10,53%, dan protein sebesar 0,06 μg/gr. Hasil pengukuran parameter penunjang menunjukkan pH berkisar antara 2 hingga 8, suhu berkisar antara 27-32oC, dan salinitas berkisar antara 19-25 ppt. Berdasarkan penelitian ini, dapat disimpulkan bahwa pemberian sumber nitrogen anorganik yang berbeda memberikan pengaruh berbeda nyata terhadap pertumbuhan, biomassa, kandungan nutrisi, dan karotenoid Thraustochytrids. sumber nitrogen Potassium nitrat memiliki hasil yang optimal terhadap biomassa, lipid, karotenoid dan protein Thraustochytrids. Sedangkan amonium nitrat dan amonium sulfat menghasilkan lipid tertinggi dan sodium nitrat menghasilkan nilai bimassa tertinggi.
English Abstract
Thraustochytrids are a group of marine heterotrophic protists belonging to the class Labyrinthulomycetes. Thraustochytrids are single-celled organisms commonly found in marine environments, especially in sediments and seawater. Thraustochytrids are known for their ability to produce high amounts of fatty acids, especially docosahexaenoic acid (DHA) and omega-3 fatty acids. Due to their significant production of DHA, Thraustochytrids have received attention in biotechnology and aquaculture. Nitrogen is an essential nutrient for the growth and development of Thraustochytrids, and organic nitrogen sources have been shown to effectively increase the production of biomass and bioactive compounds. Different inorganic nitrogen sources have been used for growing Thraustochytrids. This nitrogen source contains a variety of amino acids, peptides and vitamins, which provide not only nitrogen but also other essential nutrients required for optimal Thraustochytrids growth and metabolism. However, more variations of inorganic nitrogen sources are needed in this study. The inorganic nitrogen sources used were ammonium sulfate, ammonium nitrate, potassium nitrate and sodium nitrate. Therefore, this study was conducted to evaluate the provision of inorganic nitrogen sources in the media on growth, biomass, nutrient content, and carotenoids in Thraustochytrids. The method used in this study was the experimental method using a completely randomized design (CRD) with 4 treatments and 3 replications. Treatment of giving inorganic nitrogen sources used are ammonium sulfate, ammonium nitrate, potassium nitrate and sodium nitrate. The main parameters tested were growth, biomass, lipids, carbohydrates, proteins and carotenoids. Supporting parameters measured in the study were pH, temperature, salinity. The results of this study indicate that the provision of inorganic nitrogen sources has a significant effect on growth, biomass, and nutritional content of Thraustochytrids. Inorganic nitrogen sources Potassium nitrate had optimal results for biomass of 2.853 g/L, carbohydrates of 22.90%, carotenoids of 0.1312 μg/gr, lipids of 10.53%, and proteins of 0.06 μg/gr. The results of the supporting parameter measurements showed that the pH ranged from 2 to 8, the temperature ranged from 27-32oC, and the salinity ranged from 19-25 ppt. Based on this study, it can be concluded that the provision of different inorganic nitrogen sources had significantly different effects on the growth, biomass, nutrient content, and carotenoids of Thraustochytrids. nitrogen source Potassium nitrate has optimal results on biomass, lipids, carotenoids and proteins of Thraustochytrids. While ammonium nitrate and ammonium sulfate produced the highest lipids and sodium nitrate produced the highest mass value.
