Abstract
kan zebra dapat digunakan banyak penelitian seperti pengembangan genetik, gangguan metabolisme, toksikologi dan perilaku. Heat stress merupakan keadaan dimana tubuh menyerap panas diatas kemampuannya akan tetapi tubuh hewan tidak mampu untuk mengeluarkan panas dari tubuhnya. Likopen merupakan salah satu antioksidan yang dapat membantu melawan radikal bebas. Pemberian likopen diketahui dapat memberi efek yang menguntungkan, namun penelitian tentang variasi suhu heat stress pada proses in vitro culture terhadap denyut jantung, derajat penetasan dan survival rate pada ikan zebra yang disuplementasi likopen belum pernah dilakukan. Tujuan penelitian ini untuk mengetahui pengaruh likopen terhadap denyut jantung, derajat penetasan dan survival rate pada embrio ikan zebra yang dipapar heat stress. Penelitian dimulai dari telur ikan zebra hasil pemijahan diseleksi untuk memisahkan telur fertil dan non fertil. Embrio ikan zebra yang fertil dan sudah memasuki fase gastrula, dibagi menjadi 4 kelompok suhu yaitu non heat stress (NHS), heat stress (HS) dengan suhu 30oC, 35oC dan 40oC. Masing-masing kelompok diberikan tiga perlakuan yang berbeda yaitu perlakuan kontrol dengan 2 μL solution (P1), 1 μL solution + 1 μL likopen (P2), dan 2 μL likopen (P3) serta dilakukan ulangan sebanyak tiga kali. Pengamatan dilakukan menggunakan mikroskop inverted setiap tiga jam sekali hingga 48 hpf, kemudian dilanjutkan dengan pengamatan lima jam sekali hingga 96 hpf. Hasil dari penelitian yang didapatkan, antioksidan yang terkandung dalam likopen berpengaruh positif terhadap denyut jantung, derajat penetasan dan survival rate yang ditunjukkan signifikansi terhadap denyut jantung (bpm) HS35P3 (175,3±2,08; P=0,009), presentase derajat penetasan HS35P3 (90±0%; P=0,000) serta presentase survival rate HS35P3 (90±0%; P=0,000).
English Abstract
Zebrafish can used in many researches such as genetic development, metabolic disorders, toxicology and behavior. Heat stress is condition the body absorbs heat beyond its capacity but the animal's body unable to remove heat from its body. Lycopene an antioxidant can help fight free radicals. Giving lycopene is known to have beneficial effect, but research on heat stress temperature variations in the IVC process on heart rate, hatching rate and survival rate in zebrafish supplemented with lycopene has never been done. The purpose of this study was to determine the effect of lycopene on heart rate, hatching rate and survival rate in zebrafish embryos exposed to heat stress. Research started from zebrafish eggs spawning were selected to separate fertile and non-fertile. Zebrafish embryos were fertile and had entered the gastrula phase were divided into 4 temperature groups namely non heat stress (NHS), heat stress (HS) with temperatures of 30oC, 35oC and 40oC. Each group was given three different treatments, namely control treatment with 2 μL solution (P1), 1 μL solution + 1 μL lycopene (P2) and 2 μL lycopene (P3) and repeated three times. Observations made using inverted microscope every three hours up to 48 hpf, then followed by observations every five hours up to 96 hpf. Results of study found that antioxidants contained in lycopene had a positive effect on heart rate, hatching rate and survival rate showed significance on heart rate (bpm) HS35P3 (175.3±2.08; P=0.009), percentage of hatching rate HS35P3 (90±0%; P=0.000) and survival rate HS35P3 (90±0%; P=0.000
