Abstract
Proses kristalisasi spermatozoa saat pembekuan semen menyebabkan menurunya tingkat fertilitas selain itu juga dapat menyebabkan peningkatan kematian sel spermatozoa dan abnormalitas morfologi spermatozoa. Penambahan Oviduct Spesific Glycoprotein (OVGP) pada penelitian ini diharapkan dapat menjadi krioprotektan eksogenous, OVGP pada penelitian ini ditambahkan pada pengencer semen (AndroMed® dan Kuning Telur Sitrat). Tujuan dari penelitian ini adalah untuk mengetahui pengaruh penambahan beberapa media pengencer (AndroMed® dan Kuning Telur Sitrat) yang ditambahkan OVGP pada proses pembekuan semen dalam hasil tingkat abnormalitas morfologi sperma dan tingkat HSP70. Penelitian ini menggunakan analisa Rancangan Acak Lengkap yang dibagi menjadi empat perlakuan yaitu kelompok P1 (pengencer KTS tanpa penambahan OVGP); kelompok P2 (pengencer KTS ditambahkan OVGP sebanyak 5 μl/mL) ; kelompok P3 (pengencer AndroMed® tanpa penambahan OVGP) ; kelompok P4 (pengencer AndroMed® ditambahkan OVGP sebanyak 5 μl/mL). Ekspresi HSP70 dianalisa dengan menggunakan metode imunositokimia sedangkan untuk morfologi spermatozoa menggunakan pewarnaan eosin negrosin. Hasil yang didapatkan kemudian dianalisa dengan menggunakan One Way ANOVA (p< 0.05) apabila terdapat perbedaan yg nyata maka akan diteruskan dengan uji Tukey sedangkan analisa data morfologi diamati dianalisa secara deskriptif. Dari penelitian ini, hasil ekspresi HSP70 pada perlakuan P1 sebesar 2,64±1,11, perlakuan P2 0,57±0,22, perlakuan P3 1,84±0,40, dan perlakuan P4 0,58±0,46. Data yang didapat pada jumlah abnormalitas spermatozoa post thawing adalah P1 sebesar 38, perlakuan P2 21, perlakuan P3 29, dan perlakuan P4 15. Berdasarkan hasil tersebut, dapat disimpulkan bahwa penambahan OVGP kedalam bahan pengencer semenbeku dapat menurunkan ekspresi HSP70 pada spermatozoa serta terdapat menurunkan presentase abnormalitas spermatozoa.
English Abstract
The process of crystallization of spermatozoa during freezing of semen causes a decrease in the fertility rate but it can also cause an increase in spermatozoa cell death and morphological abnormalities of spermatozoa. The addition of Oviduct Specific Glycoprotein (OVGP) in this study is expected to be an exogenous cryoprotectant, whereas OVGP in this study was added to cement diluents (AndroMed® and Egg Yolk Citrate). The purpose of this study was to determine the effect of the addition of several diluent media (AndroMed® and Egg Yolk Citrate) which was added with OVGP to the semen freezing process on the results of sperm morphology abnormalities and HSP70 levels. This study used a completely randomized design analysis which was divided into four treatments, namely group P1 (KTS diluent without addition of OVGP); group P2 (KTS diluent added with 5 μl /mL of OVGP); group P3 (AndroMed® diluent without the addition of OVGP); group P4 (AndroMed® diluent was added with 5 μl/mL OVGP). Expression of HSP70 was analyzed using immunocytochemistry method while for spermatozoa morphology using eosin negrosin staining. The results obtained were then analyzed using One Way ANOVA (p < 0.05) if there was a significant difference, it would be continued with the Tukey test, while the morphological data analysis was observed to be analyzed descriptively. From this study, the results of HSP70 expression in the P1 treatment were 2.64 ± 1.11, the P2 treatment was 0.57 ± 0.22, the P3 treatment was 1.84 ± 0.40, and the P4 treatment was 0.58 ± 0. 46. The data obtained on the total of post-thawing spermatozoa abnormalities were P1 of 38, P2 treatment of 21, P3 treatment of 29, and P4 treatment of 15. Based on these results, it can be concluded that the addition of OVGP to frozen semen diluent can decrease HSP70 expression in spermatozoa and there is a decrease in the percentage of abnormal spermatozo
