Abstract
Budidaya ikan tidak akan lepas dari permasalahan penyakit baik secara infeksi maupun non infeksi. Pernyakit bakterial yang sering menyerang udang dan ikan laut adalah bakteri Vibrio parahaemolyticus merupakan patogen utama pada ikan air laut maupun udang. Bakteri ini menyebabkan timbulnya penyakit Acute Hepatopancreatic Necrosis Disease (AHPND) atau sering dikenal sebagai Early Mortality Syndrome (EMS) pada udang. Gejala klinis yang muncul pada ikan yang terinfeksi bakteri ini yaitu pembusukan pada sirip, borok pada bagian tubuh, dan mulut memerah. Serangan dari bakteri ini mampu mengakibatkan kematian ikan dan udang dalam waktu yang singkat dan dalam jumlah yang besar. Salah satu alternative yang digunakan untuk mengatasi permasalahan ini untuk menghindari penggunaan yang meluas dari antibiotik adalah menggunakan antibiotic alami seperti bunga telang. Bunga telang mengandung senyawa antibakteri yaitu flavonoid, alkaloid dan tanin. Penelitian ini dilaksanakan di Laboratorium Mikrobiologi, Laboratorium Sentra Ilmu Hayati, Universitas Brawijaya, Malang. Penelitian ini dilaksanakan pada bulan Januari-Februari 2022. Tujuan penelitian ini adalah untuk mengetahui daya hambat ekstrak kasar bunga telang terhadap bakteri V. parahaemolyticus secara in-vitro. Metode penelitian ini adalah metode eksperimental dengan rancangan percobaan berupa Rancangan Acak Lengkap (RAL). Rancangan penelitian dilakukan dengan 5 perlakuan, 3 ulangan dan 2 kontrol. Dosis ekstrak bunga telang yang digunakan yaitu A (25 ppm), B (50 ppm), C (75 ppm), D (100 ppm) dan E (125 ppm). Control positif menggunakan antibiotik kloramfenikol 30 ppm dan control negative tanpa perlakuan. Hasil penelitian menunjukkan bahwa adanya zona bening pada sekitar kertas cakram dan menghasilkan ukuran diameter zona bening tertinggi pada perlakuan E (125 ppm) dengan rerata zona hambat sebesar 11,37 ± 1,00 mm. Dosis yang menghasilkan ukuran diameter zona bening terendah didapatkan pada perlakuan A (25 ppm) dengan rerata zona hambat sebesar 8,3 ± 0,52 mm. Hubungan antara dosis ekstrak bunga telang dengan ukuran zona hambat menghasilkan pola linier dengan persamaan y = 0,0219x + 6,6841 dan koefisien R2 = 0,8289. Koefisien R2 menunjukkan daya hambat bakteri ekstrak bunga telag terhadap bakteri V. parahaemolyticus sebesar 82% Kesimpulan hasil penelitian bahwa ekstrak kasar bunga telang (C. ternatea) mampu membunuh bakteri V. parahaemolyticus secara in-vitro. Hal tersebut dikarenakan senyawa yang terkandung didalam bunga telang yang bersifat sebagai antibakteri. Diameter zona hambat ekstrak bunga telang. Diameter zona hambat ekstrak bunga telang berkisar 8,3-11,37 mm. Hasil ini termasuk dalam kategori sedang-kuat. Ekstrak kasar bunga telang telah dibuktikan bakteriosidal atau dikategorikan dapat membunuh bakteri V. parahaemolyticus
English Abstract
Aquaculture cannot be separated from disease problems, both infectious and non-infectious. The bacterial disease that often attacks shrimp and marine fish is the bacterium Vibrio parahaemolyticus which is the main pathogen in seawater fish and shrimp. These bacteria cause Acute Hepatopancreatic Necrosis Disease (AHPND) or often known as Early Mortality Syndrome (EMS) in shrimp. Clinical symptoms that appear in fish infected with this bacterium are rotting of the fins, ulcers on body parts, and reddened mouth. Attacks from these bacteria can cause the death of fish and shrimp in a short time and in large numbers. One alternative that is used to overcome this problem to avoid the widespread use of antibiotics is to use natural antibiotics such as telang flower. Telang flower contains antibacterial compounds, namely flavonoids, alkaloids and tannins. This research was conducted at the Microbiology Laboratory, Laboratory of the Center for Life Sciences, Brawijaya University, Malang. This research was conducted in January-February 2022. The purpose of this study was to determine the V. parahaemolyticus inhibition in vitro. This research method is an experimental method with an experimental design in the form of a completely randomized design (CRD). The research design was carried out with 5 treatments, 3 replications and 2 controls. The doses of telang flower extract used were A (25 ppm), B (50 ppm), C (75 ppm), D (100 ppm) and E (125 ppm). Positive control used 30 ppm chloramphenicol antibiotic and negative control without treatment. The results showed that there was a clear zone around the paper disc and the highest clear zone diameter was in treatment E (125 ppm) with an average inhibition zone of 11.37 ± 1.00 mm. The dose that produced the lowest clear zone diameter was found in treatment A (25 ppm) with an average inhibition zone of 8.3 ± 0.52 mm. The relationship between the dose of telang flower extract and the size of the inhibition zone resulted in a linear pattern with the equation y = 0.0219x + 6.6841 and the coefficient R2. = 0.8289The coefficient of R2 indicates the inhibition of the telang flower extract bacteria against V. parahaemolyticus by 82% The conclusion of the study was that the crude telang flower extract (C. ternatea) was able to kill V. parahaemolyticus in vitro. This is because the compounds contained in the pea flower are antibacterial. The diameter of the inhibition zone of the pea flower extract. The diameter of the inhibition zone of telang flower extract ranged from 8.3 to 11.37 mm. This result is included in the medium-strong category. The crude extract of telang flower has been proven to be bactericidal or categorized as being able to kill V. parahaemolyticus bacteria.
