Abstract
Ikan Koi (Cyprinus carpio) merupakan jenis ikan hias air tawar mempunyai nilai ekonomis yang penting. Perdagangan ikan hias dunia mencapai 1.600 jenis dan sekitar 46% (750 jenis) berasal dari air tawar, namun permasalahan yang sering terjadi adalah munculnya penyakit yang menyerang benih, salah satu penyakit yang menyerang Ikan Koi (Cyprinus carpio) yaitu Myxobolus sp. yang disebabkan oleh parasit Myxobolus sp. Salah satu alternatif yang dilakukan oleh para petani Ikan Koi (Cyprinus carpio) untuk mencegah terjangkitnya infeksi Myxobolus sp. yaitu dengan pemberian obat secara alami maupun pemberian probiotik. Berdasarkan penjelasan diatas, hal tersebut yang melatarbelakangi penulis untuk melakukan penelitian mengenai Efek Pemberian Probiotik Terhadap Histopatologi Dan Sistem Imun Ikan Koi (Cyprinus carpio) Yang Terinfeksi Parasit Myxobolus sp. Penelitian ini bertujuan untuk mengetahui dan menganalisis proses probiotik sebagai imunostimulan pada Ikan Koi (Cyprinus carpio) dalam mencegah parasit Myxobolus sp. serta mengetahui respon imun IL-6 pada Ikan Koi (Cyprinus carpio) yang diberi perlakuan probiotik. Metode yang digunakan dalam penelitian ini adalah metode eksperimental. Metode analisis yang digunakan antara lain uji histopatologi dan IHK (Imunohistokimia). Pengamatan pada penelitian ini dilakukan antara lain yaitu perlakuan ikan kontrol negatif yang merupakan ikan sehat, kontrol positif ikan yang terinfeksi Myxobolus sp. tanpa perlakuan pemberian probiotik, Ikan Koi (Cyprinus carpio) terinfeksi Myxobolus sp. (P1, P2, dan P3), dan Ikan Koi (Cyprinus carpio) sehat (P4, P5 dan P6) dengan pemberian dosis masing-masing A=0,60 ml, B=1,2 ml, dan C=1,8 ml. Aktifitas gen IL-6 diamati pada organ usus ikan uji pada setiap ikan perlakuan. Hasil uji histopatologi ditemukan jenis kerusakan edema, hiperplasia dan nekrosis. Masing-masing perlakuan didapatkan nilai paling rendah yaitu edema pada perlakuan P6 (3%) dan tertinggi P3 (27%) dan hasil terbaik pada perlakuan P1 (13%). Pada kerusakan hiperplasia hasil paling rendah pada perlakuan P4,P5,P6 (3%) dan tertinggi P2,P3 (27%) dan hasil terbaik pada perlakuan P1 (17%). Pada kerusakan nekrosis hasil paling rendah pada perlakuan P5,P6 (3%) dan tertinggi P2,P3 (23%) dan hasil terbaik pada perlakuan P1 (13%). Sedangkan untuk hasil imunohistokimia (IHK) diperoleh nilai ekspresi IL-6 paling rendah pada perlakuan P4 ulangan pertama (17,7%) dan tertinggi P3 ulangan ketiga (76,4%) dan hasil terbaik pada perlakuan P1 ulangan pertama (60,2%). Berdasarkan analisa imunohistokimia, pemberian probiotik berpengaruh terhadap ekspesi IL-6, dan perlakuan P1 (pemberian probiotik dengan dosis 0,60 ml) merupakan perlakuan paling baik, melalui hasil pengamatan melalui aplikasi ImageJ dengan plugin immunoratio dimana pada ikan yang terinfeksi Myxobolus sp. memiliki ekspresi sitokin yang tinggi dan pada ikan yang terinfeksi Myxobolus sp. dengan pemberian probiotik memiliki respon IL-6 yang lebih rendah, sehingga probiotik mempunyai peranan penting dalam pengaturan mekanisme sistem kekebalan tubuh Ikan Koi (Cyprinus carpio) yang terinfeksi Myxobolus sp.
English Abstract
Koi fish (Cyprinus carpio) is a type of freshwater ornamental fish that has important economic value. The world trade in ornamental fish reaches 1,600 species and about 46% (750 species) comes from fresh water, but the problem that often occurs is the emergence of diseases that attack seeds, one of the diseases that attack Koi Fish (Cyprinus carpio) is Myxobolus sp. caused by the parasite Myxobolus sp. One of the alternatives made by Koi Fish (Cyprinus carpio) farmers to prevent infection with Myxobolus sp. namely by giving natural drugs or giving probiotics. Based on the explanation above, this is the background for the author to conduct research on the Effects of Giving Probiotics on Histopathology and Immune System of Koi Fish (Cyprinus carpio) Infected with Myxobolus sp. This study aims to determine and analyze the probiotic process as an immunostimulant in Koi Fish (Cyprinus carpio) in preventing the parasite Myxobolus sp. and to determine the immune response of IL-6 in Koi Fish (Cyprinus carpio) treated with probiotics. The method used in this research is the experimental method. The analytical methods used include histopathological tests and CPI (Imunohistochemistry). Observations in this study were carried out, among others, namely the treatment of negative control fish which were healthy fish, positive control fish infected with Myxobolus sp. without treatment with probiotics, Koi Fish (Cyprinus carpio) infected with Myxobolus sp. (P1, P2, and P3), and healthy Koi (Cyprinus carpio) (P4, P5 and P6) with doses of A=0.60 ml, B=1.2 ml, and C=1.8 respectively. ml. The activity of the IL-6 gene was observed in the intestinal organs of the test fish in each treated fish. Histopathological test results found types of damage edema, hyperplasia and necrosis. Each treatment got the lowest score, namely edema in treatment P6 (3%) and the highest in P3 (27%) and the best results in treatment P1 (13%). In hyperplastic damage, the lowest results were in treatment P4,P5,P6 (3%) and the highest was P2,P3 (27%) and the best results were in treatment P1 (17%). In the necrotizing damage, the lowest yield was in treatment P5,P6 (3%) and the highest was P2,P3 (23%) and the best result was in treatment P1 (13%). Meanwhile, for the results of immunohistochemistry (IHK), the lowest IL-6 expression value was in the first P4 treatment (17.7%) and the highest was P3 in the third replicate (76.4%) and the best results were in the first P1 treatment (60.2%). Based on immunohistochemical analysis, the administration of probiotics had an effect on IL-6 expression, and treatment P1 (administration of probiotics at a dose of 0.60 ml) was the best treatment, based on observations through the ImageJ application with the immunoratio plugin where in fish infected with Myxobolus sp. have high cytokine expression and in fish infected with Myxobolus sp. with the administration of probiotics has a lower IL-6 response, so that probiotics have an important role in regulating the immune system mechanism of Koi Fish (Cyprinus carpio) infected with Myxobolus sp.
