Abstract
Budidaya ikan gurami mengalami peningkatan produksi dan menyebabkan peralihan sistem budidaya ikan dari sistem budidaya tradisional ke sistem budidaya intensif. Namun, dampak negatif dari sistem budidaya intensif adalah serangan penyakit pada ikan gurami yaitu penyakit edwardsiellosis akibat bakteri E.tarda. Bahan alami digunakan untuk mengatasi berbagai macam penyakit yaitu sebagai anti bakteri. Lidah buaya (A.vera) memiliki berbagai macam kandungan bahan aktif yang dapat menghambat dan bahkan membunuh bakteri. Penelitian ini dilaksanakan pada bulan November 2021 – Mei 2022 di Laboratorium Budidaya Ikan Divisi Penyakit dan Kesehatan Ikan, Fakultas Perikanan dan Ilmu Kelautan, Universitas Brawijaya Malang. Penelitian ini menggunakan metode eksperimen dengan rancangan acak lengkap (RAL) dan terbagi menjadi 2 tahapan penelitian. Tahap pertama meliputi ekstraksi dan karakterisasi senyawa antibakteri pada A.vera (uji fitokimia dan FTIR), uji aktivitas bakteri E.tarda meliputi uji cakram, uji MIC dan uji SEM. Tahap kedua meliputi aktualisasi ekstrak lidah buaya (A.vera) yang memiliki sifat anti bakteri pada ikan gurami yang diinfeksi bakteri E.tarda pada skala laboratorium (uji patogenitas, uji toksisitas ektrak, pengamatan profil hematologi dan histopatologi insang, pengamatan gejala klinis, kualitas air serta kelulushidupan). Analisis data yang dilakukan adalah menggunakan aplikasi software Statistical Package for the Sosial Science (SPSS) 25.0 for Windows. Hasil penelitian tahap 1 yaitu melalui uji fitokimia dan FTIR menunjukkan bahwa ekstrak kasar lidah buaya (A.vera) mengandung senyawa flavonoid, tannin, triterpenoid dan saponin. Hasil uji MIC didapatkan dosis 50 mg/L sebagai konsentrasi minimal untuk menghambat pertumbuhan bakteri E.tarda dengan nilai absorbansi mendekati kontrol negatif Hasil uji cakram tertinggi yaitu pada dosis 200 mg/L dengan nilai 6,01 mm. Hasil penelitian tahap 2, uji patogenitas (LD50) bakteri E.tarda ddidapatkan kepadatan 2,53 x 107 sel/ml dan uji toksisitas (LC50) ekstrak kasar lidah buaya yang tidak toksik bagi ikan. Perlakuan dengan pemberian ekstrak lidah buaya (A.vera) memberikan pengaruh yang signifikan terhadap eritrosit, leukosit, hematokrit, hemoglobin dan diferensial leukosit serta gambaran histopatologi organ insang. Nilai dosis yang paling berpengaruh terhadap profil hematologi yaitu pada dosis 150 mg/L sementara pada histopatologi yang paling berpengaruh yaitu pada dosis 150 mg/L.
English Abstract
Gourami cultivation has increased production and caused a shift farming systems from the traditional to intensive aquaculture. However, the negative impact of the intensive aquaculture system is disease attack on gourami, specifically edwardsiellosis caused by the bacterium E.tarda. Natural ingredients are used to treat various diseases as anti-bacterial. Aloe vera has a variety of active ingredients that can inhibit and even kill bacteria. This study was conducted from November 2021 – May 2022 at the Fish Cultivation Laboratory, Fish Disease and Health Division, Faculty of Fisheries and Marine Sciences, Universitas Brawijaya Malang. This study used an experimental method of Completely Randomized Design (CRD) with five treatments and three replications. The first stage includes the extraction and characterization of antibacterial compounds in A. vera (phytochemical test and FTIR), E.tarda bacterial activity test including disc test, MIC test and SEM test. The second stage includes the actualization of Aloe vera (A.vera) extract which has anti-bacterial properties in gourami infected with E.tarda bacteria on a laboratory scale (pathogenicity test, extract toxicity test, observation of gill hematological and histopathological profiles, observation of clinical symptoms, water quality and survival rate). Data analysis was carried out using the Statistical Package for the Social Science (SPSS) 25.0 software application for Windows. The results of the first stage of the study, namely through phytochemical tests and FTIR, showed that the crude extract of Aloe vera contained flavonoid, tannin, triterpenoid, and saponin compounds. The results of the MIC test obtained a dose of 50 mg/L as the minimum concentration to inhibit the growth of E.tarda bacteria with absorbance values approaching the negative control. The highest disc test results were at a dose of 200 mg/L with a value of 6.01 mm. The results of the second stage of the research, the pathogenicity test (LD50) of E.tarda bacteria obtained a density of 2.53 x 107 cells/ml and a toxicity test (LC50) of aloe vera crude extract which was not toxic to fish. Treatment with aloe vera extract (A.vera) gave a significant effect on erythrocytes, leukocytes, hematocrit, hemoglobin, and leukocyte differential as well as the histopathological picture of the gill organs. The dose value that had the most effect on the hematological profile was at a dose of 150 mg/L, while the histopathology had the most effect at a dose of 150 mg/L.
