Abstract

Hiperkolesterolemia merupakan suatu keadaan dimana kondisi kolesterol melebihi batas normal didalam tubuh. Kondisi ini dapat mengakibatkan nekrosis pada ginjal. Beberapa bahan aktif diketahui mampu mencegah peningkatan kadar kolesterol. Sargassum aquifolium merupakan salah satu spesies rumput laut coklat yang berkandungan bioaktif. Beberapa bioaktif diketahui beraktivitas pencegah peningkatan kadar kolesterol. Slow juicing diketahui dapat mengekstraksi bioaktif suatu bahan. Penelitian ini bertujuan mengetahui pengaruh jus S. aquifolium terhadap pencegahan peningkatan kadar kolesterol dan kerusakan profil ginjal tikus yang diberi diet aterogenik. S. aquifolium didapatkan dari perairan Ekabuana, Teluk Ekas, Nusa Tenggara Barat. Tikus Wistar jantan (Rattus novergicus) umur 2-3 bulan dengan berat ±150 g. Pemodelan hiperkolesterol dilakukan dengan cara memberi pakan high fat selama 6 bulan. Metode fitokimia ditentukan berdasar metode Harborne. Penelitian ini terdiri dari 5 perlakuan yaitu kontrol negatif, kontrol positif, dan hewan hiperkolesterol yang diperlakukan dengan pemberian jus 1x, 2 x, dan 3x sehari. Tiap kelompok terdiri dari 3 ekor tikus. Parameter penelitian ini adalah kadar kolesterol dan profil ginjal. Analisa data yang digunakan yaitu analisis keragaman dan uji lanjutan Duncan dengan selang kepercayaan α=0,05. Hasil menunjukkan bahwa jus S. aquifolium mengandung tanin, saponin, dan steroid. Hewan coba yang diberi diet aterogenik menunjukkan peningkatan kadar kolesterol dan ginjal mengalami nekrosis. Pemberian jus S.aquifolium sebanyak 2x sehari mampu mencegah peningkatan kadar kolesterol total dan mencegah kerusakan profil ginjal dibanding perlakuan lainnya. Pemberian jus S.aquifolium 3 kali sehari memiliki kadar kolesterol paling tinggi dan memberikan efek kerusakan pada ginjal paling tinggi. Kesimpulan bahwa jus S.aquifolium mengandung senyawa bioaktif dan pemberiannya 2 kali sehari mampu mencegah peningkatan kadar kolesterol dan kerusakan profil ginjal tikus yang diberi diet aterogenik.

English Abstract

Hypercholesterolemia is a condition in which cholesterol exceeds normal limits in the body. This condition can lead to necrosis of the kidneys. There are active ingredients that can prevent the rise in cholesterol levels. Sargassum aquifolium is a brown alga with bioactive content that can be used as a functional food to prevent elevated cholesterol levels. The content of active ingredients can be obtained by using the slow juicing method. This study aims to prevent cholesterol rise and kidney necrosis in rats. S. aquifolium was obtained from Ekabuana Waters, Ekas Bay, West Nusa Tenggara. Wistar rats (Rattus novergicus) were used as test animals for 2-3 months, weighing ±150 g. Hypercholesterolemia was modelled by feeding a high-fat diet for six months. The phytochemical method was determined based on the Harborne method. This study consisted of 5 treatments, each group containing three rats, namely positive control, negative control, and hypercholesterolemic test animals, which were treated with juice 1x, 2x and 3x a day. The parameters observed were cholesterol levels and kidney profiles. The analysis of the data used is diversity analysis and Duncan's follow-up test with a confidence interval of = 0.05. The results showed that S. aquifolium juice contained tannins, saponins and steroids. Experimental animals fed an atherogenic diet showed increases in cholesterol and MDA levels and renal necrosis. The results showed that administering S. aquifolium juice two times a day could prevent an increase in total cholesterol and prevent damage to the kidney profile. Giving S.aquifolium juice three times a day had the highest cholesterol levels and had the most severe damage to the kidneys. The conclusion is that S.aquifolium juice contains bioactive compounds. Its administration twice a day can prevent increased cholesterol levels and prevent damage to the kidney profile of rats fed an atherogenic diet.
