Abstract
Penelitian ini telah dilakukan di Laboratorium outdoor Jurusan Perikanan, Fakultas Pertanian dan Peternakan Universitas Muhammadiyah Malang di Desa Tegalgondo, Kecamatan Karangploso, Kabupaten Malang, Jawa Timur, Indonesia dari tanggal 20 Mei sampai dengan 01 Agustus 2022. Penelitian ini bertujuan untuk menentukan dosis yang tepat pada probiotik petrofish komersial dengan bakteri yang terkandung didalamnya adalah Lactobacilus plantarum, Bacillus circulan, Bacillus subtilis dan Nitrobacter sp.) untuk menumbuhkan bioflok yang baik sehinga bisa dimanfaatkan oleh ikan lele dalam meningkatkan Kesehatan dan kandungan protein dalam plasma darah sehinga dapat mempertahankan sintasan dan meningkatkan pertumbuhan. Dosis yang diujikan adalah 0 ml/40L air (P0/control), 40 ml/40L air (P1), 60ml/40L air (P2) dan 80ml/40L air (P3), dengan sumber carbon pellet dan gula pasir pada C/N ratio 16 dengan padat tebar 1000 ekor/m3. Metode yang digunakan dalam penelitian ini adalah metode eksperimental rancangan acak lengkap dengan 4 perlakuan dan setiap perlakuan diulang sebanyak 3 kali. Hasil dari pengamatan volume flok yang terbaik terdapat pada P2 yaitu 128,0ml/L dan kualitas air meliputi oksigen terlarut, pH pada semua perlakuan masih dalam tahap normal sedangkan suhu pada saat penelitian terdeteksi rendah yaitu antara 22- 260C, Sementara kadar amonia, nitrit dan nitrat pada perlakuan P2 bernilai normal (0,33mg/L, 0,43mg/L dan 1,19 mg/L) sedangkan amonia dan nitrit pada perlakuan P0, P1 dan P3 bernilai di atas normal. Nilai kualitas air dapt mempengaruhi kandungan bioflok sehinga dapat mempengaruhi status hematologi yaitu sel eritrosit, leokosit dan hemoglobin pada P2 (2,9 x 1012/l, 15,1x1012/l dan 9,6 g/dl) nilai-nilai tersebut dikategorikan normal. Nilai eritrosit dan hemoglobin pada P3 kategori normal. Sedangkan P0 dan P1 nilai eritrosit dan hemoglobin rendah. Kadar leukosit pada perlakuan (P0, P1 dan P3) diatas nilai normal (tinggi) karena pengaruh padat tebar serta kandungan ammonia dan nitrit yang tinggi. Kadar protein dengan metode elektroforesis SDS Page menunjukan bahwa P2 (67,5 kda lebih tinggi dari P3 (45,5kda), P1 (40,5kda) dan P0 (35,7kda. Kandungan bioflok yang baik pada perlakuan P2 mempengaruhi kualitas air dan nilai hematologi, total protein yang tinggi sehinga pertambahan bobot ikan lele mutiara Clarias sp.meningkat yaitu (2215,65gr, FCR 0,90 dan SR. 95%) Berdasarkan hasil pengamatan pada semua variabel dapat dinyatakan bahwa perbedaan dosis dari probiotik petrofish komersial memberikan perbedaan yang sangat nyata terhadap kandungan bioflok sehinga mempengaruhi status hematologi, total protein dan pertumbuhan ikan lele mutiara Clarias sp yaitu pada dosis 60 ml/40L air.
English Abstract
This research was conducted at the outdoor laboratory of the Department of Fisheries, Faculty of Agriculture and Animal Husbandry, Muhammadiyah University of Malang in Tegalgondo Village, Karangploso District, Malang Regency, East Java, Indonesia from May 20 to first of August, 2022. This study aims to determine the right dose of commercial petrofish probiotics with bacteria contained in it are Lactobacilus plantarum, Bacillus circulan, Bacillus subtilis and Nitrobacter sp.) to grow good biofloc so that it can be utilized by catfish in maintaining health and increasing protein content in blood plasma so that it can maintain survival and increase growth. The doses tested were 0 ml/40L water (P0/control), 40 ml/40L water (P1), 60 ml/40L water (P2) and 80 ml/40L water (P3), with carbon sources from pellet and sugar at a C/N ratio of 16, with stocking density of 1000 fish.m-3. The method used in this study was experimental method of complete randomized design with 4 treatments and each treatment was repeated 3 times. The results of the observation of the best floc volume were found in P2 which was 128.0 ml.L-1 and water quality including dissolved oxygen, pH in all treatments were still in the normal stage while the temperature at the time of the study was detected to be low, between 22-260C, while the levels of ammonia, nitrite and nitrate in the P2 treatment were normal (0.33mg/L, 0, 43mg/L and 1.19 mg/L) while ammonia and nitrite in the P0, P1 and P3 treatments are above normal. The value of water quality can affect the content of bioflocs so that it can affect the hematological status of erythrocytes, leocytes and hemoglobin in P2 (2.9 x 1012.L-1, 15.1x1012.L-1), (9.6 g/dl) these values are categorized as normal. Erythrocyte and hemoglobin values in P3 are normal. While P0 and P1 erythrocyte and hemoglobin values are low. Leukocyte level in the treatment (P0, P1 and P3) were above normal values (high) due to the effect of stocking density and high ammonia and nitrite content. Protein levels using SDS Page electrophoresis method showed that P2 (67.5kda) was higher than P3 (45.5kda), P1 (40.5kda) and P0 (35.7kda). Good biofloc content in the P2 treatment affects to the water quality, hematological values, high total protein so that the weight gain of Clarias sp. catfish increases (2215.65gr, FCR 0.90 and SR. 95%) Based on the results of observations on all variable it can be stated that different doses of commercial petrofish probiotics make a very significant difference to the content of biofloc so that it affects hematological status, total protein and growth of Clarias sp catfish at a dose of 60 ml. 40L-1 of water.
