Abstract
Krim pemutih kulit merupakan salah satu jenis kosmetik yang bertujuan untuk mencerahkan dengan memberikan efek yang merata pada penampilan kulit. Asam retinoat atau tretinoin adalah retinoid dari turunan vitamin A dalam bentuk asam all-trans-retinol. Asam retinoat sering digunakan sebagai bentuk sediaan vitamin A topikal untuk mengobati jerawat dan kerusakan akibat sinar matahari, hanya dengan resep dokter. Asam retinoat merupakan bahan yang dilarang dalam kosmetik berdasarkan BPOM Nomor 23 Tahun 2019. Asam retinoat dapat menimbulkan efek samping seperti iritan, karsinogen, dan teratogenik. Analisis kadar asam retinoat diperlukan dalam sampel krim kosmetik tanpa izin edar dari BPOM untuk mencegah bahaya yang ditimbulkan oleh penggunaannya. Penentuan kadar asam retinoat pada sampel krim pemutih dapat menggunakan metode High-Performance Liquid Chromatography (HPLC). Kondisi HPLC yang digunakan adalah kolom VP-ODS Shim-pack (150 x 4,6 mm i.d 5 m) pada suhu 30ᵒC; laju aliran 1,4 ml/menit; fase gerak metanol-air-asam asetat glasial (85:15:0.5) v/v/v; dan volume injeksi 20μL. Verifikasi metode yang dilakukan meliputi spesifisitas (panjang gelombang standar dan sampel terdeteksi pada 353 nm; waktu retensi sampel standar dan analit ±10,5 menit), linieritas (kurva standar dengan konsentrasi 0,5 ppm; 0,6 ppm; 0,7 ppm). ; 0,8 ppm). ppm; 1 ppm, y=52603x – 4040,4, r=0,9958, LOD (0,067 ppm), LOQ (0,203 ppm), kisaran hasil %recovery (99,31%-105,67%), dan hasil berkisar antara %RSD (0,19%-0,88%) . Kadar sampel krim pemutih tanpa izin edar dari BPOM yang mengandung asam retinoat meliputi sampel K1 (0,000859%), K3 (0,000658%), dan K5 (0,000659%).
English Abstract
Skin whitening cream is one type of cosmetic that aims to brighten by giving an even effect on the appearance of the skin. Retinoic acid or tretinoin is a retinoid of the vitamin A derivative in the form of all-trans-retinol acid. Retinoic acid is often used as a topical vitamin A dosage form to treat acne and sun damage, only with a doctor's prescription. Retinoic acid is an ingredient that is prohibited in cosmetics based on BPOM Number 23 of 2019. Retinoic acid can cause side effects such as irritants, carcinogens, and teratogenics. Analysis of retinoic acid levels is required in samples of cosmetic creams without a marketing authorization from BPOM to prevent the harm caused by their use. Determination of retinoic acid levels in whitening cream samples can use the High-Performance Liquid Chromatography (HPLC) method. The HPLC conditions used were VP-ODS Shimpack column (150 x 4.6 mm i.d 5 m) at 30ᵒC; flow rate 1.4 ml/min; mobile phase methanol-water-glacial acetic acid (85:15:0.5) v/v/v; and injection volume of 20μL. Verification of the methods carried out included specificity (standard and sample wavelengths detected at 353 nm; retention time of standard and analyte samples ±10.5 minutes), linearity (standard curve with a concentration of 0.5 ppm; 0.6 ppm; 0.7 ppm). ; 0.8 ppm). ppm; 1 ppm, y=52603x – 4040.4, r=0.9958, LOD (0.067 ppm), LOQ (0.203 ppm), yield range %recovery (99.31%-105.67%), and yields ranged from %RSD (0.19%-0.88%).
