Efektivitas Ekstrak Etanol Daun Melinjo (Gnetum gnemon L.) dalam Menghambat Pertumbuhan Porphyromonas gingivalis Secara In Vitro

Abstract

Periodontitis merupakan penyakit dengan prevalensi tertinggi keenam di seluruh dunia yang dapat menyebabkan kerusakan pada jaringan pendukung gigi. Salah satu etiologi utama dalam perkembangan periodontitis adalah bakteri Porphyromonas gingivalis. Daun melinjo (Gnetum gnemon L.) merupakan komoditas lokal yang mengandung senyawa aktif seperti flavonoid, alkaloid, saponn, dan tannin. Penelitian ini bertujuan untuk mengetahui efektifitas ekstrak etanol daun melinjo dalam menghambat P. gingivalis. Penelitian ini termasuk dalam penelitian eksperimental laboratoris dengan pendekatan post-test only control group design dan menggunakan metode difusi cakram. Sampel dibagi menjadi enam kelompok perlakuan yaitu ekstrak etanol daun melinjo dengan konsentrasi 22,5% ; 20% ; 17,5% ; 15% ; 12,5% ; dan 10% serta dua kelompok kontrol yaitu DMSO (kontrol positif) dan metronidazole (kontrol negatif). Hasil penelitian menunjukkan bahwa zona hambat mulai terbentuk pada besar konsentrasi 12,5% sebesar 12,7 mm dan terus meningkat dengan semakin tingginya konsentrasi ekstrak. Uji Kruskall-Wallis menunjukkan nilai signifikansi <0,05 sehingga dapat disimpulkan bahwa ekstrak etanol daun melinjo (Gnetum gnemon L.) efektif dalam menghambat pertumbuhan P. gingivalis dengan nilai KHM yaitu 12,5%. Hasil uji Post-Hoc menggunakan Mann-Whitney menunjukkan terdapat perbedaan yang bermakna pada setiap pasangan kelompok uji, kecuali pada pasangan kelompok uji K(-) dengan konsentrasi 10% serta konsentrasi 12,5% dengan 15%.

English Abstract

Periodontitis is a disease with the sixth highest prevalence worldwide that can cause damage to the supporting tissues of the teeth. One of the main etiologic in the development of periodontitis is Porphyromonas gingivalis. Melinjo leaves (Gnetum gnemon L) are local commodities that contain active compounds such as flavonoids, alkaloids, saponins, and tannins. This study aims to determine the antibacterial activity of the ethanol extract of melinjo leaves and the effect of inhibiting bacteria Porphyromonas gingivalis. This research is experimental and uses a disk diffusion method. The samples were divided into six groups, namely ethanol extract of melinjo leaves with a concentration of 22.5%; 20% ; 17.5% ; 15% ; 12.5% ; and 10% and two control groups, DMSO as a positive control group and metronidazole antibiotics as a negative control group. The results showed that the inhibition zone began to form at a concentration of 12.5% of 12.7 mm and continued to increase in line with the higher the concentration of the extract. The Kruskal–Wallis test resulted in p < 0.05, indicated that the ethanol extract of melinjo leaves (Gnetum gnemon L.) is effective in inhibiting the growth of Porphyromonas gingivalis with a MIC value of 12.5%. The data processing was followed by Mann–Whitney U-test, and the results showed that there were significant differences among the test groups except for the test group pair K (-) with a concentration of 10% and a concentration of 12.5% with 15%.
