Abstract

Inseminasi buatan dengan semen cair merupakan metode yang tepat untuk perbaikan kualitas genetik di daerah yang sulit mendapatkan nitrogen cair. Penelitian ini bertujuan untuk mendapatkan kombinasi air kelapa dan kuning telur yang dapat digunakan sebagai pengencer semen cair cold storage kambing Boer. Metode yang digunakan adalah eksperimen laboratorium dengan 4 perlakuan dan 10 ulangan sebagai kelompok. Komposisi pengencer yaitu: P0 = CEP-3 + 10% kuning telur + 0,40% putih telur, P1 = air kelapa + 10% kuning telur, P2 = air kelapa + 15% kuning telur dan P3 = air kelapa + 20% kuning telur. Variabel yang diamati dalam penelitian ini adalah motilitas individu, viabilitas, abnormalitas, membran plasma utuh dan akrosom utuh. Data dianalisis menggunakan Analysis of Variance dan terdapat perbedaan antar perlakuan kemudian dilanjutkan dengan Uji Jarak Berganda Duncan. Ada empat perlakuan dan 10 ulangan sebagai kelompok. Data dianalisis menggunakan Analysis of Variance dan terdapat perbedaan antar perlakuan kemudian dilanjutkan dengan Duncan’s (DMRT). Hasil pengujian kualitas semen segar kambing Boer volume 0,99+0,11 ml, berwarna putih kekuningan. Motilitas = 73±3,5%, viabilitas = 77,19±4,54%, abnormalitas = 8,96±1,05 dan konsentrasi spermatozoa 4,120±441,46 juta/ml. Hasil Analisis Varians menunjukkan bahwa pengamatan motilitas individu, viabilitas, membran plasma utuh dan akrosom utuh menunjukkan perbedaan yang sangat nyata (P<0,01), sedangkan kelainan menunjukkan hasil yang tidak berbeda nyata (P>0,05), antara perlakuan selama cold storage pada suhu 3-50C. Disimpulkan bahwa penambahan kuning telur dengan persentase 20% dalam pengencer air kelapa dapat dengan mudah mempengaruhi kualitas semen cair kambing Boer dalam penyimpanan dingin sampai hari ke-3 dengan motilitas 40,41%.

English Abstract

Artificial insemination using liquid semen is an appropriate method for genetic quality improvement in areas where liquid nitrogen is difficult to obtain. This study aims to find a combination of coconut water and egg yolk that can be used as a diluent for liquid semen of Boer goat cold storage. The method used was an experimental laboratory with 4 treatments and 10 replicates as groups. The composition of the diluent namely: P0 = CEP-3 + 10% egg yolk + 0.40% egg white, P1 = coconut water + 10% egg yolk, P2 = coconut water + 15% egg yolk and P3 = coconut water + 20% egg yolk. The variables observed in the study were of individual motility, viability, abnormality, intact plasma membrane and intact acrosome. The data were analyzed using Analysis of Variance and there were different between treatment then continued with Duncan Multiple Range Test. There were four treatment and 10 replicate on as groups. Data were analysed using Analysis of Variance and there were different between treatments then continued with Duncan’s (DMRT). The test results of fresh semen quality of Boer goat are volume 0.99+0.11 ml, with a yellowish white colour. Motility = 73±3.5%, viability = 77.19±4.54%, abnormality = 8.96±1.05 and spermatozoa concentration 4,120±441.46 million/ml. The result of Analysis of Variance showed that the observation of individual motility, viability, intact plasma membrane and intact acrosome indicated a very significantly different (P<0.01), while the abnormality showed result that were not significantly different (P>0.05), between treatment during cold storage at 3-50C. Concluded that the addition of egg yolk with a percentage of 20% in coconut water diluent can easily affect the quality of liquid semen of Boer goat in cold storage until the 3 days with a motility 40.41%.
