Abstract

Tujuan penelitian ini adalah mengetahui pengaruh penggunaan kombinasi kalsium dan ampas jamu pada pakan itik petelur Mojosari terhadap kualitas eksternal dan internal telur melalui metode in vivo dan in silico. Materi yang digunakan yaitu 120 ekor itik petelur Mojosari betina fase layer berumur 24 minggu yang dibagi menjadi 24 flock. Analisis Variasi (ANOVA) dilakukan untuk menganalisis data secara statistik. Dilanjutkan Uji Jarak Berganda Duncan’s (UJBD) apabila data hasil penelitian menunjukkan perbedaan secara signifikan antar perlakuan. Pola Rancangan Acak Lengkap (RAL) dilakukan dalam percobaan ini menggunakan 6 perlakuan dan 4 ulangan. Setiap ulangan terdiri dari 5 ekor itik petelur mojosari. Pakan yang diberikan adalah P0 = pakan basal, P1 = pakan basal + 0,25% zinc bacitracin, P2 = pakan basal + 0,25% kalsium dan ampas jamu, P3 = pakan basal + 0,5% kalsium dan ampas jamu, P4 = pakan basal + 0,75% kalsium dan ampas jamu dan P5 = pakan basal + 1% kalsium dan ampas jamu. Hasil percobaan didapatkan bahwa penggunaan kalsium dan ampas jamu sampai level 1% pada pakan memberikan perbedaan signifikan (P<0,01) terhadap berat telur, berat kerabang telur, volume putih dan kuning telur, dan warna kuning telur. Hasil docking tersebut dapat disimpulkan bahwa cynaroside (-9,2 kcal/mol), 14-Deoxy-11,12-didehydroandrographolide (-9,1 kcal/mol), rutin (-8,8 kcal/mol), andrographidine E (-8,6 kcal/mol), dan cianidanol (-7,8 kcal/mol) memiliki binding affinity yang lebih baik daripada loureirin B (-7.4 kcal/mol). Kandidat terbaik sebagai agen inhibisi GLP1R adalah andrographidine E dari tanaman Andrographis paniculate. Kesimpulan kombinasi kalsium dan ampas jamu dengan dosis 1% dapat diaplikasikan sebagai feed additive natural pada pakan itik petelur Mojosari karena mampu menggantikan fungsi dari antibiotik. Selaras dengan data in silico dimana diketahui interaksi antara senyawa aktif pada ampas jamu (andrographidine E) dapat bertindak sebagai agonis pada protein GLP1R yang lebih baik dibandingkan Loureirin B. Seperti yang diketahui bahwa gen pembentuk protein GLP1R merupakan salah satu key regulator sebagai kontrol gula darah dalam tubuh itik petelur Mojosari, berpengaruh pada kinerja reproduksi dan kualitas telur.

English Abstract

The purpose of this study aimed to determine the effect of using a combination of calcium and herbal waste on mojosari laying duck feed on the external and internal quality of eggs through in vivo and in silico methods. The material used was 120 24-week-old female Mojosari laying ducks divided into 24 flocks. Variation Analysis was used to statistically examine the data (ANOVA). Duncan's Multiple Distance Test is used if the study's data show a substantial difference between treatments. A Complete Randomized Design as employed in the trial, with 6 treatments and 4 tests, each containing 5 mojosari laying ducks. The feed given is P0 = basal feed, P1 = basal feed + 0.25% zinc bacitracin, P2 = basal feed + 0.25% calcium and herbal waste, P3 = basal feed + 0.5% calcium and herbal waste, P4 = basal feed + 0.75% calcium and herbal waste and P5 = basalt feed + 1% calcium and herbal waste. The results of the experiment found that the use of calcium and herbal waste up to the level of 1% in feed did not provide a significant difference (P>0.05) in the egg index, egg shell, volume, egg white index, yolk index, as well as egg yolk cholesterol but showed a significant difference (P<0.01) to egg weight, egg cage weight, white and yolk volume and yolk color. The docking results can be concluded that cynaroside (-9.2 kcal/mol), 14-Deoxy-11,12-didehydroandrographolide (-9.1 kcal/mol), rutin (-8.8 kcal/mol), andrographidine E (-8.6 kcal/mol), and cianidanol (-7.8 kcal/mol) had stronger binding affinity than loureirin B (-7.4 kcal/mol). Andrographidine E, derived from the plant Andrographis paniculata, is the best candidate for GLP1R agonist. The conclusion of the combination of calcium and herbal waste with a dose of 1% can be applied as a natural feed additive to the feed of Mojosari laying ducks because it is able to replace the function of antibiotics. The best candidate as a GLP1R inhibitory agent is andrographidine E from the plant Andrographis paniculate. The conclusion of the combination of calcium and herbal waste with a dose of 1% can be applied as a natural feed additive to the feed of Mojosari laying ducks because it is able to replace the function of antibiotics. In line with the data in silico where it is known that the interaction between active compounds in herbal waste (andrographidine E) can act as an agonist in the GLP1R protein which is better than Loureirin B. As it is known that the gene forming the GLP1R protein is one of the key regulators as a blood sugar control in the body of Mojosari laying ducks, affecting reproductive performance and egg quality.
