Abstract

Reseptor masuk SARS-CoV-2, yaitu ACE2 ditemukan di berbagai jaringan mukosa mulut. Prosedur perawatan gigi memiliki risiko tinggi sehingga direkomendasikan obat kumur antiseptik sebagai tindakan pra-prosedur. Obat kumur povidone iodine terbukti dapat membunuh 99,99% virus SARS-CoV-2 dalam waktu 15 detik. Namun, penggunaan jangka panjang dapat mempengaruhi kondisi rongga mulut. Getah tanaman jarak cina (Jatropha multifida Linn.) banyak digunakan untuk mengobati luka dan diyakini memiliki efek antiseptik yang berpotensi sama baik dengan povidone iodine. Tujuan penelitian ini untuk mengetahui potensi kandungan flavonoid dan tanin getah tanaman jarak cina sebagai bahan obat kumur alami untuk mencegah virus Covid-19 dibandingkan dengan povidone iodine melalui uji in silico. Dilakukan uji fitokimia, pembuatan obat kumur berbahan getah tanaman jarak cina dengan konsentrasi 1%, 3%, dan 5%. Evaluasi obat kumur meliputi uji organoleptis, uji homogenitas, uji pH, dan uji viskositas. Molecular docking dengan software Molegro Virtual Docker 5 licenced. Visualisasi dengan Biovia Discovery Studio ver 21.1.1 dan Pymol, lalu dilakukan analisis Lipinski Rule Of Five. Hasil penelitian menunjukkan senyawa quercetin, acacetin, apigenin, dan proanthocyanidin dalam getah tanaman jarak cina memiliki ikatan diantara protein Spike SARS- CoV-2 dan ACE2 dengan energi ikatan yang lebih rendah dari povidone iodine. Dapat disimpulkan obat kumur yang mengandung senyawa flavonoid dan tanin getah tanaman jarak cina memiliki potensi yang lebih baik untuk pencegahan virus Covid-19 dibandingkan obat kumur povidone iodine melalui uji in silico.

English Abstract

The SARS-CoV-2 entry receptor, ACE2, is found in various tissues of the oral mucosa. Dental procedures have a high risk, so it’s recommended to use an antiseptic mouthwash as a pre-procedure. Povidone iodine mouthwash is proven to kill 99.99% of the SARS- CoV-2 virus within 15 seconds. However, long-term use can affect the condition of the oral cavity. The latex of the Chinese Jatropha plant (Jatropha multifida Linn.) is widely used to treat wounds and is believed to have an antiseptic effect as good as povidone iodine. The purpose of this study was to determine the potential content of flavonoids and tannins from the latex of the Chinese Jatropha as a natural mouthwash to prevent the Covid-19 virus compared to povidone iodine through an in silico test. The research was conducted by using phytochemical tests, making mouthwash made from the latex of the Chinese jatropha plant with concentrations of 1%, 3%, and 5%. Evaluation of the mouthwash which included organoleptic tests, homogeneity tests, pH tests, and viscosity tests. In silico method with Molegro Virtual Docker 5 software licensed as molecular docking, Biovia Discovery Studio ver 21.1.1, and Pymol as visualization and Lipinski Rule Of Five analysis was performed. The study showed that the compounds quercetin, acacetin, apigenin, and proanthocyanidin contained in the latex of the Chinese jatropha plant showed the lower binding energy than povidone iodine to SARS-CoV-2 Spike Protein and ACE2. It can be said that mouthwash containing flavonoid and tannins compounds from the Chinese jatropha plant has better potential for preventing the Covid-19 virus than povidone iodine mouthwash through an in silico test
