Abstract

Akhir-akhir ini seleksi pada ternak mulai dikembangkan melalui seleksi berdasarkan marka penanda atau Marker Assisted Selection (MAS) yang dinilai mampu mengidentifikasi keragaman genetik pada ternak. Tujuan penelitian ini adalah untuk mengkaji keragaman gen Growth Hormone (GH) dan asosianya terhadap bobot badan, lingkar dada, panjang badan dan lingkar ekor pada domba ekor gemuk (DEG). Materi yang digunakan dalam penelitian ini adalah 37 ekor ternak DEG, yang terdiri dari 13 ekor jatan dan 24 ekor betina, umur 1-2 tahun. Metode yang digunakan adalah metode survey dan sampel diambil secara purposive sampling. Koleksi sampel darah sebanyak 5 ml kemudian dilakukan ekstraksi DNA. Fragmen DNA selanjutnya diamplifikasi dan diidentifikasi keragaman genotipenya melalui metode PCR-RFLP dengan menggunakan enzim restriksi AluI dan MspI. Polimorfisme gen GH dan terhadap bobot badan, lingkar dada, panjang badan dan lingkar ekor pada domba ekor gemuk (DEG) dianalisis menggunakan analisis ragam. Hasil penelitian menunjukkan bahwa gen GH-MspI bersifat polimorfik dengan, ditemukan Single Nucleotide Polymorphism (SNP) pada titik 573 dengan perubahan basa nukleotida A → G. Keseimbangan Hardy-Weinberg menunjukkan bahwa populasi dalam keadaan seimbang. Ditemukan tiga genotipe gen GH-MspI (AA, AG, dan GG) dengan dua alel (A dan G). Frekuensi genotipe AA, AG, dan GG secara berurutan adalah 0,30; 0,51; dan 0,19; sedangkan frekuensi alel A dan G masing-masing 0,55 dan 0,45. Hasil analisis ragam menunjukkan bahwa tidak terdapat asosiasi antara keragaman genotipe gen GH-MspI terhadap bobot badan, lingkar dada, panjang badan dan lingkar ekor pada domba ekor gemuk (DEG). Adapun gen GH-AluI tidak menunjukkan adanya keragaman genetik atau bersifat monomorfik.

English Abstract

Selection based on markers or Marker Assisted Selection (MAS) is considered capable of identifying genetic diversity in which related with produstion of livestock. The aim of this study was to examine the diversity of Growth Hormone (GH) genes and their associations with body weight, chest circumference, body length and tail circumference in fat tailed sheep (fts). The material used in this study was 37 DEG cattle, consisting of 13 males and 24 females, aged 1-2 years. The method used is a survey method and samples were taken by purposive sampling. Collection of 5 ml of blood samples was then performed DNA extraction. The DNA fragments were then amplified and identified for their genotypic diversity through the PCR-RFLP method using the restriction enzymes AluI and MspI. The polymorphism of the GH gene and its effects on body weight, chest circumference, body length and tail circumference in fat tailed sheep (FTS) were analyzed using analysis of variance. The results showed that the GH-MspI gene was polymorphic with Single Nucleotide Polymorphism (SNP) found at point 573 with changes in the nucleotide bases A G. Hardy-Weinberg equilibrium indicates that the population is in a balanced state. Three genotypes of the GH-MspI gene (AA, AG, and GG) with two alleles (A and G) were found. The genotype frequencies of AA, AG, and GG were 0.30; 0.51; and 0.19; while the allele frequencies of A and G were 0.55 and 0.45, respectively. The results of the analysis of variance showed that there was no association between the genotype diversity of the GH-MspI gene on body weight, chest circumference, body length and tail circumference in fat tailed sheep (FTS). The GH-AluI gene does not show any genetic diversity or is monomorphic.
