Abstract
Infeksi jamur menjadi salah satu permasalahan yang dialami oleh Indonesia. Salah satu akibat dari infeksi jamur adalah adanya masalah kulit yang sering dikeluhkan oleh masyarakat Indonesia. Penyebab kandidiasis tersering adalah Candida albicans, diikuti oleh Candida glabrata, Candida tropicalis, Candida krusei dan Paracandida. Candida albicans mengalami resistensi terhadap beberapa antifungi fluconazole sebesar 34,07% sehingga diperlukan adanya alternative terapi. Tanaman sanrego (Lunasia amara) memiliki banyak manfaat salah satunya sebagai antifungi. Bakteri endofit asal tanaman Sanrego ini diketahui menghasilkan senyawa metabolit yang mirip dengan ianangnya. Tujuan dari penelitian untuk mengetahui adanya aktivitas antifungi metabolit bakteri endofit Sanrego terhadap pertumbuhan Candida albicans secara in vitro serta menentukan KHM dan KBM. Bakteri endofit A-1 dikultur pada media cair Tryptic Soy Broth (TSB) dan dinkubasi selama ± 3 hari. Suspensi bakteri endofit yang telah diinkubasi selanjutnya disentrifus dengan kecepatan 4500 rpm selama 15 menit pada suhu ruang. Hasil sentrifus disaring menggunakan membran filter 0.22 mikrometer untuk mendapatkan metabolit bakteri endofit A-1. Pengujian aktivitas antifungi dilakukan metode dilusi cair dengan 5 konsentrasi yaitu 30%, 40%, 60%, 80%,100%, kontrol positif berisi suspensi Candida albicans dan kontrol negatif berisi media Sabouraud Dextrose Broth (SDB). Hasil uji dilusi cair didapatkan tidak ditemukan hambatan pertumbuhan Candida albicans ditandai dengan suspensi yang keruh. Hasil analisis data Kruskal Wallis menunjukkan adanya perbedaan yang signifikan secara analisis anatar konsentrasi metabolit bakteri endofit terhadap nilai Optical Density ditunjukkan dengan nilai p-value 0.005 (<0.05). Hasi uji Korelasi Pearson menunjukkan hasil p>0.05 yang artinya tidak ada korelasi antara kenaikan konsentrasi dengan pertumbuhan Candida albicans. Kesimpulan penelitian ini adalah hasil uji dilusi cair antara supernatan bakteri endofit A-1 Sanrego dengan Candida albicans tidak ditemukan adanya Konsentrasi Hambat Minimal (KHM), sehingga Konsentrasi Bunuh Minimal (KBM) tidak dapat ditentukan.
English Abstract
Fungal infection is one of the problems experienced by Indonesia. One of the consequences of fungal infections is skin problems that are often complained of by Indonesian people. The most common cause of candidiasis is Candida albicans, Candida glabrata, Candida tropicalis, Candida krusei, and Paracandida. Candida albicans experienced resistance to several fluconazole antifungals of 34.07%, so an alternative therapy was needed. The sanrego plant (Lunasia amara) has many benefits, one of which is as an antifungal. This endophytic bacterium from the Sanrego plant is metabolites similar to its host. This study aimed to determine the antifungal activity of Sanrego endophytic bacterial metabolites on the growth of Candida albicans in vitro and to determine MIC and MBC. Endophytic bacteria A-1 were cultured in Tryptic Soy Broth (TSB) liquid medium and incubated for ± three days. The endophytic bacterial suspension that had been incubated was then centrifuged at 4500 rpm for 15 minutes at room temperature. Centrifuge results were filtered using a 0.22-micrometer filter membrane to obtain endophytic bacterial metabolites A-1. Testing for antifungal activity was carried out by the liquid dilution method with 5 concentrations, namely 30%, 40%, 60%, 80%, 100%, the positive control contained Candida albicans suspension and the negative control contained Sabouraud Dextrose Broth (SDB) media. The results of Kruskal Wallis test data analysis showed that there was an analytically significant difference between the concentration of endophytic bacterial metabolites and the Optical Density value indicated by a p-value of 0.005 (<0.05). The results of the Pearson Correlation test showed p> 0.05, there was no correlation between the increase in concentration metabolites endophytic and the growth of Candida abicans. This study concludes that the results of the liquid dilution test between the supernatant of endophytic bacteria A-1 Sanrego and Candida albicans did not find any Minimal Inhibitory Concentration (MIC), so the Minimum Bactericidal Concentration (MBC) could not be determined.
