Abstract
Latar Belakang Tendinopati Achilles merupakan penyakit yang diderita oleh 8%-9% atlet pelari. Penyakit tersebut menyebabkan penurunan kemampuan beraktivitas dan kualitas hidup sehari-hari. Tendinopati terjadi karena overuse tendon sehingga menyebabkan micro damages berulang yang mengganggu homeostasis tendon dan akan menyebabkan inflamasi. Pada proses inflamasi terdapat infiltrasi sel imun yang melepaskan mediator inflamasi seperti TNF-α dan CXCL12. Metode induksi tendinopati Achilles dengan lari pada treadmill digunakan untuk menyebabkan overuse tendon pada tikus Wistar. Tujuan Untuk membuktikan induksi tendinopati Achilles dengan metode lari pada treadmill dapat meningkatkan kadar TNF-α dan CXCL12 tendon pada tikus Wistar. Metode Penelitian ini merupakan eksperimental in vivo pada hewan coba tikus putih (Rattus novergicus strain Wistar) dengan post test control group design serta simple random sampling. Tikus dibagi menjadi 2 kelompok, kelompok kontrol (tidak berlari) dan perlakuan (berlari) dengan jumlah tikus tiap kelompok 9 ekor dengan total 18 ekor. Data penelitian didapat dengan pengukuran kadar TNF-α tendon dan CXCL12 tendon dengan menggunakan Kit ELISA dari sampel jaringan tendon tikus Analisis data dilakukan dengan uji komparatif berupa uji Independent T test didahului uji normalitas (saphiro wilk) dan uji homogenitas (levene). Hasil Tidak ada perbedaan yang signifikan kadar TNF-α tendon pada kelompok kontrol dan kelompok perlakuan (p=0.632) dan tidak ada perbedaan yang signifikan kadar CXCL-12 tendon pada kelompok kontrol dan kelompok perlakuan (p=0.061). Kesimpulan Tidak terdapat pengaruh induksi tendinopati Achilles dengan metode lari pada treadmill terhadap kadar TNF-α dan CXCL-12 tendon pada tikus Wistar.
English Abstract
Background Achilles tendinopathy is a disease that affects 8% -9% runner athletes. The disease causes a decrease in the ability to move and quality of daily life. Tendinopathy occurs due to overuse of tendon, causing repeated micro damage that disrupts tendon homeostasis and will cause inflammation. During inflammatory process there is infiltration of immune cells which release inflammatory mediators such as TNF-α and CXCL12. Achilles tendinopathy induction method by running on a treadmill is used to induce tendon overuse in Wistar rats. Objective To prove that induction of Achilles tendinopathy by treadmill running method can increase TNF-α and CXCL12 tendon levels in Wistar rats. Methods This study was an in vivo experimental study on white rats (Rattus novergicus Wistar strain) with a post test control group design and simple random sampling. The rats were divided into 2 groups, the control group (not running) and the treatment group (running) with the number of rats in each group are 9 and total are 18 mice. The research data was obtained by measuring both levels of TNF-α and CXCL12 using the ELISA Kit from samples of tendon tissue from rats. Data analysis was carried out using a comparative test in the form of an Independent T test, preceded by a normality test (Saphiro Wilk) and a homogeneity test (levene). Results There was no significant difference in tendon TNF-α levels in the control and treatment groups (p=0.632) and there was no significant difference in tendon CXCL-12 levels in the control and treatment groups (p=0.061). Conclusion There was no effect of Achilles tendinopathy induction by treadmill running method on TNF-α and CXCL-12 tendon levels in Wistar rats.
