Abstract
Pneumonia merupakan salah satu penyakit infeksi yang masih menjadi permasalahan dan penyebab kematian terbesar pada anak-anak di seluruh dunia. Salah satu penyebab utama pneumonia adalah bakteri Klebsiella pneumoniae. Strain Klebsiella pneumoniae telah menjadi resisten terhadap hampir semua antibiotik yang ada saat ini, sehingga diperlukan adanya alternatif terapi. Tanaman sanrego (Lunasia amara) banyak dimanfaatkan oleh masyarakat, salah satunya sebagai antiinfeksi. Bakteri endofit asal tanaman sanrego diketahui dapat menghasilkan senyawa metabolit yang mirip dengan inangnya. Tujuan dari penelitian ini adalah untuk mengetahui aktivitas antibakteri dari metabolit bakteri endofit asal tanaman sanrego terhadap Klebsiella pneumoniae secara in vitro, serta menentukan KHM dan KBM. Pada penelitian ini, uji antibakteri dilakukan dengan metode uji dilusi cair dengan konsentrasi metabolit bakteri endofit 100%, 80%, 60%, 40%, dan 30%. Hasil uji dilusi cair menunjukkan bahwa tidak terdapat aktivitas antibakteri dari metabolit yang dihasilkan bakteri endofit asal tanaman sanrego terhadap Klebsiella pneumoniae, sehingga KHM dan KBM tidak dapat ditentukan. Selain itu, pada uji korelasi Spearman, didapatkan bahwa konsentrasi supernatan bakteri endofit A-1 asal tanaman sanrego dan pertumbuhan bakteri Klebsiella pneumoniae memiliki hubungan yang signifikan (p = 0,000) dengan koefisien korelasi = 0,975. Hal ini menunjukkan bahwa semakin tinggi konsentrasi metabolit bakteri endofit A1 asal tanaman sanrego, semakin tinggi pertumbuhan bakteri Klebsiella pneumoniae. Pada penelitian ini, didapatkan kesimpulan bahwa tidak terdapat aktivitas antibakteri dari metabolit yang dihasilkan bakteri endofit A-1 asal tanaman sanrego terhadap Klebsiella pneumoniae secara in vitro. Sebaliknya, metabolit yang dihasilkan bakteri endofit A-1 asal tanaman sanrego justru meningkatkan pertumbuhan Klebsiella pneumoniae, sehingga KHM dan KBM tidak dapat ditentukan.
English Abstract
Pneumonia is an infectious disease that is still a problem and as the biggest cause of death in children worldwide. One of the main causes of pneumonia is the Klebsiella pneumoniae. Klebsiella pneumoniae strains have become resistant to almost all currently available antibiotics, so alternative therapies are needed. The sanrego plant (Lunasia amara) is widely used by the community, one of them is as an anti-infective. Endophytic bacteria from the sanrego plant are known to produce metabolites similar to their host. The purpose of this study was to determine the antibacterial activity of endophytic bacterial metabolites from the sanrego plant against Klebsiella pneumoniae in in vitro test, and to determine the MIC and MBC. In this study, the antibacterial test was carried out using the liquid dilution test method with endophytic bacterial metabolite concentrations of 100%, 80%, 60%, 40%, and 30%. The results of the liquid dilution test showed that there was no antibacterial activity from metabolites produced by endophytic bacteria from the sanrego plant against Klebsiella pneumoniae, so the MIC and KBM could not be determined. In addition, in the Spearman correlation test, it was found that the concentration of supernatant endophytic bacteria A-1 from the sanrego plant and the growth of Klebsiella pneumoniae bacteria had a significant relationship (p = 0.000) with a correlation coefficient = 0.975. This shows that the higher concentration of the endophytic bacteria A-1 metabolites from the sanrego plant, the higher the growth of Klebsiella pneumoniae bacteria. In this study, it was concluded that there was no antibacterial activity from metabolites produced by endophytic bacteria A-1 from the sanrego plant against Klebsiella pneumoniae based on in vitro test. Against this, the metabolites produced by endophytic bacteria A-1 from the sanrego plant actually increased the growth of Klebsiella pneumoniae, so the MIC and KBM could not be determined.
