Abstract
Lidah buaya (Aloe vera) mengandung senyawa aktif antihiperglikemik polisakarida seperti acemmanan, glukomannan, dan fruktan yang memodulasi bakteri dalam usus melalui peningkatan ekspresi gen. Penelitian ini dilakukan untuk melihat pengaruh ekstrak lidah buaya (Aloe vera) yang mengandung acemmanan dan fruktan terhadap ekspresi gen penyandi GLP-1 dan menentukan dosis efektif untuk terapi DM Tipe 2. Studi eksperimental murni menggunakan metode post-test only with control group design. Tikus model DM Tipe 2 dipilih dengan sistem simple random sampling kemudian dibagi kedalam lima kelompok dari 20 tikus, diantaranya “k positif”, “k2 metformin”, “k3 ekstrak lidah buaya (Aloe vera) sebanyak 250 mg/kg/BB”, “k4 ekstrak lidah buaya (Aloe vera) sebanyak 300 mg/kg/BB”, “k5 ekstrak lidah buaya (Aloe vera) sebanyak 350 mg/kg/BB”. Terdapat perbedaan ekspresi gen GLP-1 yang signifikan antara kelompok yang diberikan ekstrak lidah buaya dan tidak diberikan ekstrak lidah buaya (p<0.05) yaitu kelompok dengan dosis 250 mg/kg/BB dan dosis 300 mg/kgBB namun tidak signifikan pada dosis 350 mg/kg/BB. Ekstrak lidah buaya (Aloe vera) sebesar 300 mg/kgBB merupakan dosis efektif dalam meningkatkan ekspresi gen GLP-1 dibandingkan dosis 250 mg/kgBB (p<0.05).Penelitian ini membuktikan ekstrak lidah buaya (Aloe vera) mampu menstimulasi ekspresi gen GLP-1 melalui dosis pemberian ekstrak sebesar 300 mg/kg/BB.
English Abstract
Aloe vera contains polysaccharide antihyperglycemic active compounds such as acemannan, glucomannan, and fructans, which modulate bacteria in the gut through increasing gene expression. This research was conducted to see the effect of an aloe vera extract containing acemannan and fructan on the expression of GLP-1-encoding genes and to determine the effective dose for the treatment of type 2 diabetes. A pure experimental study using the post-test only method with a control group design Type 2 DM model rats were selected using a simple random sampling system and then divided into five groups of 20 mice, including "k positive," "k2 metformin," "k3 extract of aloe vera as much as 250 mg/kg/BB," "k4 extract of aloe vera as much as 300 mg/kg/BB," and "k5 extract of aloe vera as much as 350 mg/kg/BB." There was a significant difference in GLP- 1 gene expression between groups given aloe vera extract and those not given aloe vera extract (p<0.05), namely the groups at a dose of 250 mg/kg/BB and at a dose of 300 mg/kg/BB, but not at a dose of 350 mg/kg/BB. Aloe vera extract at 300 mg/kg BB is an effective dose for increasing GLP-1 gene expression compared to a dose of 250 mg/kg BB (p<0.05). This study shows that a dose of 300 mg/kg/BB of aloe vera extract can stimulate GLP-1 gene expression.
