Abstract

Tongkol jagung merupakan salah satu produk samping yang dihasilkan dari pengolahan industri pertanian. Ketersediaan tongkol jagung yang melimpah menjadikan tongkol jagung berpotensi sebagai salah satu penghasil bioproduk dengan nilai dan manfaat yang lebih tinggi. Tongkol jagung memiliki komponen hemiselulosa yang tinggi dan berpotensi sebagai bahan baku pembuatan prebiotik xilo-oligosakarida (XOS). Penelitian ini bertujuan untuk mengetahui perbandingan metode hidrolisis asam dan metode kombinasi hidrolisis asam dengan enzimatis secara one-pot berdasarkan konsentrasi xilo-oligosakarida yang diperoleh dan kombinasi variasi perlakuan yang memberikan konsentrasi xilo-oligosakarida maksimum. Proses ekstraksi pada penelitian ini yaitu kombinasi metode praperlakuan hidrolisis asam dan hidrolisis enzimatis yang dilakukan secara one-pot menggunakan katalis asam oksalat dan enzim xilanase dengan variasi konsentrasi asam sebesar 0%, 0,5%, 1%, dan 1,5% pada suhu 121ºC selama 30 dan 60 menit. Kondisi hidrolisis enzimatis pada suhu 55ºC selama 24 jam dengan kecepatan 100 rpm. Selain itu dilakukan hidrolisis asam tanpa penambahan enzim dengan kondisi perlakuan yang sama sebagai perlakuan pembanding. Hasil penelitian yang diperoleh yaitu konsentrasi xilo-oligosakarida maksimal dari proses hidrolisis asam sebesar 15,69 g/L dan dari metode hidrolisis enzimatis one-pot sebesar 11,47 g/L yang dihidrolisis dengan 0,5% asam oksalat selama 60 menit. Konsentrasi xilosa maksimal didapatkan dari metode hidrolisis enzimatis one-pot sebesar 16,71 g/L dengan 1,5% asam oksalat selama 60 menit. Konsentrasi gula pereduksi maksimal pada hidrolisis asam sebesar 43,34 g/L dengan 1,5% konsentrasi asam oksalat selama 30 menit. Selain itu, berdasarkan analisis neraca massa ditunjukkan bahwa metode hidrolisis asam menghasilkan 15,70 g XOS dan 6,14 g xilosa dan hidrolisis enzimatis one-pot menghasilkan 11,47 g XOS dan 7,41 g xilosa dari 100 g tongkol jagung.

English Abstract

Corn cob are one of the by-products from the agricultural industry. The abundant availability of corn cob makes corn cob potential as a raw material for bioproducts with higher value and benefits. Corn cob have high hemicellulose components and potential as raw materials to produce xylo-oligosaccharides (XOS) prebiotics. This study aims to to find out the comparison of acid hydrolysis and combination of acid and enzymatic hydrolysis through one pot based on the xylo-oligosaccharide contents and to find out the best treatment combination that resulting maximum xylo-oligosaccharide. The extraction process in this study is a combination of acid hydrolysis pre-treatment and enzymatic hydrolysis, which was carried out in one pot using the oxalic acid and xylanase enzyme as the catalysts with various acid concentrations of 0,5%, 1%, and 1,5% at 121ºC for 30 and 60 minutes. As for enzymatic hydrolysis were carried at a temperature of 55ºC for 24 hours at 100 rpm. In addition, acid hydrolysis without enzyme carried out as comparison. The results showed that the maximum concentration of xylo-oligosaccharides from the acid hydrolysis method was 15,69 g/L with 0,5% oxalic acid for 60 minutes. Besides, 11,47 g/L was obtained with 0,5% acid concentration for 60 minutes from the one pot enzymatic hydrolysis method. The maximum xylose concentration obtained from the one pot hydrolysis method was 16,71 g/L with 1,5% oxalic acid for 60 minutes. The maximum concentration of reducing sugar on acid hydrolysis was 43,34 g/L with 1,5% concentration of oxalic acid for 30 minutes. Furthermore, based on mass balance analysis showed that 15,70 g XOS and 6,14 g xylose were produced through acid hydrolysis method and 11,47 g XOS and 7,41 g XOS were produced trough enzymatic hydrolysis using one-pot method from 100 g oven dried raw corn cob.
