Pengaruh Pemberian Ekstrak Etanol Daun Kenikir (Cosmos caudatus Kunth.) terhadap Persentase Apoptosis dan Siklus Sel pada Kultur Candida albicans

Abstract

Kandidiasis adalah penyakit ginekologis akibat jamur yang paling umum terjadi pada manusia. Penggunaan anti jamur seperti Fluconazole dilaporkan telah meningkatkan resistensi pada kandidiasis sebesar 7%. Flavonoid telah terbukti digunakan sebagai anti jamur pada Candida albicans. Flavonoid dapat mengganggu fungsi mitokondria, mempengaruhi potensial membran pada mitokondria, sehingga menghambat siklus sel dan menginduksi apoptosis. Penelitian ini menggunakan desain penelitian true experimental dengan post test only with control group design. Isolat Candida albicans diperoleh dari Laboratorium Mikrobiologi Universitas Brawijaya Malang, Indonesia dengan kode C. albicans SV-1148. Isolat ditanam pada medium Sabouraud Dextrose Agar (SDA) dan Sabouraud Dextrose Broth (SDB). Kelompok sampel dibagi menjadi kelompok kontrol negatif dan kontrol positif dengan fluconazole (60 μg/ml) serta kelompok perlakuan dengan dosis konsentrasi ekstrak etanol daun kenikir (Cosmos caudatus Kunth.) 5%,10%, 20%, 40%, dan kombinasi (konsentrasi ekstrak etanol daun kenikir sebanyak 20% + Flukonazol 30 μg/ml). Penelitian ini menghasilkan peningkatan persentase apoptosis pada kelompok perlakuan yang diberi ekstrak etanol daun kenikir (Cosmos caudatus Kunth.) dan kelompok perlakuan kombinasi dibandingkan dengan kelompok kontrol negatif. Ada penurunan persentase siklus sel pada fase S (sintesis DNA) pada kelompok perlakuan yang diberi ekstrak etanol daun kenikir (Cosmos caudatus Kunth.) dan kelompok perlakuan kombinasi dibandingkan dengan kelompok kontrol negatif. Penelitian ini menunjukkan bahwa peningkatan persentase apoptosis pada konsentrasi ekstrak etanol daun kenikir (Cosmos caudatus Kunth.) sebanyak 10% disebabkan kandungan flavonoid pada daun Kenikir (Cosmos caudatus Kunth.) yang dapat menginduksi apoptosis. Penurunan persentase siklus sel disebabkan oleh kerusakan DNA karena adanya flavonoid pada daun Kenikir (Cosmos caudatus Kunth.). Daun Kenikir (Cosmos caudatus Kunth.) dapat meningkatkan persentase apoptosis dan menurunkan persentase siklus sel fase S (sintesis DNA) pada kultur Candida albicans.

English Abstract

Candidiasis is a fungal gynecological disease that is most common in humans. The use of antifungal agents such as Fluconazole has been reported to increase resistance to candidiasis by 7%. Flavonoids have been shown to be used as an anti-fungal in Candida albicans. Flavonoids can interfere with mitochondrial function, affecting membrane potential in the mitochondria, thus inhibiting the cell cycle and inducing apoptosis. This research designed true experimental of post-test only with control group design. Candida albicans isolate was obtained from the Microbiology Laboratory of Universitas Brawijaya Malang, Indonesia with the Candida albicans SV-1148 code. Isolates were planted in Sabouraud Dextrose Agar (SDA) medium and Sabouraud Dextrose Broth (SDB). The sample group was divided into negative control group and positive control of fluconazole (60 μg / ml) and the treatment group with a dose of ethanol extract concentration of kenikir leaves (Cosmos caudatus Kunth.) 5%, 10%, 20%, 40%, and combinations (concentration ethanol extract of kenikir leaves as much as 20% + Fluconazole 30 μg / ml). This study showed an increase in the percentage of apoptosis in the treatment group given an ethanolic extract of kenikir leaves (Cosmos caudatus Kunth.) and the combination treatment group compared to the negative control group. There was a decrease in the percentage of cell cycles in the S phase (DNA synthesis) in the treatment group given an ethanolic extract of kenikir leaves (Cosmos caudatus Kunth.) and the combination treatment group compared to the negative control group. The study showed that an increase in the percentage of apoptosis in the concentration of ethanol extract of kenikir leaves (Cosmos caudatus Kunth.) as much as 5% due to the flavonoid content of Kenikir leaves (Cosmos caudatus Kunth.) which can induce apoptosis. The decrease in the percentage of cell cycles is caused by DNA damage due to the presence of flavonoids in Kenikir leaves (Cosmos caudatus Kunth.). Kenikir leaves (Cosmos caudatus Kunth.) can increase the percentage of apoptosis and decrease the percentage of S-phase cell cycles (DNA synthesis) in the culture of Candida albicans.
