M2 Macrophage Polarized by Baicalin Induces VEGF Autocrine Signalling for Immunomodulation of CD8+ T cells

aicalin is a flavone glycoside, the glucuronide of baicalin, extracted from the roots of Scutellaria baicalensis and has been known to promote M2 macrophage polarization. Some studies showed that M2 macrophage has angiogenic capability by production of pro matrix metalloproteinase-9 zymogen (MMP-9). Since evidence that one of the most important factors in angiogenesis is vascular endothelial growth factor (VEGF), we investigated the potential involvement of M2 macrophage induced by baicalin in angiogenic activity related to the signalling of VEGF and its receptor. Our result indicated that baicalin, alike IL-4, can promote M2 polarization of Raw 264.7 cells. M2 macrophage polarized by baicalin was shown round shape in morphology and significantly increased M2 gene transcripts including interferon regulatory factor 4 (IRF4), arginine-1 (Arg1), interleukin-10 (IL- 10). However, the expression of surface protein CD206 was not as dominant as M2 macrophage polarized by IL-4. Interestingly, qRT-PCR has shown that M2 macrophage polarized by baicalin significantly increased the expressions of VEGF-A, VEGFR1 and VEGFR2 compared with M2 macrophage polarized by IL-4. The secreted VEGF-A protein in conditioned medium of M2 macrophage polarized by baicalin was also significantly increased. Meanwhile, the expression of PD-L1 was significantly increased in M2 macrophages induced by baicalin. Interestingly, the PD-L1 expression in M2 macrophages induced by baicalin was suppressed by VEGFR inhibitor (Axitinib). After injection of M2 macrophages induced by baiclin to mice, CD8+ T cells were decresed in the peripheral blood and spleen, and CD4+/CD25+ T cells were increasd in bone marrow and the spleen. Finally, we conclude that the concomitant expression of VEGF-A and its receptor in M2 macrophage polarized by baicalin involves an autocrine signalling for immunomodulation of T cells possibly through PD-1-PD-L1 interaction.
