Abstract
Inseminasi buatan (IB) menggunakan semen cair tidak membutuhkan nitrogen cair di dalam media penyimpanannya. Semen cair membutuhkan pengencer serta media simpan untuk membawa semen cair dalam aplikasi IB yang mampu mempertahankan kualitas semen cair. Penelitian ini bertujuan untuk mengetahui kualitas semen cair sapi PO menggunakan pengencer berbeda yang disimpan dalam media simpan berbeda. Penelitian dilakukan di Laboratorium Sumber Sekar Fakultas Peternakan Universitas Brawijaya, Malang. Materi penelitian adalah sapi PO yang ditampung 2 kali seminggu. Metode penelitian adalah eksperimen laboratorium dengan 2 pengencer 5 media simpan 5 ulangan yaitu pengencer : P1 (CEP-3 (Bovine Serum Albumin (BSA) dalam pengencer CEP-2 diganti dengan 0,4% putih telur) + 10% kuning telur) dan P2 (tris aminomethan tanpa raffinosa + 20% kuning telur), media simpan : M1 (termos berisi es batu), M2 (refrigerator suhu 3-5o C), M3 (termos berisi air es), M4 (termos berisi air) dan M5 (termos kosong). Variabel yang diamati adalah motilitas, viabilitas, abnormalitas, integritas membran dan total spermatozoa motil. Analisis data pengencer pada setiap media menggunakan uji T tidak berpasangan, analisis data media simpan pada setiap pengencer menggunakan Rancangan Acak Kelompok (RAK) dan jika terdapat perbedaan signifikan dilanjutkan dengan uji jarak berganda (Duncan). Total spermatozoa motil diuji menggunakan Pearson's Chi Square dengan nilai harapan 40 juta motil/ml. Hasil penelitian menunjukkan P2 (tris aminomethan tanpa raffinosa + 20% kuning telur) merupakan pengencer terbaik dan M1 (termos berisi es batu) sebagai media simpan terbaik untuk membawa semen cair dalam aplikasi IB di lapangan karena mampu mempertahankan kualitas semen cair
English Abstract
Artificial insemination (AI) using liquid semen does not require liquid nitrogen in its storage media. Liquid semen requires extender and storage media to carry liquid semen in the application of AI capable of maintaining the semen quality. The research was to know the quality of liquid semen Ongole crossbred using different extender stored in different storage media. Research was conducted at Sumber Sekar Laboratory, Faculty of Animal Husbandry, Brawijaya University, Malang. The research material used Ongole crossbred which where accommodated 2 times a week. Research method was laboratory experiment using treatment 2 extender 5 storage media and 5 replications there were extender : P1 (CEP-3 (Bovine Serum Albumin (BSA) in CEP-2 substituted with 0,4% albumen) + 10% egg yolk) and P2 (tris aminomethan without raffinose + 20% egg yolk, storage media : M1 (thermos containing ice cubes), M2 (refrigerator), M3 (thermos containing ice water), M4 (thermos containing water), and M5 (empty thermos). The variables observed were motility, viability, abnormality, sperm membrane integrity and total motile sperm. Analysis of extender data on each storage media using unpaired T test, to analyze data of storage media in each extender using Randomized Block Design (RBD) and if there was significant difference between the treatments then tested with multiple range test (Duncan). Total motile sperm tested using Pearson's Chi Square with expectation value of 40 million motile sperm/ml. The results showed that P2 (tris aminomethan without raffinose + 20% egg yolks) was the best extender and M1 (thermos containing ice cubes) as the best storage media to carried liquid semen in AI applications in the field as it was able to maintain the quality of liquid semen.
