Abstract
Udang putih Pasifik atau udang vaname (L. vannamei), merupakan udang laut asli daerah pantai belahan bumi barat, dan kini telah berhasil diperkenalkan dan dibudidayakan di air payau dan di air tawar di daratan Cina. Udang vaname mulai masuk ke Indonesia pada tahun 1999 sebagai alternatif udang budidaya selain spesies udang windu (Penaeus monodon) yang sebelumnya mengalami penurunan produksi budidaya karena berbagai hal. Pada tanggal 10 Oktober 2000 Pemerintah Indonesia mengizinkan impor udang putih Pasifik hanya untuk tujuan penelitian. Udang vaname memiliki kemampuan untuk menjaga regulasi osmotik karena berbagai jenis salinitas, spesies ini mampu menghuni perairan dengan salinitas berkisar antara 0,5 sampai 40 ppt. Akan tetapi, perbedaan salinitas dapat mempengaruhi tingkat kelangsungan hidup udang itu sendiri. Pada budidaya intensif, variasi salinitas dapat merusak homeostasis, menyebabkan stress, pertumbuhan lambat, dan tingkat kelangsungan hidup yang rendah serta cenderung berkulit tipis. Penelitian tentang dampak salinitas yang berbeda terhadap ekspresi gen udang masih banyak yang harus dikaji. Salah satunya adalah tentang ekspresi gen Molt Inhibiting Hormone (MIH) udang vaname. Tujuan dari penelitian ini adalah menganalisis pola ekspresi gen MIH, laju pertumbuhan udang vaname, serta mutasi yang terjadi pada udang vaname yang dipelihara di budidaya air tawar payau dan laut. Penelitian ini dilaksanakan pada bulan November 2018 sampai dengan Januari 2019. Sampel udang vaname (L. vannamei) diambil dari Tambak yang ada di Kabupaten Lamongan. Kabupaten Gresik, dan Kabupaten Situbondo. Sampel udang disimpan dan selanjutnya dibawa ke Laboratorium Sentral Ilmu Hayati Universitas Brawijaya Malang untuk dilakukan uji PCR. Selama penelitian di lapangan, dilakukan analisis kualitas air, meliputi pH, Suhu, dan oksigen terlarut. Penelitian ini menggunakan metode deskriptif berdasarkan kajian molekuler. Karakter Molekuler dari gen MIH udang vaname dari sampel udang vaname yang budidaya di air laut, payau, dan tawar teridentifikasi sebagai gen MIH pada udang vaname dengan Query cover antara 95-99%, nilai identity antara 99-100 dan E-value 0.0. Berdasarkan hasil elektroforesis, gen MIH ditemukan pada basepair 212 – 237. Average Daily Growth (ADG) tertinggi didapat pada udang vaname laut dengan 0,22 gr/hari dan Average Body Weight (ABW) tertinggi didapatkan oleh udang vaname payau dengan 7,65 gram. Terjadi mutasi pada udang yang dipelihara di air laut, payau, dan tawar. Mutasi transisi (satu basa purin menjadi satu basa purin atau satu basa pirimidin menjadi satu basa pirimidin) terletak pada nukleutida ke 58 (Laut, Payau, dan Tawar), dan mutasi transversi (perubahan satu basa purin menjadi satu basa pirimidin atau sebaliknya) terletak pada nukleutida ke 66 (Payau, Tawar), 103 (Laut).
English Abstract
The Pacific white shrimp (L. vannamei), is a sea shrimp native to the western hemisphere coastal region, and has now been successfully introduced and cultivated in brackish water and in freshwater on the Chinese mainland. Vaname shrimp began to enter Indonesia in 1999 as an alternative to cultivated shrimp in addition to the tiger shrimp species (Penaeus monodon) which had previously decreased cultivation production due to various things. On October 10, 2000 the Indonesian Government allowed the import of Pacific white shrimp only for research purposes. Vaname shrimp has the ability to maintain osmotic regulation due to various types of salinity, this species is able to inhabit waters with salinity ranging from 0.5 to 40 ppt. However, differences in salinity can affect the survival rate of the shrimp itself. In intensive cultivation, variations in salinity can damage homeostasis, cause stress, slow growth, and low survival rates and tend to be thin- skinned. Research on the effects of different salinity on shrimp gene expression is still much to be studied. One of them is about the expression of the Molten Inhibiting Hormone (MIH) vaname shrimp gene. The purpose of this study was to analyze the pattern of MIH gene expression, the growth rate of vaname shrimp, and the mutations that occur in vaname shrimp which are maintained in brackish freshwater and marine aquaculture. This research was conducted from November 2018 to January 2019. Samples of vaname shrimp (L. vannamei) were taken from ponds in Lamongan Regency. Gresik Regency and Situbondo Regency. Shrimp samples were taken to the Central Laboratory of Life Sciences, University of Brawijaya Malang for PCR testing. During research in the field, an analysis of water quality was carried out, including pH, temperature, and dissolved oxygen. This study uses descriptive methods based on molecular studies. Molecular character of MIH from vaname shrimp samples cultivated in seawater, brackish, and freshwater was identified as MIH gene in vaname shrimp with cover queries of 95-99%, identity values between 99-100 and E-value 0.0. Based on the results of electrophoresis, the MIH gene is found in basepair 212- 237. The highest average Daily Growth (ADG) is found in sea vaname shrimp with 0.22 gr / day and the highest Average Body Weight (ABW) obtained by brackish vaname shrimp with 7.65 grams . Mutations occur in shrimp that are kept in sea water, brackish, and fresh. Transition mutations (one purine base into one purine base or one pyrimidine base into one pyrimidine base) are located at 58th nucleutide (Sea, Brackish, and Freshwater), and transversion mutations (changes in one purine base into one pyrimidine base) are located at the nucleutide 66 (Brackish, Freshwater), 103 (Sea).
