Abstract
Hedyotis corymbosa secara lokal dikenal sebagai Peh-Hue-Juwa-Chi-Cao dari famili Rubiaceae, tersebar luas di daerah tropis Asia. Berdasarkan penggunaan tradisional, para peneliti memberikan bukti ilmiah yang substansial mengungkapkan dampak menguntungkan dari tanaman ini menyoroti antikanker, hepatoprotektif, antiulcer, antioksidan, antimalaria, antibakteri dan aktivitas antijamur. Ini mendorong kami untuk melakukan penelitian ini, di mana kami mengisolasi dan mengevaluasi konstituen bioaktif berdasarkan sifat biologisnya kegiatan. Penelitian ini bertujuan untuk menyaring dan mengidentifikasi konstituen bioaktif dari ekstrak metanol seluruh tanaman H. corymbosa dan evaluasi aktivitas antimikroba seperti difusi sumur agar untuk antimikroba uji kepekaan dan konsentrasi hambat minimum. Metanol ekstrak H. corymbosa dipartisi secara berurutan menggunakan etil asetat dan n-butanol.
English Abstract
Hedyotis corymbosa is locally known as Peh-Hue-Juwa-Chi-Cao from the Rubiaceae family, widely distributed in tropical regions of Asia. Based on the traditional uses, researchers provided substantial scientific evidence revealing the beneficial impact of this plant highlighting its anticancer, hepatoprotective, antiulcer, antioxidant, anti-malarial, antibacterial and antifungal activities. This prompted us to conduct the present study, where we isolated and evaluated the bioactive constituents based on their biological activities. This study aims to screen and identify bioactive constituents from the methanol extract of H. corymbosa whole plant and evaluate their antimicrobial activities such as agar well diffusion for antimicrobial susceptibility test and minimum inhibitory concentration. The methanol extract of H. corymbosa was sequentially partitioned using ethyl acetate and n-butanol. The ethyl acetate layer was further fractionated and isolated using chromatographic techniques to obtain pure compounds. The bioactive compound structure was determined using spectroscopic analysis especially the Nuclear Magnetic Resonance (NMR) and Mass Spectrum (MS). The investigation of H. corymbosa resulted in the isolation of eight compounds, including two triterpenes, two sterols, two aromatics and two anthraquinones. The compounds were identified as, β-sitosterol (S1), stigmastane-3,6-dione (S2), ferulic acid (AR1), scopoletin (AR2), 2-hydroxy-1- methoxyanthraquinone (An1), 3-hydroxy-1,2-dimethoxyanthraquinone(An2), ursolic acid (T1) and 3β-hydroxyolean-11-en-28,13β-olide (T2).The antimicrobial effect of the crude extract, partitions and fractions were evaluated using agar well diffusion for antimicrobial susceptibility test and for the pure compounds were evaluated using minimum inhibitory concentration. The ethyl acetate layer and crude extract displayed the higher antimicrobial activities with a zone inhibition about 4 mm larger than n-butanol and water layer. The Minimum Inhibitory Concentration (MIC) for the pure compound was shown that most of the compounds have the ability to inhibit human pathogenic bacteria with average 100 μg/mL. The antimicrobial activities showed by the crude extracts, fractions, and pure compounds of H. corymbosa can be used as a commercial product for antimicrobial agent against S. aureus, Salmonella enterica, E. coli and Bacillus substilis.
