Abstract

Karamunting (Rhodomyrtus tomentosa (Ait.) Hassk) termasuk jenis hortikultura dengan senyawa unggul didalamnya. Pemanfaatan bagian buah telah digunakan sebagai herbal tradisional di Malaysia, Vietnam, Thailand dan China. Di Indonesia, tanaman ini tumbuh sebagai tanaman liar di semak belukar dan hutan Pulau Sumatra, Kalimantan dan Sulawesi. Degradasi senyawa bioaktip dari buah karamunting dapat diminimalisir dengan proses ekstraksi. Tujuan penelitian adalah untuk diperoleh kondisi proses ekstraksi dan kualitas pada titik optimum dari buah. Metode yang dipilih untuk proses ekstraksi adalah Microwave Assisted Extraction (MAE) dan kondisi optimum ditentukan dengan Response Surface Methodology (RSM). Kebutuhan daya dan energi dari MAE dikalibrasi berdasarkan parameter berbasis energi, yaitu densitas daya yang diserap (W/mL) dan densitas energi yang diserap (J/mL). Hasil uji ANOVA diketahui pengaruh signifikan (p-value<0.05) terhadap respon rendemen ekstrak dengan nilai p-value 0.0006, respon total flavonoid 0.0001, serta respon total antosianin 0.0005. Nilai p-value pada lack of fit pada masing-masing respon terdapat pengaruh tidak signifikan (p-value>0.05) yang diartikan model dapat digunakan sebagai prediksi optimasi. Hasil optimasi variabel faktor adalah lama ekstraksi 190 detik dan rasio bahan:pelarut 1:19.05 (w/v). Verifikasi data optimum dihasilkan 1.07% rendemen ekstrak, 3.78 mg/gram total antosianin dan 15.47 mg/gram total flavonoid. Semua respon berada dalam ambang batas prediksi.

English Abstract

Rhodomyrtus tomentosa (Ait.) Hassk including horticulture products with superior bioactive compounds. The benefit of fruit parts has been used as traditional herbs in Malaysia, Vietnam, Thailand and China. In this research, bioactive compounds from Rhodomyrtus tomentosa were obtained by Mcrowave Assisted Exraction (MAE). MAE is a modern extraction methods that used microwaves to enhancement the effectivenesss and efficiency of cell rupture. The optimum results of extraction process was determined by Response Surface Methodology (RSM) toward the extract yields, total anthocyanins content and total flavonoids content. Power and energy from MAE can be calibration with paramter Absorbed Power Density (APD in W/mL) and Absorbed Energy Density (AED in J/mL). Based on ANOVA test results, there is a significant influences on each responses with p-value<0.05. The p-value of the lack of fit in each responses has an insignifant influences (p-value>0.05) which means the model can be used as an optimization predictions. The optimum conditions for MAE process were 190 seconds of extraction times and 1:19.05 (w/v) of sample to solvents ratio. The verification of optimum conditon are 1.07% of extract yields, 3.78 mg/gram of anthocyanins and 15.47 mg/gram of flavonoids, which all of responses are in the threshold of the predicted value.
