Abstract
Bakteri Edwardsiella tarda menyebabkan penyakit edwardsiellosis. Penyakit ini telah menyerang berbagai jenis spesies ikan. Infeksi penyakit ini menyebabkan kerugian bagi para pembudidaya ikan. Oleh karena itu diperlukan upaya untuk mengobati serangan bakteri E. tarda ini. Pengobatan antibiotik yang sering dilakukan namun memberikan efek samping yaitu bakteri resisten. Upaya alternatif lain yang dapat dilakukan yaitu dengan penggunaan bahan alami salah satu contohnya yaitu lengkuas putih (A. galanga). Hal ini dikarenakan di lengkuas putih terkandung senyawa yang berguna sebagai antibakteri alami. Penelitian ini bertujuan untuk mengetahui pengaruh dan dosis optimal pemberian ekstrak lengkuas putih (A. galanga) terhadap pertumbuhan bakteri E. tarda. Penelitian ini dilakukan di Laboratorium Sentra Ilmu Hayati (LSIH), Universitas Brawijaya, Malang pada bulan Desember 2021 Metode penelitian yang digunakan yaitu metode eksperimen untuk mengetahui secara langsung pengaruh pemberian berbagai dosis ekstrak lengkuas putih (A. galanga) terhadap pertumbuhan bakteri E. tarda. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan 5 perlakuan dan 3 ulangan. Dosis perlakuan ekstrak kasar lengkuas putih (A. galanga) yaitu: A (50 ppm), B (100 ppm), C (150 ppm), D (200 ppm) dan E (250 ppm). Hasil penelitian menunjukkan bahwa ekstrak kasar lengkuas putih (A. galanga) memberikan pengaruh sangat nyata terhadap daya hambat pertumbuhan bakteri E. tarda. Pengamatan zona hambat dilakukan setelah inkubasi selama 24 dan 48 jam. Hasil pengamatan setelah inkubasi selama 24 jam menunjukkan dosis ekstrak dengan rata-rata diameter zona bening tertinggi yaitu pada perlakuan E sebesar 10,05 mm dan dosis ekstrak dengan rata-rata zona bening terendah pada perlakuan A sebesar 6,01 mm. Pengamatan setelah inkubasi selama 48 jam didapatkan hasil bahwa zona bening mengecil yang menandakan sifat antibakteri sebagai bakteriosidal. Penelitian ini menunjukkan bahwa hubungan antara pemberian ekstrak kasar lengkuas putih (A. galanga) terhadap pertumbuhan bakteri E. tarda membentuk pola kuadratik dengan persamaan y = 5,45 – 0,013x dan koefisien nilai determinasi (R2) sebesar 0,84. Kesimpulan pada penelitian ini yaitu pemberian ekstrak kasar lengkuas putih (A. galanga) efektif sebagai bahan antibakteri alami bakteri E. tarda. Bahan alami ini bersifat bakteriostatik, yaitu mampu menghambat pertumbuhan bakteri. Perlakuan dosis yang tertinggi dari persamaan grafik hasil uji polynomial orthogonal yaitu pada dosis 250 ppm, sedangkan perlakuan dengan dosis 250 ppm pada uji cakram dengan hasil rerata zona hambat yaitu 9,05 mm menghasilkan nilai zona hambat tertinggi.
English Abstract
Bacterium Edwardsiella tarda which can cause edwardsiellosis. This disease has attacked various types of fish species. Infection of E. tarda cause loss for fish farmers. Therefore, efforts are needed to treat the attack of E. tarda bacteria. Antibiotic treatment is often done but has a side effect, namely bacterial resistance. Another alternative effort that can be done is by using natural ingredients, one example is white galangal (A. galanga). This is because white galangal contains compounds that are useful as natural antibacterials. This study aims to determine the effect and optimal dose of white galangal (A. galanga) on the growth of E. tarda bacteria. This research was conducted at the Laboratorium Sentra Ilmu Hayati (LSIH), Universitas Brawijaya, Malang in Desember 2021. The research method used is an experimental method to determine directly the effect of giving various doses of white galangal (A. galanga) on the growth of E. tarda bacteria. This study used a completely randomized design with 5 treatments and 3 replications. The treatment doses of white galangal (A. galanga) were: A (50 ppm), B (100 ppm), C (150 ppm), D (200 ppm) and E (250 ppm). The results showed that the crude extract of white galangal (A. galanga) leaves had a very significant effect on the inhibition of the growth of E. tarda bacteria. The zone of inhibition was carried out after 24 and 48 hours of incubation. The results of observations after incubation for 24 hours showed that the extract dose with the highest average clear zone diameter was 11.48 mm in treatment C and the extract dose with the lowest average clear zone diameter was 8.85 mm in treatment E. After incubation for 48 hours, the results showed that the clear zone was reduced which indicated its antibacterial properties as bakteriosidal. This study showed that the relationship between the crude extract of white galangal (A. galanga) and the growth of E. tarda bacteria formed a quadratic pattern with the equation y = 5,45 – 0,013x and the coefficient of determination (R2) of 0,84 The conclusion of this study is that the crude extract of white galangal (A. galanga) is effective as a natural antibacterial agent for E. tarda bacteria. This natural material is bacteriostatic, which is able to inhibit the growth of bacteria. The highest dose treatment from the equation of the polynomial orthogonal test results is at a dose of 250 ppm, while the treatment with a dose of 250 ppm in the disc test with an average inhibition zone of 9,05 mm produces the highest inhibition zone value.
