Abstract

Dewasa ini, Cardiovascular Diseases (CVDs) kian meningkat kasusnya di Indonesia maupun di dunia. Tercatat pada tahun 2018 Kemenkes RI menyatakan 33,3 % dan skala global WHO menyatakan 87% penduduk pada tahun 2021 mengalami CVDs yang disebabkan karena stroke. Stroke disebabkan oleh trombosit yang mengalami proses trombolisis oleh reseptor α-trombin (IIa) dengan faktor koagulasi dalam pembuluh darah jantung. Obat kimia seperti heparin, aspirin, dan hirudin untuk mengobati trombolitik telah ada, namun apabila digunakan tidak sesuai anjuran akan berdampak negatif bagi kesehatan. Fokus penelitian ini yaitu pada aktivitas dan interaksi peptida bioaktif anti-trombotik sebagai agen pencegahan trombotik serta keamanannya melalui metode in silico pada protein makanan (Food-derived bioactive peptide) kasein susu, kedelai, dan kefir. K-kasein dari kefir dengan aktivitas 0,037 A berdasarkan analisis BIOPEP dengan total 17 peptida anti-trombotik. ToxinPred dan AlgPred menunjukkan tidak adanya potensi peptida yang berbahaya akan tetapi subunit β-konglisinin mampu menjadi allergen yang dikenali oleh sistem imun IgE. Hasil visualisasi analisis molecular docking menggunakan PyMol, PyRx, dan Discover studio berhasil mendapat peptida GPR dengan binding affinity -6.4 kcal/mol memiliki interaksi pada sisi katalitik serta pocket S1 reseptor α-trombin (IIa). Interaksi tersebut mampu mengganggu faktor V, VII, XI, dan XIII dalam meregulasi trombolisis.

English Abstract

Currently, Cardiovascular Diseases (CVDs) are increasingly cases in Indonesia and the wild world. In fact, in 2018 the Indonesian Ministry of Health stated 33.3% and the WHO global scale stated that 87% of the population in 2021 experienced CVD caused by stroke. Stroke is a disease in which affect by platelets undergo thrombolysis by the α-thrombin (IIa) receptor with coagulation factors on the heart blood vessel. Chemical or synthesis drugs such as heparin, aspirin, and hirudin to treat thrombolytics already exists, but the lack of these drugs if used inappropriately will harm health. Therefore, this study focused on the activity and interaction of anti-thrombotic bioactive peptides as thrombotic prevention agents and their safety used in silico method on the food-derived bioactive peptide of casein milk, soy, and kefir. K-casein from kefir with 0.037 A activity based on BIOPEP analysis consists of 17 anti-thrombotic peptides library. ToxinPred and AlgPred showed there are no potentially harmful peptides but in the variant of β-conglycinin subunit is capable of being an allergen known to the immune system IgE. The results of visualization of molecular docking analysis using PyMol, PyRx, and Discover studio obtained a GPR peptide that has the highest binding affinity with total energy -6.4 kcal/mol having bond interactions on the catalytic side and pocket S1 receptor α-thrombin (IIa). The interaction is able to interfere with factors V, VII, XI, and XIII of thrombolysis regulation.
