Abstract

Buah jeruk (Citrus sp.) adalah buah hortikultura utama di Indonesia. Keanekaragaman hayati yang tinggi pada buah jeruk belum sepenuhnya dikarakterisasi. Karakterisasi dapat berupa karakterisasi morfologi, biokimia serta molekuler. Karakterisasi morfologi dapat berupa karakter morfologi polen dengan parameter yang dapat diamati berupa unit, bentuk, ukuran dan tipe aperture. Karakterisasi molekuler dapat menggunakan penanda ISSR karena reproduksibiltas serta sensitivitas yang tinggi. Penelitian ini bertempat di Balai Penelitian Tanaman Jeruk dan Buah Subtropika (Balitjestro) pada bulan November 2021 - Januari 2022. Tujuan dari penelitian ini adalah untuk menganalisis kekerabatan jeruk berdasarkan karakter polen dan penanda ISSR. Data hasil karakterisasi molekuler dianalisis berdasarkan munculnya pita dan dibuat dendogram kekerabatan menggunakan software NTSYs. Hasil penelitian menunjukkan morfologi polen jeruk memiliki unit monad, bentuk prolate, aperture tetracolporate serta ukuran minuta dan media. Berdasarkan dendogram kekerabatan morfologi polen dan karakter molekuler, sepuluh aksesi jeruk dikelompokkan menjadi dua kelompok dan dari kedua dendogram memiliki perbedaan hasil hubungan kekerabatan. Perbedaan dapat disebabkan karena karakter morfologi polen hanya mencakup sebagian kecil sifat genotipe sehingga tidak cukup kuat untuk menduga hubungan kekerabatan dari sepuluh aksesi jeruk.

English Abstract

Citrus fruit (Citrus sp.) is the main horticultural fruit in Indonesia. The high biodiversity of citrus fruits has not been fully characterized. Characterization can be in the form of morphological, biochemical and molecular characterization. Morphological characterization can be in the form of pollen morphological characters with observable parameters in the form of unit, shape, size and type of aperture. Molecular characterization can use the ISSR marker because of its high reproducibility and sensitivity. This research took place at the Research Institute for Citrus and Subtropical Fruits (Balitjestro) from November 2021 - January 2022. The purpose of this study was to analyze the relationship between oranges based on pollen characters and ISSR markers. Molecular characterization data were analyzed based on the appearance of bands and a kinship dendrogram was made using NTSYs software. The results showed that the morphology of orange pollen had monad units, prolate shape, tetracolporate aperture, minute and media sizes. Based on the dendrogram of pollen morphology and molecular characters, ten citrus accessions were grouped into two groups and from the two dendrograms had different results of kinship relationships. The difference could be caused because the morphological characteristics of the pollen only cover a small part of the genotypic traits so that it is not strong enough to predict the kinship relationship of the ten citrus accessions.
