Abstract

Ulkus merupakan suatu keadaan patologis dimana jaringan epitel menghilang akibat mengelupasnya jaringan nekrotik yang telah meluas ke lamina propria di bagian bawahnya. Salah satu penyebab ulkus yaitu adanya trauma, yang disebut ulkus traumatik. Proses penyembuhan luka ulkus traumatik dapat terjadi selama 7-10 hari tanpa pengobatan (self-limiting) dengan cara menghilangkan faktor-faktor penyebabnya. Nanotransfersome gel ekstrak jeruk nipis (Citrus aurantifolia Swingle) mengandung flavonoid yang memiliki efek anti inflamasi. Nanotransfersome memiliki sifat yang dapat melindungi obat dari degradasi lingkungan dan mampu menghantarkan obat melalui celah sempit antar sel dengan baik. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian nanotrasnfersome gel ekstrak jeruk nipis terhadap jumlah sel Fibroblas pada proses penyembuhan luka ulser mukosa labial tikus wistar. Metode penelitian ini adalah randomized post test dengan sampel 32 ekor tikus wistar yang dibagi menjadi 8 kelompok yaitu kelompok K, kelompok A, kelompok B, dan kelompok C. Pemberian nanotransfersome gel ekstrak jeruk nipis yang dioleskan pada luka mukosa labial dua kali sehari dengan time series hari 3 dan 7. Fibroblas diberi pewarnaan HE dan dihitung menggunakan mikroskop cahaya dengan pembesaran 400x per lima lapang pandang. Berdasarkan hasil uji One Way Anova menunjukkan perbedaan yang bermakna jumlah sel fibroblas (p<0,05) antar masing-masing kelompok. Pada hasil uji Post Hoc Tukey menunjukkan adanya perbedaan pada kelompok K dibandingkan kelompok A, B, dan C (p<0,05). Hasil uji korelasi Pearson adalah positif yang menunjukkan rata-rata jumlah fibroblas semakin meningkat. Kesimpulan penelitian ini adalah nanotransfersome gel ekstrak jeruk nipis berpengaruh terhadap peningkatan jumlah sel Fibroblas pada tikus dengan luka ulser mukosa labial.

English Abstract

Ulcer is a pathological condition in which the epithelial tissue disappears as a result of peeling off the necrotic tissue that has extended to the lower lamina propria. One of the causes of ulcers is trauma, which is called traumatic ulcer. Healing process of traumatic ulcer wound can occur for 7-10 days without treatment (self-limiting) by eliminating the factors that cause it. Nanotransfersome gel extract of lime skin (Citrus aurantifolia Swingle) contains flavonoids which have anti-inflammatory effects. Nanotransfersome can protect drugs from environmental degradation and be able to deliver drugs through narrow gaps between cells. The aim of this study was to determine the effect of lime extract nanotransfersome gel on the number of fibroblasts in the wound healing process of wistar rats labial mucosa ulcers. This research method is randomized post test with 32 Wistar rats divided into 8 groups, namely group K, group A, group B, and group C. Treatment is done by giving the lime extract nanotransfersome gel which is applied to the labial mucosal wound twice a day with time series 3 and 7 days. The fibroblasts were stained with HE staining and counted using a light microscope with 400x magnification per five fields of view . The data obtained performing the One Way Anova test and Tukey's Post Hoc test. Based on the results of the One Way Anova test, there was a significant difference in the number of fibroblasts (p <0.05) between each group. The results of Tukey's Post Hoc test showed a difference in K group compared to A, B, and C groups (p <0.05). The Person correlation result is positive and showed that the average number of fibroblasts is increasing. The conclusion of this study was the lime extract nanotransfersome gel had an effect on increasing the number of fibroblasts in Wistar rat with labial mucosal ulcer wounds.
