Abstract

Jumlah perokok di Indonesia semakin bertambah dari tahun ketahun. Rokok serta zat berbahaya didalamnya berafiliasi dengan perubahan parameter hematologi termasuk leukosit dan hematokrit. Peningkatan kadar leukosit bisa menyebabkan disfungsi endotel. Tingginya nilai hematokrit sudah terbukti terkait dengan meningkatnya risiko penyakit jantung. Penelitian ini bertujuan untuk mengetahui korelasi antara kadar nikotin dan metabolitnya dengan kadar leukosit dan nilai hematokrit pada perokok serta mengetahui perbedaan kadar nikotin dan metabolitnya, leukosit, dan hematokrit pada perokok dan nonperokok. Penelitian ini menggunakan metode cross-sectional dengan desain analisis korelasional dan deskriptif komparatif. Sampel terdiri dari 34 perokok dan 23 nonperokok yang diambil melalui teknik total sampling. Variabel bebas penelitian ini ialah kadar nikotin dan metabolitnya sedangkan variabel terikatnya ialah kadar leukosit dan nilai hematokrit. Pengukuran kadar cotinine menggunakan ELISA (Enzyme-linked immunosorbent assay) Kit sedangkan pengukuran kadar leukosit dan nilai hematokrit menggunakan Sysmex XT-2000i Automated Hematology Analyzer. Analisis data korelasional menggunakan uji korelasi Pearson sedangkan analisis data perbandingan menggunakan uji T-Independent. Hasil analisis korelasi didapatkan p-value cotinine-leukosit (0,84), cotinine-hematokrit (0,67). Sedangkan hasil uji beda didapatkan p-value cotinine (0,00), leukosit (0,33), dan hematokrit (0,00). Kesimpulan penelitian ini ialah tidak ada korelasi signifikan antara kadar cotinine dengan kadar leukosit dan nilai hematokrit, ditemukan perbedaan yang bermakna pada kadar cotinine dan nilai hematokrit namun tidak ditemukan perbedaan yang bermakna pada kadar leukosit perokok dengan nonperokok.

English Abstract

The number of smokers in Indonesia is increasing from year to year. Cigarettes and harmful substances in them are associated with changes in hematological parameters including leukocytes and hematocrit. Elevated leukocyte levels can cause endothelial dysfunction. High hematocrit values are associated with an increased risk of heart disease. This study aims to determine the correlation between levels of nicotine and its metabolites with leukocyte levels and hematocrit values in smokers and to determine differences in levels of nicotine and its metabolites, leukocytes, and hematocrit in smokers and non-smokers. This study used a cross-sectional method with a comparative descriptive and correlational analysis design. The sample consisted of 34 smokers and 23 non-smokers who were taken through total sampling technique. The independent variable in this study was the level of nicotine and its metabolite (Cotinine), while the dependent variable was the leukocyte level and hematocrit value. Measurement of cotinine levels using the ELISA (Enzyme-linked immunosorbent assay) Kit while the measurement of leukocyte levels and hematocrit values using the Sysmex XT-2000i Automated Hematology Analyzer. The correlational data analysis used the Pearson correlation test, while the comparative data analysis used the T-independent test. The results of the correlation analysis obtained p-values of cotinine-leukocytes (0.84), cotinine-hematocrit (0.67). While the results of the different tests obtained p-value cotinine (0.00), leukocytes (0.33), and hematocrit (0.00). This study concludes that there is no significant correlation between cotinine levels with leukocyte levels and hematocrit values, significant differences were found in cotinine levels and hematocrit values but no significant differences were found in leukocyte levels in smokers and non-smokers.
